FluoCells™ prepared microscope slides contain fixed, multi-labeled cell preparations for observation by fluorescence microscopy. FluoCells™ Prepared Slide #1 contains bovine pulmonary artery endothelial cells (BPAEC). MitoTracker™ Red CMXRos was used to stain the mitochondria in the live cells, with accumulation dependent upon membrane potential. Following fixation and permeabilization, F-actin was stained with Alexa Fluor™ 488 phalloidin, and the nuclei were counterstained with the blue-fluorescent DNA stain DAPI.

Do you have any slides of pre-labeled cells or tissues that I can use to demonstrate with?

Some of the most popular are the FluoCells demonstration slides, which have cells or tissue sections that are labeled with three different dyes. FluoCells 1 and 2 have cultured cells, while FluoCells 3 is a tissue section (mouse kidney). Slides with fluorescent beads are also available.

DAPI and Hoechst dyes are quite similar to each other. Why would I choose one over the other?

DAPI is a very common blue-fluorescent dye for nuclear counterstaining and gives very bright labeling on nuclei in fixed and permeabilized cells and tissues. However, it is considered to be a semi-permeant to impermeant stain and provides inconsistent staining of live cells. Hoechst 33342 dye is cell-permeant and stains with the same binding mechanism and fluorescent color; it is preferred for live-cell imaging and is just as good as DAPI for fixed cell labeling.
