L-Glutamine (200 mM) (Gibco™)
L-Glutamine is an amino acid that is required for cell culture. L-Glutamine participates in the formation of purine and pyrimidine nucleotides, amino sugars, glutathione, L-glutamate, and other amino acids, as well as in protein synthesis and glucose production. Unlike most other amino acids, L-glutamine is not stable in solution. The rate at which degradation proceeds is a function of time, temperature, and pH. Gibco™ L-glutamine is a ready-to-use 200 mM stock solution that acts as a cell culture supplement. The optimal concentration is dependent upon the cell type and medium used to culture the cells, but generally falls in the range of 2–6 mM.
We also offer GlutaMAX™ Supplement as a stable alternative to L-glutamine.

EfficientFeed™ A+ AGT™ Supplement (Gibco™)
Gibco® EfficientFeed™ A+ (Plus) AGT™ Supplement is a dry-format single-part supplement for fed batch addition to the culture of multiple cell lines to assist with process development and maximization of bioreactor utilization. Compared to original CHO CD EfficientFeed™ A AGT™ nutrient supplement, EfficientFeed™ A+ supplement has components that are uniquely combined to allow simpler single-part pH-neutral reconstitution, as well as the ability to super-concentrate the supplement from 66.4 g/L (1X) to 199.2 g/L (3X) for reduction of product dilution or increased titers with additional feeding, when appropriate. The product is an animal origin-free, chemically defined formulation that contains no proteins, hydrolysates, or components of incompletely defined composition.
Which EfficientFeed™ + Supplement to Choose?
Consider EfficientFeed™ A+ AGT™ Supplement when:
• You have used CHO CD EfficientFeed™ A supplement in the past and wish to explore simpler reconstitution and super-concentration benefits of new technology for delivery of key components
• You are currently using CD OptiCHO™ as your base cell growth medium
Consider EfficientFeed™ B+ AGT™ Supplement when:
• You have used CHO CD EfficientFeed™ B supplement in the past and wish to explore simpler reconstitution and super-concentration benefits of new technology for delivery of key components
• You are currently using CD CHO as your base cell growth medium
Consider EfficientFeed™ C+ AGT™ Supplement when:
• You have used CD EfficientFeed™ C supplement in the past and wish to explore simpler reconstitution and super-concentration benefits of new technology for delivery of key components
• You want higher productivity in GS CHO, CHO-K1, & CHO-S® derived cell lines

• You are using a base medium with greater than 6 g/L glucose or a hydrolysate at day 0
Increasing Titers
As a complete feed, EfficientFeed™ + supplements have been shown to provide multiple-fold increases in titers at 1X concentration versus batch culture, and are an excellent starting point for process development. Flexibility to further increase titers are enhanced with the ability to super-concentrate the product by reconstituting with less water to deliver up to 199 g/L of fed nutrients (see figure).
Maximizing Bioreactor Utilization
Have you ever wished you could have more working volume at the start of a bioreactor run to get more protein produced, but needed too much feeding volume space to accomplish your optimal goal? Have you ever thought that room for a little more feeding might get you to a new level of titer from a process and maybe even reduce the number of bioreactor runs that might be needed per year? In either of these cases, it is important to have the flexibility to super-concentrate your feeding supplements so you get less product dilution, more space for an additional feeding, and/or more working volume to start your run (see figure). In fact, you may gain about 20% more bioreactor utilization by exploring the unique ability to super-concentrate this EfficientFeed A+ product.
Simplified Handling to Reduce Risk in Scale-Up
The ability to scale up in an AGT™ dry format provides ease of handling and storage in manufacturing and other bioprocess settings. Fewer parts and steps in reconstitution mean fewer opportunities for operator error. Further, ability to store this feed, even when super-concentrated, for the length of a bioreactor run at room temperature, in the dark, may allow you fewer chances of contamination during closed system connections by reducing the number of connections to the bioreactor to one.

Sodium Pyruvate (100 mM)
Sodium Pyruvate is commonly added to cell culture medium as an energy source in addition to glucose. Since the cells make sodium pyruvate as an intermediate metabolite in the glycolysis pathway, it is not necessary to supplement all cultures. However, if the cells have been grown in medium supplemented with sodium pyruvate, we recommend the supplementation is continued as the cells become accustomed to this addition and their growth will lag without it. Gibco® Sodium Pyruvate, 100 mM Solution is formulated as 11 g per liter of water. The final concentration of sodium pyruvate used in most cell culture medium is 1 mM.

Feed Kit A+, B+, C+ (Gibco™)
The Feed Kit A+, B+, C+ provides three supplements and a supplementation strategy designed to optimize cell growth and/or productivity in CHO cells. The three supplements, EfficientFeed™ A+, EfficientFeed™ B+, and EfficientFeed™ C+ are based on the chemically defined CHO CD EfficientFeed™ A, CHO CD EfficientFeed™ B, and CD EfficientFeed™ C nutrient supplements. Gibco™ EfficientFeed+ AGT™ supplements are dry-format, single-part supplements for fed-batch addition to the culture of multiple cell lines to assist with process development and maximization of bioreactor utilization. Compared to the original EfficientFeed AGT nutrient supplements, EfficientFeed+ supplements have components that are uniquely combined to allow simpler single-part pH-neutral reconstitutions, as well as the ability to super-concentrate the supplements for reduction of product dilution or increased titers with additional feeding. By using the three feed supplements independently or in combination, you can optimize fed-batch protocols for both cell growth and protein yield.
Criteria for supplementation
Specific CHO cell lines and clones react differently to alternative supplementation protocols. Metabolic needs vary and it is essential for optimal growth and/or productivity to determine optimal nutrient supplementation protocols for each cell line or clone. You may find that two or more different feeds are needed. General criteria to consider when designing supplementation protocol experiments are provided in the user manual (downloadable below).
Selecting the feed or combination that may fit best for your process
A chart is provided below to help in the selection of Gibco media and feeds to pair for testing based on cell type. Should you wish to gain additional insight into different methods as well as the field support available to you, consult the user guide or request support through your local Account Manager.
Simplified handling to reduce risk in scale-up
The ability to scale up using AGT dry format provides easier handling and storage in manufacturing and other bioprocess settings. Fewer parts and reconstitution steps mean fewer opportunities for operator error. Further, the ability to store this feed, even when super-concentrated, for the length of a bioreactor run at room temperature in the dark may lessen the chance of contamination by reducing the number of connections to the bioreactor to one or two . Customization of other feeding formulations is possible should that option suit your needs better after trying this kit. You can contact your local Field Application Scientist through your Account Manager to learn more about our customization capabilities.

KnockOut™ Serum Replacement - Multi-Species (Gibco™)
Gibco™ KnockOut™ Serum Replacement – Multi-Species (KnockOut SR – Multi-Species) is a more defined, FBS-free medium supplement that supports the growth of pluripotent stem cells (PSCs) cultured on fibroblast feeder cells. Designed to replace fetal bovine serum (FBS) in existing protocols, this Research Use Only product uses the same KnockOut Serum Replacement formulation that has been trusted by many for almost 20 years. For pre-clinical or clinical applications, please see Cat. No. 10828028.
With KnockOut Serum Replacement – Multi-Species, you can:
• Culture mouse, human, and other mammalian feeder-dependent PSCs with confidence
• Rely on the improved performance of a qualified, FBS-free formulation
• Enjoy the peace of mind of having a more stable price and supply compared to FBS
Culture mammalian PSCs with confidence
The KnockOut SR – Multi-Species formulation has been cited in more than 2000 publications. It has been shown to support feeder-dependent culture of embryonic stem cells (ESCs) and induced pluripotent stem cells (iPSCs) from mouse, human, and other mammalian species. For mouse and human PSCs, it has also been shown to support reprogramming, embryoid body (EB) formation, in vitro differentiation, cryopreservation, ESC derivation, and many other applications.
Rely on improved performance
KnockOut SR – Multi-Species has a more defined formulation compared to FBS, which contains many undefined and variable components. Unlike most FBS products, each lot is tested for its ability to support PSCs. Because of this, culturing mouse and human PSCs with KnockOut SR – Multi-Species leads to less differentiation and more compact colony morphology compared to FBS.
Enjoy the peace of mind of having stable price and supply
KnockOut SR – Multi-Species is not affected by the limited supply and fluctuating prices often seen with FBS. This makes it easier to plan your budget and buy what you need, especially since it is more affordable than most FBS products.
