PureLink™ Pro 96 total RNA Purification Kit (Invitrogen™)
The PureLink® Pro 96 total RNA Purification Kit provides high-throughput isolation of high-quality total RNA from the widest range of sample types and sizes. Isolate high yields of total RNA with low genomic DNA contamination from bacteria, yeast, plant, or mammalian samples (Figures 1 and 2).
The one-hour procedure requires no handling or disposal of organic waste. RNA binds to a silica membrane and impurities are removed by washing through vacuum filtration or centrifugation. Benefits of MagMAX magnetic bead–based purification
The magnetic bead–based purification format of the kit allows for easy scaling of the number of samples processed either manually with a magnetic stand or with a magnetic particle processor such as the KingFisher Duo Prime or KingFisher Flex processors.
Magnetic beads offer many benefits compared to other technologies for isolating nucleic acids. Beads bind the RNA and DNA more efficiently than glass fiber filters, resulting in higher and more consistent yields. LeukoLOCK™ Fractionation & Stabilization Kit (Invitrogen™)
The LeukoLOCK™ Fractionation & Stabilization Kit is part of the LeukoLOCK™ Total RNA Isolation System, an innovative method for cellular fractionation of whole blood and total RNA stabilization and extraction from the leukocyte population. The LeukoLOCK™ system is comprised of a Fractionation & Stabilization kit (available separately here) and a LeukoLOCK™ Total RNA Isolation Kit, and is optimized for use with human blood. Blood is a storehouse of cellular information; however, the presence of globin mRNA in RNA prepared from whole blood can interfere with downstream expression profiling applications. The LeukoLOCK™ system employs filter-based leukocyte-depletion technology to isolate leukocytes from whole blood and RNAlater® Solution to stabilize the cells on the filter. By excluding red blood cells, the RNA that is purified from captured leukocytes is inherently depleted of globin mRNA, which improves sample utility for expression profiling and other applications (see Figure).
LeukoLOCK™ Fractionation & Stabilization Kit
A 9–10 mL sample of anticoagulated blood is passed through a LeukoLOCK™ filter, which captures the total leukocyte population while eliminating red blood cells (including reticulocytes), platelets, and plasma. After rinsing with phosphate-buffered saline, the filter is flushed with RNAlater® Solutionto stabilize the RNA in the captured leukocytes (see protocol schematic). The RNA can be isolated immediately, or stabilized cells can be maintained for several days at room temperature or for longer periods at –20°C or –80°C (see Figure).

These tubes are designed for direct collection of 3 mL of blood into a plastic, evacuated blood collection tube containing RNA Stabilizing Reagent. The RNA, and therefore the expression profile, is stable in the collection tube for up to five days at room temperature and minimally 7 days at 4°C (see Figure).

Optimized RNA Sample Prep for 1-10 Cells
The Ambion® Single Cell Lysis Kit enables RNA preparation from 1-10 cells. The Single Cell Lysis kit generates a concentrated cell lysate through the optimized combination of lysis buffer, stop solution, and DNase. The resulting lysate is compatible with common enzymatic reactions such as reverse transcription. Sample preparation is fast and can be completed in only 7 minutes with 2 pipetting steps (Figure 1).

The LeukoLOCK™ Total RNA Isolation System is an innovative method for cellular fractionation of whole blood and total RNA stabilization and extraction from the leukocyte population. After rinsing with phosphate-buffered saline, the filter is flushed with RNAlater® solution to stabilize the RNA in the captured leukocytes (see protocol schematic). RNA Isolation
Purification of the RNA is done by first disrupting the captured cells in a guanidinium thiocyanate–based solution that releases the RNA while simultaneously inactivating nucleases. The cell lysate is collected and briefly treated with Proteinase K. RNA is then purified from the lysate using MagMAX™ magnetic bead-based technology for washing, followed by DNase treatment (with TURBO DNase™) and final clean-up.
Expression Studies
Both quantitative RT-PCR and microarray expression studies have validated the suitability of the purified RNA for reliable transcriptome profiling (see Figure). Compared to RNA processed with a commonly used blood RNA isolation procedure, RNA isolated with the LeukoLOCK™ system produces longer median aRNA after amplification and higher percent present calls without requiring additional post-extraction steps to remove globin mRNA (see Figure). Since globin aRNA represents 25–40% of the aRNA peak from whole blood total RNA, the yield of total RNA is understandably lower with methods that deplete unwanted globin transcripts. RNA amplified after LeukoLOCK™ purification offers greater array sensitivity due to the dramatic reduction in competing globin transcripts.

RiboPure™-Blood souris inclut des tubes à échantillons pré-chargés de la solution RNAlater® afin d’empêcher la détérioration de l’ARN et de préserver les profils d’ARN.
