The kit addresses the requirements stipulated by the European Network of Forensic Science Institutes (ENFSI) and European DNA Profiling Group (EDNAP) and includes the expanded European Standard Set of Loci (ESSL). The NGM SElect™ Express kit simultaneously amplifies the same 16 loci included in the NGM™ kit (D3S1358, vWA, D16S539, D2S1338, D8S1179, D21S11, D18S51, D19S433, TH01, FGA, Amelogenin, D10S1248, D22S1045, D2S441, D1S1656, D12S391) with the addition of the highly polymorphic SE33 locus in the PET (red) dye channel.

Powerful Discrimination
The combination of loci in the NGM SElect™ Express kit generates an extremely high power of discrimination of any AmpFℓSTR® kit (see Table 1).

Maximizes Concordance
The NGM SElect™ Express kit maintains identical primer sequences for the core STR loci common to the SGM Plus®, Identifiler® and SEfiler Plus™ kits, thereby:

• Maximizing concordance with existing data sets for simplified historical comparisons
• Minimizing requirements for repeat analyses necessary to resolve non-concordant events
• Maintaining sufficient similarity of configuration with previous kits to reduce training and implementation requirements

The primers for the SE33 locus were redesigned from those used in the SEfiler Plus™ kit to relocate the amplicon within the multiplex. However, the new primer sequences have been optimized and tested thoroughly to maintain concordance with the SEfiler Plus™ kit.

Easy to Implement, Easy to Use
Every aspect of the NGM SElect™ Express kit has been designed with ease of implementation and use in mind. This is a single kit solution that is designed to deliver high quality results for both database and casework samples, optimizing the forensic DNA workflow while reducing operating costs. The kit workflow is very similar to other AmpFℓSTR® kits, enabling forensic laboratories to streamline validation and implementation. However, the enzyme is now contained within the master mix, simplifying reaction set-up. Cycling times have been shortened to less than one hour, allowing laboratories to achieve higher quality results more quickly. For compatibility with both manual and automated systems, two package sizes are available, the standard 200 reaction kit and a larger 1000 reaction kit. The larger package includes larger bottles to facilitate use on liquid handling platforms.

AmpFLSTR™ Yfiler™ PCR Amplification Kit (Applied Biosystems™)

The AmpFLSTR® Yfiler® PCR Amplification Kit permits the analysis of complex male:female DNA mixtures that often yield ambiguous or inconclusive results with autosomal STRs.

Key Features of the AmpFLSTR® Yfiler® PCR Amplification Kit:

• Analyze trace quantities of male DNA that may be present in a large background of female DNA
• Combine the inherent male specificity of Y-STRs with the robust performance of an AmpFLSTR® kit
• Amplify 17 Y-STR loci in a single PCR reaction which yields the highest discriminatory capacity of any commercially available Y-STR kit
• Search the user-friendly on-line haplotype database to easily determine the frequency of any Yfiler® kit haplotype
• Use this powerful tool for the analysis of cases that contain a suspect(s)

Combines the Inherent Male Specificity of Y-STRs with the Robust Performance of an AmpFLSTR® Kit
Mixtures of body fluids and/or tissues from a male and female are frequently encountered with sexual assault casework and other violent crimes. These mixed samples often contain trace quantities of male DNA present in a large background of female DNA. Typical samples can include vaginal swabs containing a small number of sperm cells, body swabs from female victims of oral assaults, and fingernail scrapings/clippings from female victims of violent assaults. Standard autosomal STR analysis may be unable to detect the minor male contributor in these types of samples, while the Yfiler® kit can routinely detect a minor male contributor present at less than 1:1000.

Provides the Highest Discriminatory Capacity of Any Commercially Available Y-STR Kit
The AmpFLSTR® Yfiler® PCR Amplification Kit amplifies 17 Y-STR loci in a single PCR reaction using 5-dye chemistry to provide the highest discriminatory capacity of any commercially available Y-STR kit. The Yfiler® kit can distinguish more than 98% of all unrelated males from one another, and therefore offers the discriminatory power necessary to conduct meaningful forensic DNA analysis.

A Powerful Tool for Suspect Casework
The Yfiler® kit is a powerful tool for the analysis of cases that contain a suspect(s). A significant proportion of DNA cases contain a suspect who has been identified by conventional law enforcement techniques. For these cases, the Yfiler® kit is a powerful tool that can be used to include or exclude a suspect(s) associated with an investigation.
