HT Supplement (100X) (Gibco™)

Gibco® HT Supplement is a liquid mixture of sodium hypoxanthine (10 mM) and thymidine (1.6 mM). HT-supplemented medium is suitable for post-selection rescue to overcome the effects of residual intracellular aminopterin. Once the de novo biosynthesis pathway for nucleosides has been reestablished, HT supplementation can be discontinued.

Gibco® EfficientFeed™ C+ (Plus) AGT™ Supplement is a dry-format single-part supplement for fed batch addition to the culture of multiple cell lines to assist with process development and maximization of bioreactor utilization. This product is designed to work with CHO cell lines expressing endogenous glutamine synthetase (GS) as a single feed without the need for additional GS supplements. Compared to the original CD EfficientFeed™ C AGT™ nutrient supplement, EfficientFeed™ C+ supplement has components that are uniquely combined to allow simpler pH-neutral reconstitution, as well as the ability to super-concentrate the supplement from 81.2 g/L (1X) to 162.4 g/L (2X) for reduction of product dilution or increased titers with additional feeding, when appropriate. HuMEC Supplement Kit (Gibco™)

HuMEC Supplement Kit contains growth factors and other components and is designed to work with HuMEC Basal Serum-Free Medium (12753018). Together, these products are engineered specifically for the culture of human mammary epithelial cells (A10565). This translates to greater experimental validity, more reproducible results, and reduction of lot-to-lot variability and aberrations that can occur with the addition of serum. Human mammary epithelial cells cultured in HuMEC Medium have gene expression profiles consistent with normal mammary epithelial cells, as tested on a cDNA microarray.

Applications:
HuMEC Medium is optimized specifically for the growth of human mammary epithelial cells, and has been performance tested in a growth assay using normal human mammary epithelial cells. HuMEC Medium promotes superior growth of normal human mammary epithelial cells, making it ideal for researchers in breast cancer, drug discovery, and other aspects of breast biology.

Contents and Storage:
The HuMEC Supplement Kit includes 5 ml of a supplement mix containing epidermal growth factor, hydrocortisone, isoproterenol, transferrin, and insulin, and 25 mg of bovine pituitary extract. Store the supplement mix at -5°C to -20°C.

L-Glutamine (200 mM) (Gibco™)

L-Glutamine is an amino acid that is required for cell culture. L-Glutamine participates in the formation of purine and pyrimidine nucleotides, amino sugars, glutathione, L-glutamate, and other amino acids, as well as in protein synthesis and glucose production. Unlike most other amino acids, L-glutamine is not stable in solution. The rate at which degradation proceeds is a function of time, temperature, and pH. Gibco™ L-glutamine is a ready-to-use 200 mM stock solution that acts as a cell culture supplement. The optimal concentration is dependent upon the cell type and medium used to culture the cells, but generally falls in the range of 2–6 mM.

We also offer GlutaMAX™ Supplement as a stable alternative to L-glutamine.

When used, this dry-format, single-part total feeding supplement can shift glycan profiles from heavily G0F to mostly G1F and G2F glycans. The product is an animal origin–free, chemically-defined formulation that contains no proteins, hydrolysates, or components of unknown composition. GlycanTune A+ Total Feed provides an easy-to-reconstitute, pH-neutral solution that can be concentrated from the suggested 73.0 g/L (1X) up to 219.0 g/L (3X). Dialing in a glycan profile
By following the protocol recommended to test with GlycanTune Total Feed, it is possible to shift the glycan protein expression profiles from primarily G0 to primarily G1 and G2.

Increasing titers
As a complete feed, GlycanTune total feeds have been shown to provide multiple-fold increases in titers at 1X concentration versus batch culture; as such, they are an excellent starting point for process development when a shift in glycan profile is desired. Gibco® Large Vessel Endothelial Supplement (LVES) is a 50X, complete, ready-to-use media supplement optimized for use with Gibco® Medium 200 and Medium 200PRF for culture of primary human umbilical vein endothelial cells (HUVEC), human aortic endothelial cells (HAEC), and human pulmonary artery endothelial cells (HPAEC). This blended supplement has been designed to deliver optimal cell growth and to enable superior cell performance for angiogenesis applications, such as cell migration and tube formation. LVES contains fetal bovine serum, hydrocortisone, human epidermal growth factor, basic fibroblast growth factor, heparin, and ascorbic acid.

Microvascular Growth Supplement (MVGS) (Gibco™)

A sterile, concentrated (20X) solution intended for use with Medium 131 to culture human microvascular endothelial cells. Each 25 ml bottle of MVGS contains all of the growth factors, hormones, and tissue extracts necessary for the culture of human microvascular endothelial cells, and is the correct amount of supplement for a 500 ml bottle of Medium 131. When a 500 ml bottle of medium is supplemented with MVGS, the final concentrations of the components in the supplemented medium are:
• fetal bovine serum (4.9% v/v final concentration)
• hydrocortisone (1 µg/ml)
• human fibroblast growth factor (3 ng/ml)
• heparin (10 µg/ml)

Sodium Pyruvate (100 mM) (Gibco™)

Sodium pyruvate is commonly added to cell culture media as a carbon source in addition to glucose. Since cells make sodium pyruvate as an intermediate metabolite in the glycolysis pathway, it is not a required supplement for all cell cultures. However, if cells have been grown in medium that is supplemented with sodium pyruvate, we recommend continuing use of the supplement as cell growth may lag without it. Gibco® Insulin,-Transferrin-Selenium-Ethanolamine (ITS-X) is used as a basal medium supplement in order to reduce the amount of fetal bovine serum (FBS) used to culture cells. Insulin promotes glucose and amino acid uptake, lipogenesis, intracellular transport, and the synthesis of proteins and nucleic acids. Transferrin is an iron carrier and may also help to reduce toxic levels of oxygen radicals and peroxide. Selenium, as sodium selenite is a co-factor for glutathione peroxidase and other proteins that is used as an anti-oxidant in media. Ethanolamine is a precursor of phospho-glycerides which are essential to the structure of the plasma membrane and cellular organelles.

A sterile, concentrated (100X) solution intended for use as one component in a complete culture environment for the growth of human mammary epithelial cells. Each 5 ml bottle of MEGS contains all of the growth factors, hormones, and tissue extracts necessary for the culture of mammary epithelial cells and is the correct amount of supplement for a 500 ml bottle of Medium 171.

When a 500 ml bottle of medium is supplemented with MEGS, the final concentrations of the components in the supplemented medium are:

• Bovine pituitary extract (BPE) (0.4% v/v)
• Recombinant human insulin-like growth factor-I (1 µg/ml)
• Hydrocortisone (0.5 µg/ml)

N-2 Supplement (100X) (Gibco™)

N-2 Supplement is a chemically-defined, serum-free supplement based on Bottenstein’s N-1 formulation. It is recommended for growth and expression of neuroblastomas as well as post-mitotic neurons in primary cultures from both the peripheral nervous system and the central nervous system. N-2 Supplement is provided as a 100X concentrate and is intended for use with Neurobasal Medium supplemented with growth factors such as bFGF and EGF. It may also be used with DMEM.

Resurge™ CD Pak (Gibco™)

Resurge CD Pak are a diverse set of chemically defined supplement formulations to complement a variety of base media. The CD PAK contains one of each Resurge supplement: CD1, CD2, CD3, CD4, and CD5 at 100 grams each. The supplements have been developed using several host cell lines and in various culture systems, including shaker flasks and bench-scale bioreactors. In order to increase versatility of the formulation, glucose and L-glutamine are not included in the final formulations. Therefore, these two components should be added by users and monitored as appropriate.

Low Serum Growth Supplement (LSGS) (Gibco™)

Low Serum Growth Supplement (LSGS) is a sterile, concentrated (50X) solution intended for use as one component in a complete culture environment for the growth of human dermal fibroblasts and endothelial cells derived from large vessels. LSGS is intended for use with Medium 106 (fibroblasts) or Medium 200 (endothelial cells). LSGS contains: fetal bovine serum, basic fibroblast growth factor, heparin, hydrocortisone, and epidermal growth factor. L-Glutamine (200 mM) (Gibco™)

L-Glutamine is an amino acid that is required for cell culture. L-Glutamine participates in the formation of purine and pyrimidine nucleotides, amino sugars, glutathione, L-glutamate, and other amino acids, as well as in protein synthesis and glucose production. Unlike most other amino acids, L-glutamine is not stable in solution. The rate at which degradation proceeds is a function of time, temperature, and pH. Gibco® L-glutamine is a ready-to-use 200 mM stock solution that acts as a cell culture supplement. The optimal concentration is dependent upon the cell type and medium used to culture the cells, but generally falls in the range of 2–6 mM.

Criteria for supplementation
Specific CHO cell lines and clones react differently to alternative supplementation protocols. Metabolic needs vary and it is essential for optimal growth and/or productivity to determine optimal nutrient supplementation protocols for each cell line or clone. You may find that two or more different feeds are needed. General criteria to consider when designing supplementation protocol experiments are provided in the user manual (downloadable below).

Increasing Titers
As a complete feed, EfficientFeed™ + supplements have been shown to provide multiple-fold increases in titers at 1X concentration versus batch culture, and are an excellent starting point for process development. Flexibility to further increase titers are enhanced with the ability to super-concentrate the product by reconstituting with less water to deliver up to 158 g/L of fed nutrients (see figure).

Maximizing Bioreactor Utilization

Insulin-Transferrin-Selenium-Sodium Pyruvate (ITS-A) (100X) (Gibco™)

ITS-A (Insulin-Transferrin-Selenium-Sodium Pyruvate) Supplement is used as a basal medium supplement in order to reduce the amount of Fetal Bovine Serum (FBS) used to culture cells. ITS-A is used for adherent cell cultures. Originally used with RPMI 1640 and Minimum Essential Medium (MEM), ITS-A is now routinely used with other basal media to support a wide variety of cell types in the presence of 2–4% FBS. We also offer ITS-G and ITS-X supplements.

Using ITS-A Supplement
Insulin promotes glucose and amino acid uptake, lipogenesis, intracellular transport, and the synthesis of proteins and nucleic acids. Transferrin is an iron carrier and it may also help to reduce toxic levels of oxygen radicals and peroxide. Selenite is a co-factor for glutathione peroxidase and other proteins, and is used as an anti-oxidant in media. Sodium Pyruvate is a precursor to amino acids, fatty acids, and cholesterol; it is an important intermediate in many biosynthetic pathways. The complete formulation is available.
