Sodium Pyruvate (100 mM) (Gibco™)

Sodium pyruvate is commonly added to cell culture media as a carbon source in addition to glucose. Since cells make sodium pyruvate as an intermediate metabolite in the glycolysis pathway, it is not a required supplement for all cell cultures. However, if cells have been grown in medium that is supplemented with sodium pyruvate, we recommend continuing use of the supplement as cell growth may lag without it. Gibco® Sodium Pyruvate (100 mM) is formulated using 11 g per liter of water. The final concentration of sodium pyruvate used in most cell culture media is 1 mM.

Bovine Pituitary Extract (BPE) (Gibco™)

The protein concentration of each lot of Bovine Pituitary Extract is determined by the Bradford method. Each vial of BPE contains not less than 25 mg of protein.

L-Glutamine (200 mM) (Gibco™)

L-Glutamine is an amino acid that is required for cell culture. L-Glutamine participates in the formation of purine and pyrimidine nucleotides, amino sugars, glutathione, L-glutamate, and other amino acids, as well as in protein synthesis and glucose production. Unlike most other amino acids, L-glutamine is not stable in solution. The rate at which degradation proceeds is a function of time, temperature, and pH. Gibco® L-glutamine is a ready-to-use 200 mM stock solution that acts as a cell culture supplement. The optimal concentration is dependent upon the cell type and medium used to culture the cells, but generally falls in the range of 2–6 mM.

We also offer Gibco® GlutaMAX™ Supplement as a stable alternative to L-glutamine.

HepaRG™ Maintenance/Metabolism Medium Supplement (Gibco™)

HepaRG™ Maintenance/Metabolism Medium Supplement is a specialized media supplement for use in culturing HepaRG™ cells. HepaRG™ Maintenance/Metabolism Medium Supplement is specifically used for multiple day growth of HepaRG™ cells.

Media Preparation
Working media is prepared by the addition of thawed HepaRG™ Maintenance/Metabolism Medium Supplement to 100 ml of William’s Medium E (A1217601 or 12551032) and 1 ml of GlutaMAX™-I (35050061). The simple two-step process helps reduce contamination opportunities.

Simplified, Relevant and Reproducible Studies
HepaRG™ cells exhibit many characteristics of primary human hepatocytes and are essentially a single donor. These features allow users to obtain physiological relevant results for metabolism-based drug-drug interactions without the concern of donor variability and limited lot sizes that come with relying on donor tissue. HAT Supplement (50X)

Gibco® HAT Supplement is a liquid mixture of sodium hypoxanthine (5 mM), aminopterin (20 µM) and thymidine (0.8 mM). HAT-supplemented medium is suitable for post-fusion selection against unfused or self-fused HGPRT–myeloma cells. Hypoxanthine and thymidine supply preformed purines and pyrimidines for DNA synthesis by hybridomas via the salvage pathway that utilizes HGPRT- contributed by the fused spleen cell. Aminopterin, a folic acid antagonist, inhibits the de novo nucleoside biosynthesis pathway.

EfficientFeed™ C+ AGT™ Supplement (Gibco™)

Gibco® EfficientFeed™ C+ (Plus) AGT™ Supplement is a dry-format single-part supplement for fed batch addition to the culture of multiple cell lines to assist with process development and maximization of bioreactor utilization. This product is designed to work with CHO cell lines expressing endogenous glutamine synthetase (GS) as a single feed without the need for additional GS supplements. Compared to the original CD EfficientFeed™ C AGT™ nutrient supplement, EfficientFeed™ C+ supplement has components that are uniquely combined to allow simpler pH-neutral reconstitution, as well as the ability to super-concentrate the supplement from 81.2 g/L (1X) to 162.4 g/L (2X) for reduction of product dilution or increased titers with additional feeding, when appropriate. The product is an animal origin-free, chemically defined formulation that contains no proteins, hydrolysates, or components of incompletely defined composition.

Increasing Titers
As a complete feed, EfficientFeed™ + supplements have been shown to provide multiple-fold increases in titers at 1X concentration versus batch culture, and are an excellent starting point for process development. Flexibility to further increase titers are enhanced with the ability to super-concentrate the product by reconstituting with less water to deliver over 150 g/L of fed nutrients (see figure).

Maximizing Bioreactor Utilization
Have you ever wished you could have more working volume at the start of a bioreactor run to get more protein produced, but needed too much feeding volume space to accomplish your optimal goal? Have you ever thought that room for a little more feeding might get you to a new level of titer from a process and maybe even reduce the number of bioreactor runs that might be needed per year? In either of these cases, it is important to have the flexibility to super-concentrate your feeding supplements so you get less product dilution, more space for an additional feeding, and/or more working volume to start your run (see figure). In fact, you may gain at least 10% more bioreactor utilization by exploring the unique ability to super-concentrate this EfficientFeed C+ product.

Simplified Handling to Reduce Risk in Scale-Up
The ability to scale up in an AGT™ dry format provides ease of handling and storage in manufacturing and other bioprocess settings. Fewer parts and steps in reconstitution mean fewer opportunities for operator error. Further, ability to store this feed, even when super-concentrated, for the length of a bioreactor run at room temperature, in the dark, may allow you fewer chances of contamination during closed system connections by reducing the number of connections to the bioreactor to one.

Gibco® Glucose Solution contains D-glucose at a concentration of 200 g/L and is used as a serum-free medium supplement suitable for cell culture. Glucose Solution can be added to a variety of Gibco® cell culture media including but not limited to Dulbecco’s Modified Eagle Medium (DMEM), RPMI 1640, DMEM/F-12, IMDM, and Minimum Essential Medium.
