L-Glutamine (200 mM) (Gibco™)

L-Glutamine is an amino acid that is required for cell culture. L-Glutamine participates in the formation of purine and pyrimidine nucleotides, amino sugars, glutathione, L-glutamate, and other amino acids, as well as in protein synthesis and glucose production. Unlike most other amino acids, L-glutamine is not stable in solution. The rate at which degradation proceeds is a function of time, temperature, and pH. Gibco® L-glutamine is a ready-to-use 200 mM stock solution that acts as a cell culture supplement. The optimal concentration is dependent upon the cell type and medium used to culture the cells, but generally falls in the range of 2–6 mM.

Using L-glutamine
L-glutamine (nonsterile) powder should be made as a filter-sterilized stock solution of 200 mM (or similar) in distilled water. The optimal concentration to use is dependent upon the cell type and medium used to culture the cells, but generally falls in the range of 2–6 mM.

GlutaMAX™ Supplement (Gibco™)

Gibco® GlutaMAX™ Supplement is an alternative to L-glutamine, with increased stability that improves cell health. GlutaMAX™ Supplement is suitable for both adherent and suspension culture of mammalian cells, with no adaptation required. GlutaMAX™ Supplement is offered as a 200 mM L-alanyl-L-glutamine dipeptide in 0.85% NaCl. GlutaMAX™ Supplement is also included in a variety of media formulations. Compared to L-glutamine, GlutaMAX™ Supplement:

Minimizes toxic ammonia build-up
Improves cell viability and growth
Remains stable across a wide range of temperatures

Minimizes toxic ammonia build-up
Unlike L-glutamine, GlutaMAX™ Supplement does not spontaneously break down to form ammonia. Instead, cells cleave the dipeptide bond to release L-glutamine as needed. This system prevents waste build-up and maintains a fresh supply of L-glutamine during long-term culture.

Improves cell viability and growth
Cultures with lower levels of ammonia show improved cell viability. In addition, culture conditions are more controlled due to the consistent concentration of GlutaMAX™ Supplement that is not possible with L-glutamine.

Remains stable across a wide range of temperatures
GlutaMAX™ Supplement is room-temperature stable and ready to use. GlutaMAX™ Supplement retains full functionality for 24 months at room temperature, 2 to 8°C, or -5 to -30°C for convenient storage and handling. GlutaMAX™ Supplement is even stable at 37°C for a week or more.

GLUTAMAX I improves growth efficiency and performance of mammalian cell culture systems. GLUTAMAX I eliminates problems associated with the spontaneous breakdown of Lglutamine during incubation. It is highly soluble in aqueous solution and is heat stable. It is supplied as a 200mM (100X), liquid stock in 0.85% NaCl.
