Expi293™ Expression Medium Gibco™

Gibco Expi293 Expression Medium is a chemically defined, serum-free, protein-free medium for growth and transfection of suspension-adapted human embryonic kidney (HEK) 293 cells. It is specifically designed as a core component to support high density culture of Expi293F cells in the Expi293 Expression System for scalable transient protein expression.

• Supports growth of suspension 293 cultures to densities of over 1.5 x 10^7 cells/mL
• Transfection compatible serum-free suspension culture medium enables transfection efficiencies of greater than 70% using ExpiFectamine transfection reagent

Expi293 Expression Medium is formulated with GlutaMAX-I reagent. It is ready to use, with no supplementation required. The chemically defined formulation results in high reproducibility and lot-to-lot reliability. Expi293 Expression Medium contains no human or animal-origin components. The medium is not recommended for adherent cell cultures.

Diploid Production Serum-Free Medium (SFM) Gibco™

The Gibco Diploid Serum-Reduced Medium (SRM) system is designed to support vaccine manufacture under serum-reduced or serum-free conditions for cell types traditionally cultured in classical media with 10% serum. With this system, human diploid cells, such as MRC-5, Wi38, and KMB17, can be grown with just 1–2% serum. Additionally, it supports growth of chicken embryo fibroblasts under serum-free conditions. Virus production can then be conducted under serum-free conditions without the addition of albumin or other supplements.

The Diploid SRM system is composed of two different kits:
1. Diploid Growth Serum-Reduced Medium (SRM) kit: A basal medium and 100X growth supplement designed for cell expansion
2. Diploid Production Serum-Free Medium (SFM) kit (this page): A basal medium and 100X production supplement designed for virus production

Human cells have traditionally been used to manufacture a variety of vaccines, such as the varicella zoster, polio, smallpox, hepatitis A, measles, mumps, and rubella vaccines. Traditionally, the only way to culture these cells has been to use classical media with 10% serum for growth and 1–5% human albumin (human sourced or recombinant) for virus production. These are two of the highest cost components in vaccine production. The Diploid SRM system helps reduce or eliminate both components while maintaining high virus titers.

Cost savings
Reduction or elimination of serum can significantly reduce cost for a vaccine manufacturing process. Premium FBS is expensive with high price volatility. Given the typical cost pressures on vaccine production, both economic drivers make vaccine manufacturing to a target price extremely challenging. Diploid Growth SRM reduces the impact of this volatility in price and offers a level of cost consistency not available with processes that use serum in high volumes.

In addition, serum requires frozen storage. A 10% serum concentration in growth media can result in high frozen storage requirements for a vaccine process. A process requiring 1,000 L of growth medium would require the frozen storage of 100 L of serum. The Diploid Growth SRM kit comes with a 100X frozen supplement, enabling an 80% reduction in frozen storage requirements. 1,000 L of Diploid Growth SRM would only need 20 L of frozen storage: 10 L for FBS and 10 L for the 100X supplement.

Albumin is commonly added to virus production media. With the Diploid Production SFM, viruses can be produced at comparable titers without any additional supplementation.

No adaptation required
The Diploid SRM system was developed with existing cell banks in mind. Transitioning a vaccine process to a new medium can be challenging due to the need to adapt the cells to the new medium. For human cells such as MRC-5, there is the added risk of reaching the Hayflick limit due to the added passages necessary during cell adaptation and subsequent re-banking of the cells. The transition to the Diploid SRM system does not require adaptation or re-banking of cells when using the Diploid Growth SRM kit. Existing cGMP cells banked with serum may be thawed directly in Diploid Growth SRM supplemented with 1% FBS, allowing a simple transition to this medium.

Mitigate serum supply risk
Obtaining a steady supply of serum can be challenging, especially if the serum required is from a BSE/TSE safe region such as New Zealand or Australia. Serum supply can fluctuate based on several environmental (drought, cattle disease) and economic (feed cost, dairy pricing) factors. With Diploid Growth SRM this dependence is substantially reduced due to the reduced serum requirements.

Sf-900™ Medium (1.3X) Gibco™

Sf-900™ SFM is a complete serum-free, low protein insect cell culture medium developed for the convenient and reproducible formation of 1% agarose overlays used for plaque assays of baculovirus in Spodoptera frugiperda (Sf9 and Sf21) cells. When used with Gibco® 4% Agarose Gel, Sf900™ SFM (1.3x) allows for convenient formation of a 1% overlay with correct 1x final osmolality and nutrition required for insect cell growth during the plaque assay.

293 SFM II Gibco™

293 SFM II is a serum-free, low-protein (<10 µg/ml) formulation optimized for high-density suspension culture of 293 cells (1). (It is not suitable for adherent 293 cell cultures.) This product replaces 293 SFM (Cat. No. 10970-010). 293 SFM II is animal-origin free. Formulated without L-glutamine for increased stability; add 20 ml of 200 mM L-glutamine or 20 ml of GlutaMAX™-I Supplement per 1,000 ml of medium prior to use.

Ability to support suspension growth and transfection of 293-F cells
Gibco® 293-F, 293-H, and FreeStyle™ 293-F cells in suspension can be cultured directly into Gibco® FreeStyle™ 293 Expression Medium without adaptation. Cells adapted to other commercially available serum-free media can be subcultured directly into Gibco® FreeStyle™ 293 Expression Medium, usually without any further adaptation. Cells usually require adaptation from serum-containing formulations. Gibco® FreeStyle™ 293 Expression Medium is animal origin–free, protein-free and chemically defined, allowing for easier purification of your protein of interest. Gibco® chemically defined media contain no proteins, hydrolysates, or components of unknown composition.

Depending on the impeller design and speed, it may be necessary to supplement Gibco® FreeStyle™ 293 Expression Medium with additional Pluronic® F-68 to avoid sheer stress in the culture.

The primary manufacturing site for CTS OpTmizer T-Cell Expansion SFM is FDA-registered and has an ISO13485-certified quality management system. Superior long-term, high-density growth

Increases in maximum cell densities of 20–100% are also observed with the Lymantria dispar (Gypsy moth) and Trichoplusia ni (Tn-368; Cabbage Looper) cell lines. Traditionally, Grace's medium supplemented with 10% FBS has been used for recombinant protein expression. Sf-900™ II SFM is an improved serum-free, protein-free medium designed for growth of Sf9 and other lepidopteran cell lines and production of insect virus and rDNA proteins.

CD DG44 Medium Gibco™

CD DG44 Medium is a chemically defined, protein-free medium optimized for the growth of dihydrofolate reductase deficient (DHFR-) Chinese Hamster Ovary (CHO) cells and expression of recombinant proteins in suspension culture. CD DG44 fully chemically defined formulation contains no lysates, hydrolysates, nor proteins or peptide components of animal, plant, or synthetic origin.

When to Use CD DG44 Medium
• You use parental DG44 or DXB11 CHO cells (dhfr-)

When to Consider Another Gibco® Medium

Consider CD FortiCHO™ medium when:
• The cell line you are using is a transfected CHO K1, GS CHO or CHO-S™ cell line
• Maximum batch culture cell densities and protein titers are needed
• You have time to adapt cells into a new medium in an effort to maximize titers
• Cell health at harvest is a priority for downstream processing

Consider CD OptiCHO™ medium when:
• You are using a transfected CHO cell line other than CHO K1, GS CHO or CHO-S™ cell lines
• Optimization of feeding strategies will be part of your base medium selection testing in an effort to maximize productivity
• Your CHO cell line is “finicky," i.e. hard to grow or adapt

Consider CD CHO medium when:

Cells grown in CTS OpTmizer T-Cell Expansion SFM with or without human serum demonstrate good cytokine production profiles in polyclonally activated T cells. CTS OpTmizer T-Cell Expansion SFM maintains the desired the CD62L+ central memory T-cell phenotype.

GMP manufactured
The primary manufacturing site for CTS OpTmizer T-Cell Expansion SFM is FDA-registered and has an ISO13485-certified quality management system. The methods and controls used for the manufacturing are in conformity with current Good Manufacturing Practices for medical devices, 21 CFR Part 820, of the regulation. Alternate manufacturing sites are available, providing risk mitigation by helping to ensure security of supply.

Versatility
CTS OpTmizer T-Cell Expansion SFM has been widely adopted in clinical T-cell research applications including the culture of CAR T-cells. It works with multiple culture formats, including culture flasks, culture bags, and rocking bioreactor systems. CTS OpTmizer T-Cell Expansion SFM was designed to support expansion of cultured human T-cells without additional serum supplementation. Increased performance is generally observed when supplemented with CTS Immune Cell Serum Replacement.

What you get
CTS OpTmizer T-Cell Expansion SFM includes two components: 1 liter of CTS OpTmizer T-Cell Expansion Basal Medium and 26 mLs of CTS OpTmizer T-Cell Expansion Auxiliary Concentrated Medium, which are mixed together to create the complete medium. Full traceability documentation such as Certificate of Origin and Certificate of Analysis are available.
