EpiLife™ CF/PRF Kit (Gibco™)

A sterile, liquid medium for the long-term, serum-free culture of human epidermal keratinocytes and human corneal epithelial cells. EpiLife® CF⁄PRF is prepared without calcium chloride and phenol red. Because this product is phenol red free, this product is ideal for oncology studies and when fluorescent microscopy is to be used. EpiLife® greatly extends the in vitro lifespan of human epidermal keratinocytes compared to other serum-free media systems. When used for cellular differentiation, the basal medium requires the addition of calcium plus an appropriate growth supplement prior to use. This basal media is animal origin free and contains essential and non-essential amino acids, vitamins, other organic compounds, trace minerals, and inorganic salts. It does not contain antibiotics, antimycotics, hormones, growth factors, or proteins. EpiLife® medium is HEPES and bicarbonate buffered, and is designed for use in an incubator with an atmosphere of 5% CO2 and 95% air.

Essential 8™ Adaptation Kit (Gibco™)

The Essential 8™ Adaptation Kit includes Essential 8™ Medium and recombinant human Laminin 521 (rhLaminin-521) for optimal survival during transition from feeder-dependent to feeder-free culture. rhLaminin-521 has been proven to promote cellular survival under stressful conditions in the absence of small molecule inhibitors. In addition, rhLaminin-521 has been shown to support PSC growth for >20 passages without any signs of karyotypic abnormalities and to maintain the ability of PSCs to differentiate into all three germ line lineages. rhLaminin-521 can also be purchased separately.

The Essential 8 Adaptation Kit is:
• Robust–transition cells between feeder-dependent and feeder-free culture systems with minimal effort
• Physiologically relevant–expand cells with minimal differentiation
• Scalable–passage stem cell lines as single cells with no impact to genetic stability

Trouble-free transition
When used with Essential 8 Medium, rhLaminin-521 provides a robust culture matrix that supports pluripotent stem cell adherence and expansion during the transition from feeder-dependent culture systems.

Confident expansion
rhLaminin-521 is expressed in blastocysts and therefore provides an environment mimicking the human stem cell niche.

Easy scale-up
rhLaminin-521 promotes stable pluripotent stem cell maintenance as single cells in the absence of ROCK inhibitors.

Astrocyte Medium (Gibco™)

Astrocyte Medium has been specially formulated to support the growth of primary human and rat astrocytes while retaining their phenotypes. The medium has been internally validated and optimized to provide an optimal balance between cell growth and the maintenance of cell viability, normal phenotype, and function. If more rapid proliferation is desired, addition of an appropriate amount of epidermal growth factor (EGF) is recommended, although cells may exhibit morphological and phenotypic changes.

Astrocytes are a type of glial cell of the nervous system with a typical star shape. It has been shown in recent years that astrocytes not only function as gap fillers and structural supports, but play an active role in supporting the normal functions of neurons, oligodendrocytes, neural stem cells, and blood-brain barrier. They regulate the extracellular ionic and chemical environment, respond to CNS injury, and play a fundamental role in the pathogenesis of ischemic neuronal death, and they have remained one of the most active research areas in neuroscience.

Astrocyte Medium has three components: base medium (DMEM), N-2 Supplement, and OneShot Fetal Bovine Serum (FBS). Mixing of these three components yields a complete medium.

For established protocols using Astrocyte Medium, access our Gibco Neurobiology Protocol Handbook.
