ExpiCHO™ Expression Medium (Gibco™)

ExpiCHO™ Expression Medium is a GMP-manufactured, chemically-defined, serum-free, protein-free, animal origin–free medium specifically designed for high-density growth and transfection of suspension-adapted Chinese hamster ovary (CHO) cells. It is a core component of the ExpiCHO™ Expression System Kit (Cat. No. A29133) for scalable transient protein production.

ExpiCHO Expression Medium:
• Supports growth of suspension CHO cultures to densities of over 2 x 107 cells per mL, with >95% cell viability
• Uses a transfection-compatible, serum-free suspension culture medium that enables high transfection efficiencies of >90% with ExpiFectamine™ CHO transfection reagent
• Supports transient transfection of suspension CHO cells at a density of 6 x 106 cells per mL
• Facilitates sustained, high-level expression of high-density transiently transfected cultures, achieving yields of up to 3 g per L of recombinant protein
• Supports growth and transfection of CHO cells in culture formats from <1 mL in multi-well plates to >10 L in benchtop bioreactors

ExpiCHO Expression Medium is formulated with GlutaMAX™-I reagent. It is ready to use, with no supplementation required. The chemically-defined formulation results in high reproducibility and lot-to-lot reliability. For highest level transient protein expression, the medium is used in combination with a matched culture feed, ExpiCHO™ Feed, which is supplied as part of the ExpiFectamine™ CHO Transfection Kit (Cat. No. A29129). ExpiCHO Expression Medium is not recommended for adherent cell cultures. Cells are typically grown in Erlenmeyer-style flasks on a shaker platform in a humidified, CO2 cell culture incubator. However, cultures are scalable from volumes of <1 mL in multi-well plates to >10 L in bioreactors.

FreeStyle™ 293 Expression Medium (Gibco™)

FreeStyle™ 293 Expression Medium is an animal origin–free, chemically defined, protein-free medium specifically developed to support the growth and transfection of 293-F cells in suspension without adaptation. The medium is complete and ready-to-use, with no supplementation required. Gibco® FreeStyle™ 293 Expression Medium features:

Gibco® FreeStyle™ 293 Expression Medium is ready to use. It does not require the addition of serum, glutamine, or surfactants. Gibco® FreeStyle™ 293 Expression Medium contains GlutaMAX™ supplement to minimize toxic ammonia build-up and improve cell viability and growth in an easy-to-use format.

Ability to support suspension growth and transfection of 293-F cells
Gibco® 293-F, 293-H, and FreeStyle™ 293-F cells in suspension can be cultured directly into Gibco® FreeStyle™ 293 Expression Medium without adaptation. Cells adapted to other commercially available serum-free media can be subcultured directly into Gibco® FreeStyle™ 293 Expression Medium, usually without any further adaptation. Cells usually require adaptation from serum-containing formulations. Cells cultured in Gibco® FreeStyle™ 293 Expression Medium can be transfected using lipid based transfection reagents, such as 293fectin™ reagent and Freestyle™ MAX reagent.

Gibco® FreeStyle™ 293 Expression Medium is animal origin–free, protein-free and chemically defined, allowing for easier purification of your protein of interest. Gibco® chemically defined media contain no proteins, hydrolysates, or components of unknown composition.

Easy use as part of the FreeStyle™ 293 Expression System and FreeStyle™ MAX Expression System for protein production
The FreeStyle™ 293 Expression System is a complete, suspension cell culture system for generating large amounts of mammalian recombinant protein (see figure). The FreeStyle™ 293 Expression System combines Gibco® FreeStyle™ 293 Expression Medium with 293fectin™ reagent, a cationic-lipid formulation designed to transfect suspension FreeStyle™ 293-F cells at high efficiency. The FreeStyle™ MAX Expression System is a breakthrough technology for rapid and high-yield mammalian protein production. Scalability in spinner flasks and bioreactors
Protein production using Gibco® FreeStyle™ 293 Expression Medium can be scaled up in spinner flasks or bioreactors. The appropriate spinner or impeller speed and seeding density should be optimized for each system. Depending on the impeller design and speed, it may be necessary to supplement Gibco® FreeStyle™ 293 Expression Medium with additional Pluronic® F-68 to avoid sheer stress in the culture.

Sf-900™ III SFM (Gibco™)

Sf-900™ III SFM is a low-hydrolysate, serum-free, protein-free, animal origin–free insect cell culture medium optimized for the growth and maintenance of Spodoptera frugiperda (Sf9 and Sf21) cells and for recombinant gene expression using the baculovirus and stable insect expression systems. This medium is suitable for suspension and monolayer culture methods and supports growth of other lepidopteran cell lines.

Superior long-term, high-density growth
Spodoptera frugiperda (Sf9) cells grown in Sf-900™ III SFM achieve maximum cell densities of 10 to 14 × 106 cells/mL, a significant improvement over Sf-900™ II SFM and other commercially available serum-free media.

Optimized for recombinant protein production
Traditionally, Grace's medium supplemented with 10% FBS has been used for recombinant protein expression. Sf-900™ III SFM is an improved serum-free, protein-free, animal origin–free medium designed for growth of Sf9 and other lepidopteran cell lines and production of insect viruses and recombinant proteins.

Gibco® Sf-900™ III SFM is a serum-free, protein-free, animal origin–free medium that allows for much easier purification of your protein of interest. Sf-900™ III SFM is ready to use; it does not require addition of serum, glutamine, or surfactants. 1X CD CHO Medium (Gibco™)

CD CHO medium is a protein-free, serum-free, chemically-defined medium optimized for the growth of Chinese hamster ovary (CHO) cells and expression of recombinant proteins in suspension culture. With a proven track record of quality for more than 10 years, CD CHO medium contains no proteins or peptide components of animal, plant, or synthetic origin, as well as no undefined lysates or hydrolysates.

When to Use CD CHO Medium
• Your GS CHO cells are recommended to be grown in CD CHO medium
• Your cells have been successfully cultured in CD CHO medium in the past and your process is nearing a regulatory filing (DMF available)

When to Consider Another Gibco® Medium

Consider CD FortiCHO™ medium when:

The B-27 Plus Neuronal Culture System is a next-generation media system that provides the highest rate of in vitro survival of primary rodent and human stem cell-derived neurons. Quality and performance testing
Every lot of Gibco® PB-MAX™ Karyotyping Medium is performance tested by a certified US reference cytogenetics laboratory to ensure consistently superior performance. Peripheral blood lymphocytes from a normal adult donor are cultured for 72 hours in Gibco® PB-MAX™ Karyotyping Medium before measuring the mitotic index and chromosome banding resolution. In addition, each lot is tested for sterility, pH, osmolality, and mycoplasma.

Classic formulation
Gibco® PB-MAX™ Karyotyping Medium is an optimized RPMI 1640 medium, supplemented with Fetal Bovine Serum (FBS), L-glutamine, and phytohemagglutinin (PHA). CTS™ Essential 8™ Medium (Gibco™)

CTS Essential 8 Medium is an animal-origin-free (AOF), feeder-free medium formulated for the growth and expansion of both embryonic and induced human pluripotent stem cells (PSCs). Essential 8 Medium is the most defined PSC medium as described in the original publication by Guokai Chen et al. in the laboratory of James Thomson. CTS Essential 8 Medium enables the transition from bench to clinic through the direct replacement of research-use-only (RUO) Essential 8 Medium.

CTS Essential 8 Medium:
• Reduces the risks associated with animal origin material through an AOF formulation
• Facilitates regulatory filings through cGMP manufacturing and the availability of documentation
• Simplifies the transition from bench to clinic with the same 8-component formulation as RUO Essential 8 Medium, but with AOF components

Reduces risks from animal-origin material
CTS Essential 8 Medium contains no animal- or human-derived components at the primary component level, thereby reducing the risks associated with animal-origin components. It is the only commercially available AOF medium for pluripotent stem cell culture: Competitors’ similar-grade products are only xeno-free, containing human serum albumin (HSA), which brings additional risks in a clinical environment.

Facilitates regulatory filings
Gibco CTS products are manufactured at a site that uses methods and controls that conform with cGMP for medical devices, 21 CFR Part 820. Our FDA-registered manufacturing site is ISO 13485- and ISO 9001-certified. Simplifies transition from bench to clinic
Like RUO Essential 8 Medium, CTS Essential 8 Medium contains only the eight essential components needed for stem cell culture, but is AOF. Combined with cGMP manufacturing, this simple, fully-defined formulation improves consistency compared to other media that contain over 20 highly variable ingredients. The close similarity between Essential 8 Medium and CTS Essential 8 Medium enables a smooth transition from one medium to the other.

CHO-S-SFM II, with hypoxanthine and thymidine (Gibco™)

CHO-S-SFM II, is a complete, serum-free, low-protein (less than 100 µ/ml) medium optimized for the growth and maintenance of Chinese hamster ovary (CHO) cells in suspension culture for production of recombinant proteins. It is supplemented with hypoxanthine and thymidine (the non-supplemented equivalent is also available). This ready-to-use medium is suitable for the suspension culturing of CHO and other cells in batch, continuous, and perfusion culture systems. A CHO-S-SFM II medium Drug Master File (DMF) has been submitted to the FDA. Permission to cross-reference the Master File may be obtained by contacting Technical Services or your local Sales Representative.

This medium contains components of animal origin. If growth factors or hydrolysates, found in competitor media, are to be avoided, consider CD CHO, CD OptiCHO™ or CD FortiCHO™ media.

CD FortiCHO™ Medium (Gibco™)

CD FortiCHO™ medium is specifically designed to offer the highest batch and fed-batch culture performance and yield with recombinant CHO cells in a chemically defined environment. The protein-free, animal origin component-free formulation provides consistency and helps reduce the need to screen for adventitious agents. With CD FortiCHO™ medium cell cultures last 2+ days longer than competitive alternatives, which means decreased HCP (Host Cell Protein) concentration for removal in downstream purification.

CD FortiCHO™ medium is fortified to support maximal recombinant CHO cell growth and protein expression in suspension batch culture, offering 120% greater growth and production than select competitive media with simple glucose supplementation. HuMEC Basal Serum-Free Medium (1X) (Gibco™)

HuMEC Basal Serum-Free Medium is designed to work with HuMEC Supplement Kit (SKU# 12755013). Together, these products are engineered specifically for the culture of human mammary epithelial cells (SKU# A10565). This translates to greater experimental validity, more reproducible results, and reduction of lot-to-lot variability and aberrations that can occur with the addition of serum. Human mammary epithelial cells cultured in HuMEC Medium have gene expression profiles consistent with normal mammary epithelial cells, as tested on a cDNA microarray.

Applications:
HuMEC Medium is optimized specifically for the growth of human mammary epithelial cells, and has been performance tested in a growth assay using normal human mammary epithelial cells. HuMEC Medium promotes superior growth of normal human mammary epithelial cells, making it ideal for researchers in breast cancer, drug discovery, and other aspects of breast biology.

Contents and Storage:
HuMEC Basal Serum Free Medium should be supplemented with HuMEC Supplement Kit. HuMEC Supplement Kit and HuMEC Basal Serum Free Media can be ordered together as HuMEC Ready Medium (SKU# 12752010). The HuMEC Supplement Kit includes 5 ml of a supplement mix containing epidermal growth factor, hydrocortisone, isoproterenol, transferrin, and insulin, and 25 mg of bovine pituitary extract. Store the basal medium at +2°C to +8°C.

Hybridoma-SFM, powder (Gibco™)

Gibco® Hybridoma SFM is a serum-free, very low-protein medium specifically developed for the ability to support the growth of hybridomas for antibody production. Gibco® Hybridoma SFM features:
• Ability to support suspension and stationary hybridomas and myelomas
• Complete, ready-to-use formulation
• Serum-free, very low-protein formulation

Ability to support suspension and stationary hybridomas and myelomas
Suitable for the culture of recombinant myeloma lines as well as traditional hybridomas, Gibco® Hybridoma SFM provides high cell densities and IgG production. Cholesterol-dependent cultures such as NS0 require the addition of 250x Cholesterol Lipid Concentrate.
