Essential 6™ Medium (Gibco™)

Essential 6 Medium is a feeder-free and xeno-free medium that supports the reprogramming of somatic cells and the spontaneous or directed differentiation of human pluripotent stem cells (PSCs). In addition, Essential 6 Medium can be used as a base for custom media for the culture of PSCs. The formulation is based on a medium originally developed by Guokai Chen et. al. (1) in the laboratory of James Thomson and published as "E6." With only six essential components, Essential 6 Medium helps minimize variability. To complete your workflow with a matching reduced-variability PSC culture medium developed by the same lab, use Essential 8 Medium.

Essential 6 Medium enables:
Differentiation—does not contain bFGF, which inhibits differentiation
Reprogramming—does not contain TGFβ, which has a negative effect on reprogramming efficiency
Flexibility—provides a flexible format where the levels of TGFβ and bFGF can be adjusted and additional components can be added to match a given application

Differentiation
Unlike other media used in PSC culture, Essential 6 Medium does not contain bFGF or TGFβ. As such, Essential 6 Medium can support the formation of embryoid bodies. It has also been used as a base for the directed differentiation of various cell types in the endodermal, mesodermal, and ectodermal lineages (2), including motor neurons (3).

Reprogramming
Essential 6 Medium allows for defined and feeder-free reprogramming when used with bFGF. The formulation supports somatic cell reprogramming using a variety of methods, including episomal vectors (4) and CytoTune (Sendai virus), and is optimized to help ensure maximum cell health and pluripotency with minimal variability.

Flexibility
Essential 6 Medium is xeno-free and contains only the essential components needed for stem cell culture, minus bFGF and TGFβ. This provides a basal medium that will maximize cell health and pluripotency while allowing levels of TGFβ and bFGF to be adjusted and additional components to be added to match a given application.

Quality and performance testing
Every lot of Gibco® AmnioMAX™ - II Complete Medium is performance tested by a certified US reference cytogenetics laboratory to ensure consistently superior performance. Pooled primary human amniotic fluid samples are cultured for six days in Gibco® AmnioMAX™ - II Complete Medium before measuring the total number of colonies and total number of mitotic colonies. In addition, each lot is tested for sterility, pH and osmolality.

Unique, optimized formulation
Gibco® AmnioMAX™ - II Complete Medium contains Fetal Bovine Serum (FBS), gentamicin, and L-glutamine to maximize cell attachment and growth. This optimized medium also has an enhanced buffering system that provides greater pH stability during culture manipulations.

Sf-900™ II SFM, no methionine, no cystine (Gibco™)

Sf-900™ II SFM (without methionine and cystine) is a serum-free, protein-free insect cell culture medium optimized for the growth and maintenance of Spodoptera frugiperda (Sf9 and Sf21) cells and for large-scale production of recombinant proteins expressed using the baculovirus expression vector system (BEVS). This medium is suitable for suspension and monolayer culture methods and supports growth of other lepidopteran cell lines. Gibco® Sf-900™ II SFM features:

Superior long-term, high-density growth
Spodoptera frugiperda (Sf9) cells grown in Sf-900™ II SFM achieve maximum cell densities of 9 to 12 × 106 cells/mL, a significant improvement over competitors' formulations and Grace’s medium (see Product Manual). Increases in maximum cell densities of 20–100% are also observed with the Lymantria dispar (Gypsy moth) and Trichoplusia ni (Tn-368; Cabbage Looper) cell lines. Gibco® Sf-900™ II SFM is capable of supporting the cultures to >20 passages.

Optimized for recombinant protein production
Traditionally, Grace's medium supplemented with 10% FBS has been used for recombinant protein expression. Sf-900™ II SFM is an improved serum-free, protein-free medium designed for growth of Sf9 and other lepidopteran cell lines and production of insect virus and rDNA proteins.

Serum-free, protein-free, ready-to-use formulation
Gibco® Sf-900™ II SFM is a serum-free, protein-free medium that allows for much easier purification of your protein of interest. Sf-900™ II SFM is ready to use; it does not require addition of serum, glutamine, or surfactants. Cells adapted to other commercially available serum-free media can be subcultured directly into Sf-900™ II SFM, usually without any further adaptation. Cells usually require some adaptation from serum-containing formulations.

Scalable in bioreactors
The utility of Sf-900™ II SFM in larger-scale cell culture systems was demonstrated with a 5 L Celligen™ bioreactor. Successful infections with rAcNPV were carried out producing rβ-Gal and rEPO (see Product Manual).

Product use
Customers using Gibco® Sf-900 II SFM in a manufacturing process, who have a submission with the FDA, may request a letter of authorization from us to reference our Type II Drug Master File (DMF).

cGMP manufacturing and quality system
For supply chain continuity, we manufacture Gibco® Sf-900 II SFM at two separate facilities located in Grand Island, NY and Scotland, UK. Both sites are compliant with cGMP manufacturing requirements, are certified to ISO 13485, and are registered with the FDA as medical device manufacturers.

ExpiSf™ CD Medium (Gibco™)

ExpiSf CD Medium is a GMP-manufactured, chemically defined, yeastolate-free, serum-free, protein-free, animal origin-free medium specifically designed for high-density growth, transfection, and baculovirus infection of suspension-adapted Sf9 (Spodoptera frugiperda) insect cells.

ExpiSf CD Medium:
• Delivers high reproducibility and lot-to-lot reliability
• Facilitates high-level expression of baculovirus-infected cultures, achieving protein yields three times higher than traditional insect expression platforms

ExpiSf CD Medium is a core component of the ExpiSf Expression System. For highest level protein expression, the medium is used in combination with a matched protein expression enhancer, ExpiSf Enhancer, which is supplied as part of the ExpiSf Protein Production Kit. Sf21 cells adapted to other commercially-available serum-free media may be adapted for growth in ExpiSf CD Medium; although, sequential adaptation is required and high-density growth isn’t guaranteed. ExpiSf CD Medium is not recommended for adherent cell cultures. Cells are typically grown in Erlenmeyer-style flasks on a shaker platform in a non-humidified, non-CO2 cell culture incubator.

cGMP manufacturing
ExpiSf CD Medium is manufactured using an ISO 13485-certified process according to 21 CFR 820 medical device current good manufacturing practices (cGMP).

Dry media formulations

AIM V™ Medium (research grade), AlbuMAX™ Supplement (Gibco™)

Contains AlbuMAX® supplement (bovine serum albumin) substituted for human serum albumin. Contains L-glutamine, streptomycin sulfate at 50 µg/ml, and gentamicin sulfate at 10 µg/ml.

Classic formulation
Gibco® PB-MAX™ Karyotyping Medium is an optimized RPMI 1640 medium, supplemented with Fetal Bovine Serum (FBS), L-glutamine, and phytohemagglutinin (PHA). Gibco® PB-MAX™ Karyotyping Medium is based on formulations referenced in the Association for Cytogenetic Technologists Laboratory Manual for the culture of peripheral blood lymphocytes for cytogenetic analysis. Ability to support suspension growth and transfection of 293-F cells
Gibco® 293-F, 293-H, and FreeStyle™ 293-F cells in suspension can be cultured directly into Gibco® FreeStyle™ 293 Expression Medium without adaptation. The FreeStyle™ 293 Expression System is a complete, suspension cell culture system for generating large amounts of mammalian recombinant protein (see figure). Depending on the impeller design and speed, it may be necessary to supplement Gibco® FreeStyle™ 293 Expression Medium with additional Pluronic® F-68 to avoid sheer stress in the culture.

The formulation of this product is identical to research-grade LV-MAX Production Medium, but CTS LV-MAX Production Medium is validated for clinical and commercial therapeutic applications.

CD CHO AGT™ Medium (Gibco™)

CD CHO AGT™ (Advanced Granulation Technology) medium is a protein-free, chemically-defined medium optimized for the growth of Chinese hamster ovary (CHO) cells and expression of recombinant proteins in suspension culture. The AGT™ dry media format is designed to provide increased consistency and productivity across all stages of production from development to commercial manufacturing. CD CHO AGT™ medium contains no proteins or peptide components of animal, plant, or synthetic origin, as well as no undefined lysates or hydrolysates.

When to Use CD CHO Medium
• Your GS CHO cells are recommended to be grown in CD CHO medium
• Your cells have been successfully cultured in CD CHO medium in the past and your process is nearing a regulatory filing (DMF available)

When to Consider Another Gibco® Medium

Consider CD FortiCHO™ medium when:
• The cell line you are using is a transfected CHO K1, GS CHO or CHO-S™ cell line
• Maximum batch culture cell densities and protein titers are needed
• You have time to adapt cells into a new medium in an effort to maximize titers
• Cell health at harvest is a priority for downstream processing

Grace's Insect Medium, supplemented (Gibco™)

Grace's Insect Medium, Supplemented, was originally designed for the growth of cells from the Australian Emperor Gum Moth, Antherea eucalypti. Grace's Supplemented Insect Medium is now widely used for the growth of Spodeptera frugiperda cells, Sf9 and Sf21.

Sf9 and Sf21 cells are routinely used to express recombinant protein using the Baculovirus Expression Vector System (BEVS). We offer a variety of Gibco® media options for a range of insect cell culture applications.

Formulation
This Grace's Insect Medium contains lactalbumin hydrolysate, yeastolate, L-glutamine, and sodium bicarbonate. The complete formulation is available. Grace's Supplemented Insect Medium contains lactalbumin hydrolysate and yeastolate not commonly found in the unsupplemented version.

Dual site cGMP manufacturing and quality system
Gibco® Grace's Supplemented Insect Medium is manufactured at a cGMP-compliant facility located in Grand Island, New York. The facility is registered with the FDA as a medical device manufacturer and is certified to ISO 13485 standards. For supply chain continuity, we offer an identical product made in our Scotland facility (11605-045). Grace's Supplemented Insect Medium contains lactalbumin hydrolysate and yeastolate, and is commonly supplemented with 10% Fetal Bovine Serum (FBS) before use. Grace's Supplemented Insect Medium requires ambient CO2 in order to maintain physiological pH.

CD OptiCHO™ medium is specifically designed to offer high performance and yield with recombinant CHO cells in a chemically defined fed-batch environment. The protein-free, animal origin free formulation is designed to provide consistency and reduces the need to screen for adventitious agents. Medium 106 (Gibco™)
