Pierce™ Protein Concentrators PES, 50K MWCO, 5–20 mL Thermo Scientific™

Thermo Scientific Pierce Protein Concentrators PES are disposable ultrafiltration centrifugal devices with a polyethersulfone (PES) membrane for concentration, desalting, and buffer exchange of biological samples. This product has a molecular weight cutoff (MWCO) of 50K and can be used for processing sample volumes ranging from 5 to 20 mL.

Features of Pierce Protein Concentrators PES:
Rapid protein concentration—sample concentration of 10- to 30-fold can be achieved in 5 to 30 minutes for 10K MWCO (time may vary for a product with a different MWCO)
Excellent protein recovery—typical protein recovery is >90%
Versatility—perform sample concentration, diafiltration, or buffer-exchange of biological samples
Compatibility—may be used in both fixed angle and swing bucker rotors; collect and recover sample without invert spinning

For easy identification, the MWCO value is etched on the side of each device. To assist with sample addition and recovery, a clear window with graduations is located on the side of each device enabling visual determination of the retentate volume. The vertical design of the membrane allows high flux rates, low nonspecific binding, and negligible protein aggregation on the membrane; and it offers reliable and consistent results.

Pierce Protein Concentrators PES contain a membrane rated for retaining molecules with a molecular weight at least 2-fold greater than the MWCO rating of the PES membrane within the device. Reduced recovery may occur when using a concentrator with molecules smaller than the recommended mass. Protein recovery will vary depending on the specific protein in the sample and its starting concentration. To achieve >90% recovery of protein, the minimum protein sample concentration should be 0.1 mg/mL.

Pierce™ Centrifuge Columns, 2 mL Thermo Scientific™

Thermo Scientific Pierce Centrifuge Columns are convenient tools for handling 40 µL to 10 mL of an affinity support for purification.

Column details:

• Made from low protein binding polypropylene for compatibility with protein purification
• Total volume: 5 mL (2 mL resin bed, 3 mL reservoir)
• Resin volume: 2 mL
• Filter type, pore size: Polyethylene filter, ~30 µm pore size
• Receiver tube: Fits standard 15 mL conical centrifuge tubes
• Caps: Screw-top cap
• Twist-off bottom and press-on cap to reseal

Simply add the affinity resin to one of the polypropylene columns, remove the twist-off bottom and allow the resin to pack itself. Then add your sample and allow it to bind to the support and use a centrifuge to efficiently wash away any contaminants and elute your purified protein. Pierce Centrifuge Columns can be used in either spin or gravity-flow format. The spin format reduces sample processing time and makes column washing and elution more efficient. For gravity-flow procedures and when slow processing time is desired for binding steps with dilute samples, Column Extenders can be used to increase the column reservoir capacity. Use the any combination of processing techniques to get the best possible chromatography results.

Applications:

• Affinity purification, affinity chromatography
• Immunodepletion
• Ion exchange resins
• Gel filtration, desalting, buffer exchange

Antibody Conjugate Purification Kit, 50-100 µg conjugate Invitrogen™

Molecular Probes® Antibody Conjugate Purification kits efficiently purify antibody conjugates from unconjugated dye or biotin. This particular kit is optimized for the purification of 50–100 µg of antibody conjugate. Purification is accomplished using an easy-to-assemble size-exclusion spin column. Each kit contains sufficient material for 5 separate purifications, and can also be used to purify high-molecular weight proteins (> 30 kDa) away from unconjugated labels.

Additional Features of this Antibody Conjugate Purification Kit:

• Purified conjugate ready-to-use in ~45 minutes with ~15 minutes of hands-on time

• Designed for low-molecular weight labels (<3000 Da, e.g. Alexa Fluor® dyes); cannot be used for removing unconjugated high-molecular weight labels (>10 kDa) such as enzymes (HRP) or proteins (R-phycoerythrin)

Centri-Sep™ Spin Columns Invitrogen™

Centri-Sep™ Spin Columns are used for the fast and efficient purification of nucleic acids. The Centri-Sep™ gel provides excellent recovery of DNA fragments larger than 16 base pairs, while removing greater than 98% of salts, NTPs, and other low molecular weight impurities. Features of Centri-Sep™ columns:

• Processes samples faster than ultrafiltration columns
• Achieves rapid and efficient separation
• Permits use of custom buffers
• Convenient 20–100 µL sample size
• Dry gel format
• Room temperature storage
• Microcentrifuge format

Designed for performance
The column design is based on the description of gel filtration for the purification of DNA from nick translation reactions by Sambrook, et.al. Each Centri-Sep™ unit consists of a specially fritted microfuge tube, dry gel, a wash tube, and a sample collection tube, all specifically designed for this purpose.

Excellent recovery
The Centri-Sep™ gel provides excellent recovery of DNA fragments with sizes greater than 16 base pairs or a 25-mer, while removing >98% of salts, NTPs, and other unwanted low molecular weight impurities.

Versatile
The column gel is hydrated with either reagent-grade water or a buffer of your choice, and spun in a microcentrifuge or swinging-bucket centrifuge to remove the intersititial fluid. Your sample is then applied to the column and spun again, leaving you not only with a sample purified through removal of low molecular weight components, but also exchanged into the buffer of your choice.
