This pre-diluted version of the matrix can be taken directly from the refrigerator and dispensed into cell culture plates without further handling. Geltrex hESC-Qualified Ready-To-Use basement membrane matrix is optimized for stem cell culture applications using StemPro hESC SFM. AlgiMatrix™ 3D Culture System, 24-well plate (Gibco™)

The Algimatrix™ 3D Culture System provides a three-dimensional matrix that has the potential to create cell culture conditions that more closely reflect normal cell morphology and behavior. It has relevance for both academic and industrial researchers who want to culture hepatocytes and other spheriod forming cells. Algimatrix™ alginate sponge is provided in a 24 well format that fits well into the workflow of both lower and higher-throughput users. The larger well size enables growth of more cells for downstream applications such as transfection.

AlgiMatrix™ 3D Culture System, 6-well plate (Gibco™)

AlgiMatrix™ 3D Culture System 6 well plate is a user-friendly, animal –free bioscaffold available for the development of high-fidelity cell culture models that are more predictive of disease states and drug responses. CTS™ Vitronectin (VTN-N) Recombinant Human Protein, Truncated (Gibco™)

CTS™ (Cell Therapy Systems) Vitronectin (VTN-N) Recombinant Human Protein provides a defined surface for feeder-free culture of human pluripotent stem cells (PSCs). When used with Essential 8™ Medium (Cat. No. A1517001), CTS Vitronectin has been proven to maintain pluripotency and normal growth characteristics in multiple PSC lines. In addition, CTS Vitronectin has been shown to support PSC growth for >8 passages without any signs of karyotypic abnormalities and to maintain the ability of PSCs to differentiate into all three germ line lineages. The VTN-N variant of vitronectin supports human pluripotent stem cell attachment and survival better than wild-type vitronectin when used in conjunction with Essential 8 Medium (1).

Animal origin–free system
CTS Vitronectin is animal origin–free at the primary level to help reduce contamination risk and variability associated with animal-origin raw materials compared to human plasma–derived vitronectin and standard basement membrane extracts (BMEs).

Optimized and defined surface
Unlike standard basement membrane extracts (BMEs), CTS Vitronectin is based on the N-variant design of vitronectin (VTN-N) from the laboratory of James Thomson for use with Essential 8 Medium (1) and has been shown to support human pluripotent stem cell attachment and survival better than wild type vitronectin. CTS Vitronectin provides an optimal environment for feeder-free pluripotent stem cell culture.

Seamless transition from bench to clinic
PSC cultures expanded in other feeder or feeder-free systems can be transitioned to Essential 8 Medium and CTS Vitronectin with minimal adaptation.

Poly-D-Lysine (Gibco™)

Poly-D-Lysine is a chemically synthesized extracellular matrix used to facilitate cell adhesion to tissue culture-treated plastic and glass surfaces. Poly-D-Lysine contains polymers with a molecular weight of 50,000–150,000 Daltons, which makes this product ideal for neuronal culture applications. This ready-to-use product is provided as a liquid at a concentration of 0.1 mg/mL. When used on Nunc cell culture plastics with B-27 supplements or the B-27 Plus Neuronal Culture System, Poly-D-Lysine maintains cell viability and normal growth characteristics in multiple neuronal cell types.

Recombinant human Laminin 521 (rhLaminin-521) is a recombinant human protein that provides a defined surface for feeder-free culture of human pluripotent stem cells (PSCs). When used with Essential 8™ Medium, rhLaminin-521 has been proven to promote cellular survival under stressful conditions in the absence of small molecule inhibitors. rhLaminin-521 has been shown to support PSC growth for >20 passages without any signs of karyotypic abnormalities and to maintain the ability of PSCs to differentiate into all three germ line lineages. rhLaminin-521 also provides optimum reprogramming efficiency of human somatic cells as well as human pluripotent stem cell survival during feeder-dependent to feeder-free transitions when used in conjunction with Essential 8 Medium. The major components of Geltrex™ matrix include laminin, collagen IV, entactin, and heparin sulfate proteoglycans. It is ideal for the promotion and maintenance of differentiated phenotypes in a variety of cell cultures including primary epithelial cells, endothelial cells, smooth muscle cells, and human induced pluripotent stem cells (iPSC). Geltrex™ matrix offers, on average, a 2-fold reduction in several growth factors. AlgiMatrix™ Firming Buffer (Gibco™)

AlgiMatrix™ Firming Buffer is for use with the AlgiMatrix™ 3D Culture System to facilitate the formation of firm sponges in the 96 well and 24 well plate formats. This buffer allows the users to adjust the firmness of the sponge to meet specific cell and experimental requirements. Should be ordered as a separate component.
