CTS™ Essential 6 Medium

CTS Essential 6 Medium is a cGMP-manufactured, xeno-free, feeder-free medium that supports the reprogramming of somatic cells and the spontaneous or directed differentiation of human pluripotent stem cells (PSCs). The formulation is based on a medium originally developed by Guokai Chen, et al., in the laboratory of James Thomson (1). With only six essential components, CTS Essential 6 Medium helps minimize variability. Provides a flexible format
CTS Essential 6 Medium does not contain bFGF or TGFβ and so can support the formation of embryoid bodies. This medium can support the directed differentiation of various cell types and somatic cell reprogramming using a variety of methods.

CTS Essential 6 Medium is manufactured at a site that uses methods and controls that conform with cGMP for medical devices, 21 CFR Part 820. Simplifies transition from bench to clinic
CTS Essential 6 Medium contains the same six essential components as RUO Essential 6 Medium, and with its cGMP manufacturing process, this fully-defined formulation helps minimize variability. Like RUO Essential 6 Medium, CTS Essential 6 Medium does not contain bFGF or TGF-beta, which inhibits differentiation and negatively affects reprogramming efficiency, respectively. CTS™ Essential 8™ Medium (Gibco™)

CTS Essential 8 Medium is an animal-origin-free (AOF), feeder-free medium formulated for the growth and expansion of both embryonic and induced human pluripotent stem cells (PSCs). Essential 8 Medium is the most defined PSC medium as described in the original publication by Guokai Chen et al. in the laboratory of James Thomson. CTS Essential 8 Medium enables the transition from bench to clinic through the direct replacement of research-use-only (RUO) Essential 8 Medium.

Reduces risks from animal-origin material
CTS Essential 8 Medium contains no animal- or human-derived components at the primary component level, thereby reducing the risks associated with animal-origin components. It is the only commercially available AOF medium for pluripotent stem cell culture: Competitors’ similar-grade products are only xeno-free, containing human serum albumin (HSA), which brings additional risks in a clinical environment.

Simplifies transition from bench to clinic
Like RUO Essential 8 Medium, CTS Essential 8 Medium contains only the eight essential components needed for stem cell culture, but is AOF. Combined with cGMP manufacturing, this simple, fully-defined formulation improves consistency compared to other media that contain over 20 highly variable ingredients. The close similarity between Essential 8 Medium and CTS Essential 8 Medium enables a smooth transition from one medium to the other.

MesenPRO RS™ Medium (Gibco™)

MesenPRO RS™ Medium is a reduced serum (2%) medium specifically formulated to support the growth of human mesenchymal stem cells (MSCs) in culture. MesenPRO RS™ Medium consistently improves MSC expansion compared with conventional serum-supplemented media. Using MesenPRO RS™ Medium, MSCs can be expanded for multiple passages while maintaining their multipotential phenotype (i.e., differentiation into osteogenic, chondrogenic, and adipogenic lineages).

MesenPRO RS™ Medium consists of:

• Basal liquid medium (MesenPRO RS™ Basal Medium)
• Frozen supplement (MesenPRO RS™ Growth Supplement), which is added at the time of use.

StemPro™ hESC SFM (Gibco™)

Developed for human embryonic and human induced pluripotent stem cell culture

StemPro® hESC SFM is a fully defined, serum- and feeder-free medium (SFM) specially formulated for the growth and expansion of human embryonic (hESCs) and human induced pluripotent stem cells (iPSC). StemPro® hESC SFM has been extensively tested and is proven to maintain pluripotency in a growing list of hESC lines including BG01, BG02, BG03, H1, H9, and BG01V. In addition, StemPro® hESC SFM has been used to scale up production of hESCs to over 1 x 1010 cells. StemPro® hESC SFM has been shown to support hESC and iPSC growth for >50 passages without any signs of karyotypic abnormalities, and to maintain the ability of hESCs to differentiate into all three germ line lineages.

StemPro™ HSC Expansion Medium (Prototype) (Gibco™)

StemPro HSC Expansion Medium (Prototype) is a xeno-free, serum-free medium specifically formulated to support expansion of hematopoietic stem cells (HSCs). The phenotype targeted for expansion is the total CD34+ cell population, with enrichment of the CD34+CD90+CD45RA– cell subset. StemPro HSC Expansion Medium (Prototype) can be used to study hematopoietic cells from human mobilized peripheral blood, bone marrow, or cord blood, and is manufactured with components of human and recombinant origin.

StemPro HSC Expansion Medium (Prototype) consists of a liquid basal medium and frozen supplement, which is added at the time of use. The medium does not contain growth factors or cytokines.

Features of StemPro HSC Expansion Medium (Prototype) include:

Superior expansion and enrichment
CD34+ cells and the CD34+CD90+CD45RA– subset are found in low proportions in mobilized peripheral blood, bone marrow, and cord blood. These cells are important for proper hematopoietic and immune function, and are studied in a variety of research areas, including transplantation biology, stem cell biology, and hematopoietic development. StemPro HSC Expansion Medium (Prototype) enables superior expansion of not only the total CD34+ compartment, but also enriches for CD34+CD90+CD45RA– stem cells, increasing the quantity of cells available for research (see figures below).

Maintenance of functionality
Functionality of expanded HSCs is critical. The most widely accepted method for functional testing of HSCs is engraftment in mouse models. However, in vivo assays are not always practical, so in vitro methods are often used as a surrogate indication of functionality. One popular in vitro functional test is the colony forming unit (CFU) or colony forming cell (CFC) assay. The CFU assay assesses the proliferation and differentiation potential of HSCs by measuring their ability to form colonies when seeded in a semi-solid medium. StemPro HSC Expansion Medium (Prototype) maintains in vitro differentiation capacity to granulocytic, monocytic, erythroid, and megakaryocytic lineages (see figure below).

Xeno-free, serum-free
Xeno-free and serum-free conditions are important features of media aimed at future clinical use. StemPro HSC Expansion Medium (Prototype) contains only recombinant and human-origin components, allowing for xeno-free culture of CD34+ cells. In addition, the medium does not contain serum, nor does it require addition of serum.

StemPro™ MSC SFM XenoFree (Gibco™)

GIBCO® StemPro® MSC SFM XenoFree has been developed for the growth and expansion of human mesenchymal stem cells (MSCs) and Adipose-derived Stem Cells (ADSCs) under completely serum-free and xenogeneic (xeno)-free conditions. Using StemPro® MSC SFM XenoFree, human MSCs or ADSCs can be expanded for multiple passages while maintaining their multipotent phenotype (i.e. ability to differentiate into osteogenic, chondrogenic and adipogenic lineages). StemPro® MSC SFM XenoFree is cGMP compliant which allows for traceability and manufacturing reliability.

Essential 6™ Medium (Gibco™)

Essential 6 Medium is a feeder-free and xeno-free medium that supports the reprogramming of somatic cells and the spontaneous or directed differentiation of human pluripotent stem cells (PSCs). In addition, Essential 6 Medium can be used as a base for custom media for the culture of PSCs. The formulation is based on a medium originally developed by Guokai Chen et. al. (1) in the laboratory of James Thomson and published as "E6." With only six essential components, Essential 6 Medium helps minimize variability. To complete your workflow with a matching reduced-variability PSC culture medium developed by the same lab, use Essential 8 Medium.

Essential 6 Medium enables:
Differentiation—does not contain bFGF, which inhibits differentiation
Reprogramming—does not contain TGFβ, which has a negative effect on reprogramming efficiency
Flexibility—provides a flexible format where the levels of TGFβ and bFGF can be adjusted and additional components can be added to match a given application

Differentiation
Unlike other media used in PSC culture, Essential 6 Medium does not contain bFGF or TGFβ. As such, Essential 6 Medium can support the formation of embryoid bodies. It has also been used as a base for the directed differentiation of various cell types in the endodermal, mesodermal, and ectodermal lineages (2), including motor neurons (3).

Reprogramming
Essential 6 Medium allows for defined and feeder-free reprogramming when used with bFGF. The formulation supports somatic cell reprogramming using a variety of methods, including episomal vectors (4) and CytoTune (Sendai virus), and is optimized to help ensure maximum cell health and pluripotency with minimal variability.

Flexibility
Essential 6 Medium is xeno-free and contains only the essential components needed for stem cell culture, minus bFGF and TGFβ. This provides a basal medium that will maximize cell health and pluripotency while allowing levels of TGFβ and bFGF to be adjusted and additional components to be added to match a given application.

Essential 8™ Flex Medium Kit (Gibco™)

Essential 8™ Flex is a xeno-free and feeder-free medium that supports the culture of pluripotent stem cells (PSCs) without the need for weekend feeding. Essential 8 Flex provides all of the benefits of Essential 8 Medium: minimized variability of components, reliable and robust cultures with a xeno-free, cGMP produced media; along with the added benefit of flexibility. Maintenance of standard PSCs no longer requires being in the lab 7 days a week.

Flexibility
With Essential 8 Flex Medium, PSCs need only be fed on the day of splitting and the day after. If the cells will be left without feeding for more than 48 hours, such as over a weekend, then simply double the feed volume on Friday.

Reliable and robust cultures
Essential 8 Flex Medium has been shown to support PSC growth for >30 passages without any signs of karyotypic abnormalities and to maintain the ability of iPSCs to differentiate into all three germ line lineages.

StemPro® MSC SFM CTS™ is a serum-free medium (SFM) specially formulated for the growth and expansion of human mesenchymal stem cells (MSCs). StemPro® MSC SFM CTS™ enables superior human MSC growth and increased consistency compared to classical serum-supplemented medium (DMEM + 10% FBS). Using StemPro® MSC SFM CTS™, human MSCs can be expanded beyond 5 passages while still maintaining their tri-lineage mesoderm differentiation potential (i.e., ability to differentiate into osteogenic, chondrogenic and adipogenic lineages). Each lot of StemPro® MSC SFM CTS™ is qualified using a human MSC performance assay and human MSCs growing in serum-supplemented medium can be transitioned directly into StemPro® MSC SFM CTS™ with little or no adaptation required. The Essential 8™ Adaptation Kit includes Essential 8™ Medium and recombinant human Laminin 521 (rhLaminin-521) for optimal survival during transition from feeder-dependent to feeder-free culture. rhLaminin-521 has been proven to promote cellular survival under stressful conditions in the absence of small molecule inhibitors. In addition, rhLaminin-521 has been shown to support PSC growth for >20 passages without any signs of karyotypic abnormalities and to maintain the ability of PSCs to differentiate into all three germ line lineages. rhLaminin-521 can also be purchased separately.

PSC Neural Induction Medium (Gibco™)

Gibco® PSC Neural Induction Medium is a serum-free medium that provides high efficiency neural induction of human pluripotent stems cells (PSCs) in only 7 days. Unlike existing methodologies, use of PSC Neural Induction Medium does not require the intermediary step of embryoid body (EB) formation, which adds time, labor, and variability. Neural Stem Cells (NSCs) generated using PSC Neural Induction Medium have high expression of NSC markers and can be further differentiated into other neural cell types.

Generation of High Quality NSCs
Neural stem cells generated using PSC Neural Induction Medium have >80% expression of Sox1, Sox2, and Nestin (NSC markers), and <1% expression of Oct4 (pluripotency marker). In addition, expanded NSCs maintained phenotype, exhibited normal karyotype, and could be differentiated into neurons, astrocytes, and oligodendrocytes.

StemPro™-34 SFM (1X) (Gibco™)

StemPro®-34 SFM is a serum-free medium specifically formulated to support the development of human hematopoietic cells in culture (1). StemPro®-34 SFM can be used to study hematopoietic cells from bone marrow, peripheral blood, or neonatal cord blood. StemPro®-34 SFM is performance-tested on normal human bone marrow.
