StemPro™ MSC SFM XenoFree (Gibco™)

GIBCO® StemPro® MSC SFM XenoFree has been developed for the growth and expansion of human mesenchymal stem cells (MSCs) and Adipose-derived Stem Cells (ADSCs) under completely serum-free and xenogeneic (xeno)-free conditions. Using StemPro® MSC SFM XenoFree, human MSCs or ADSCs can be expanded for multiple passages while maintaining their multipotent phenotype (i.e. ability to differentiate into osteogenic, chondrogenic and adipogenic lineages). PSC Neural Induction Medium (Gibco™)

Gibco® PSC Neural Induction Medium is a serum-free medium that provides high efficiency neural induction of human pluripotent stems cells (PSCs) in only 7 days. Unlike existing methodologies, use of PSC Neural Induction Medium does not require the intermediary step of embryoid body (EB) formation, which adds time, labor, and variability. Neural Stem Cells (NSCs) generated using PSC Neural Induction Medium have high expression of NSC markers and can be further differentiated into other neural cell types.

Rapid Induction of NSCs

Scalable Production of NSCs
PSC Neural Induction Medium is a robust medium that has been shown to generate over 20 million NSCs from 1 million PSCs. The medium has been tested with multiple hPSC lines including hiPSCs derived from use of Episomal iPSC Reprogramming Vectors and the CytoTune®-iPS Sendai Reprogramming Kit.

Generation of High Quality NSCs
Neural stem cells generated using PSC Neural Induction Medium have >80% expression of Sox1, Sox2, and Nestin (NSC markers), and <1% expression of Oct4 (pluripotency marker). In addition, expanded NSCs maintained phenotype, exhibited normal karyotype, and could be differentiated into neurons, astrocytes, and oligodendrocytes.

StemPro™ hESC SFM (Gibco™)

Developed for human embryonic and human induced pluripotent stem cell culture

StemPro® hESC SFM is a fully defined, serum- and feeder-free medium (SFM) specially formulated for the growth and expansion of human embryonic (hESCs) and human induced pluripotent stem cells (iPSC). StemPro® hESC SFM has been extensively tested and is proven to maintain pluripotency in a growing list of hESC lines including BG01, BG02, BG03, H1, H9, and BG01V. In addition, StemPro® hESC SFM has been used to scale up production of hESCs to over 1 x 1010 cells. StemPro® hESC SFM has been shown to support hESC and iPSC growth for >50 passages without any signs of karyotypic abnormalities, and to maintain the ability of hESCs to differentiate into all three germ line lineages.

StemPro™-34 SFM (1X) (Gibco™)

StemPro®-34 SFM is a serum-free medium specifically formulated to support the development of human hematopoietic cells in culture (1). StemPro®-34 SFM can be used to study hematopoietic cells from bone marrow, peripheral blood, or neonatal cord blood. StemPro®-34 SFM is performance-tested on normal human bone marrow. It is manufactured with components of human, recombinant or synthetic origin.

StemPro®-34 SFM consists of a basal liquid medium and frozen supplement (StemPro®-Nutrient Supplement) which is added at the time of use.

CTS™ Essential 8™ Medium (Gibco™)

CTS Essential 8 Medium is an animal-origin-free (AOF), feeder-free medium formulated for the growth and expansion of both embryonic and induced human pluripotent stem cells (PSCs). Essential 8 Medium is the most defined PSC medium as described in the original publication by Guokai Chen et al. in the laboratory of James Thomson. CTS Essential 8 Medium enables the transition from bench to clinic through the direct replacement of research-use-only (RUO) Essential 8 Medium.

CTS Essential 8 Medium:
• Reduces the risks associated with animal origin material through an AOF formulation
• Facilitates regulatory filings through cGMP manufacturing and the availability of documentation

Reduces risks from animal-origin material
CTS Essential 8 Medium contains no animal- or human-derived components at the primary component level, thereby reducing the risks associated with animal-origin components. It is the only commercially available AOF medium for pluripotent stem cell culture: Competitors’ similar-grade products are only xeno-free, containing human serum albumin (HSA), which brings additional risks in a clinical environment.

Facilitates regulatory filings
Gibco CTS products are manufactured at a site that uses methods and controls that conform with cGMP for medical devices, 21 CFR Part 820. Our FDA-registered manufacturing site is ISO 13485- and ISO 9001-certified. Simplifies transition from bench to clinic
Like RUO Essential 8 Medium, CTS Essential 8 Medium contains only the eight essential components needed for stem cell culture, but is AOF. Essential 6™ Medium (Gibco™)

Essential 6 Medium is a feeder-free and xeno-free medium that supports the reprogramming of somatic cells and the spontaneous or directed differentiation of human pluripotent stem cells (PSCs). In addition, Essential 6 Medium can be used as a base for custom media for the culture of PSCs. The formulation is based on a medium originally developed by Guokai Chen et. al. (1) in the laboratory of James Thomson and published as "E6." With only six essential components, Essential 6 Medium helps minimize variability. To complete your workflow with a matching reduced-variability PSC culture medium developed by the same lab, use Essential 8 Medium.

Essential 6 Medium enables:
Differentiation—does not contain bFGF, which inhibits differentiation
Reprogramming—does not contain TGFβ, which has a negative effect on reprogramming efficiency
Flexibility—provides a flexible format where the levels of TGFβ and bFGF can be adjusted and additional components can be added to match a given application

Differentiation
Unlike other media used in PSC culture, Essential 6 Medium does not contain bFGF or TGFβ. As such, Essential 6 Medium can support the formation of embryoid bodies. It has also been used as a base for the directed differentiation of various cell types in the endodermal, mesodermal, and ectodermal lineages (2), including motor neurons (3).

Reprogramming
Essential 6 Medium allows for defined and feeder-free reprogramming when used with bFGF. The formulation supports somatic cell reprogramming using a variety of methods, including episomal vectors (4) and CytoTune (Sendai virus), and is optimized to help ensure maximum cell health and pluripotency with minimal variability.

Flexibility
Essential 6 Medium is xeno-free and contains only the essential components needed for stem cell culture, minus bFGF and TGFβ. This provides a basal medium that will maximize cell health and pluripotency while allowing levels of TGFβ and bFGF to be adjusted and additional components to be added to match a given application.

StemPro® MSC SFM CTS™ is a serum-free medium (SFM) specially formulated for the growth and expansion of human mesenchymal stem cells (MSCs). StemPro® MSC SFM CTS™ enables superior human MSC growth and increased consistency compared to classical serum-supplemented medium (DMEM + 10% FBS). Using StemPro® MSC SFM CTS™, human MSCs can be expanded beyond 5 passages while still maintaining their tri-lineage mesoderm differentiation potential (i.e., ability to differentiate into osteogenic, chondrogenic and adipogenic lineages). Each lot of StemPro® MSC SFM CTS™ is qualified using a human MSC performance assay and human MSCs growing in serum-supplemented medium can be transitioned directly into StemPro® MSC SFM CTS™ with little or no adaptation required. CTS™ StemPro™ HSC Expansion Medium (Gibco™)

CTS StemPro HSC Expansion Medium was designed to help meet regulatory compliance requirements for ancillary materials and provides more functional cells for cell therapy applications. This xeno-free, serum-free medium is manufactured in accordance with cGMP for medical devices (ISO13485 guidelines). The phenotypes of the hematopoietic stem cells (HSCs) targeted for expansion are the total CD34+ cell population with an emphasis on enrichment of the CD34+CD90+CD45RA- cell subset. CTS StemPro HSC Expansion Medium can be used to study hematopoietic cells from human mobilized peripheral blood, bone marrow, or cord blood.

CTS StemPro HSC Expansion Medium consists of a liquid basal medium and a frozen supplement that is added to the basal medium at the time of use. Growth factors or cytokines are not included.

The formulation is serum-free and xeno-free, and USP/EP grade components are preferentially chosen where possible. In addition, each lot undergoes quality control testing to ensure safety and performance for cell and gene therapy applications.

Superior expansion
CD34+ cells and the CD34+CD90+CD45RA- subset are found in low proportions in mobilized peripheral blood, bone marrow, and cord blood. They are important for proper hematopoietic and immune function and are studied in a variety of research areas, including transplantation biology, stem cell biology, and hematopoietic development. CTS StemPro HSC Expansion Medium enables superior expansion of not only the total CD34+ compartment, but also enriches for CD34+CD90+CD45RA- stem cells, providing more cells to work with (see figures below).

Maintenance of functionality
Functionality of expanded HSCs is critical to success in cell and gene therapy applications. The most widely accepted method for functional testing of HSCs is engraftment in mouse models. The theory is that if human cells persist after transplantation into immunodeficient mice, the cells have demonstrated long term repopulation and differentiation capacity, which are both defining factors of stem cells.
