MagMAX™ Cell-Free Total Nucleic Acid Isolation Kit (Applied Biosystems™)

The MagMAX Cell-Free Total Nucleic Acid Isolation Kit is designed for enrichment of all nucleic acid species present in cell-free biological samples such as plasma. The kit utilizes MagMAX magnetic-bead technology, ensuring reproducible recovery of high-quality nucleic acid that is suitable for a broad range of applications, including real-time PCR, digital PCR, and next-generation sequencing.

The magnetic bead–based purification format allows you to easily process a wide range of sample input volumes (1 mL to 6 mL). When used with the KingFisher Duo Prime or KingFisher Flex Magnetic Particle Processor, 6 to 24 plasma samples of 2-mL input volume can be isolated at once. Alternatively, samples can be processed manually with the use of a magnetic stand.

Go beyond cell-free DNA and get the total picture
Isolating and analyzing circulating cell-free total nucleic acid (cfTNA) from plasma and other cell-free samples serves as a non-invasive method to detect and monitor targets associated with certain disease states such as cancer.

The first step in attaining this valuable information is efficient nucleic acid isolation that specifically recovers the circulating nucleic acid species that contains information related to the disease state. The importance of cell-free DNA (cfDNA) is becoming established, but it may not tell the whole story. Cell-free RNA (cfRNA) sequences serve as critical biomarkers as well in the form of microRNAs and gene fusions. This inclusion of the cfRNA with the cfDNA provides an opportunity for researchers to expand their studies and advance human health.

The MagMAX Cell-Free Total Nucleic Acid kit is developed by the same Thermo Fisher Scientific scientists that brought you the MagMAX Cell-Free DNA Isolation Kit. The difference is the inclusion of all cfRNA species in the eluate, allowing you to increase the amount of information available from a single cell-free sample. Just like the cfDNA Isolation Kit, the cfTNA kit is designed for both manual and automated isolation of nucleic acid from a wide sample volume input range of 1 to 6 mL. The phenol-free, magnetic-bead format makes the kit fully amenable to automated isolation.

Thanks to the scalable format that allows for various sample inputs, it is possible to vary the number of samples processed with a single kit. For example, enough reagents are provided to process 50 samples of 2 mL plasma each. Alternatively, with the same kit it is possible to process 25 samples of 4 mL plasma each. Higher and lower input volumes are possible and the number of samples that can be processed will vary accordingly. This unique product attribute allows you to tailor the product to your specific needs.

Benefits of MagMAX magnetic bead–based purification
Magnetic beads offer many benefits compared to other technologies for isolating nucleic acid. Beads bind the DNA more efficiently than glass-fiber filters, resulting in higher and more consistent yields. Additionally, because filters and vacuum manifolds are not used, there is no risk of filter clogging due to cellular particulates in samples. This clogging issue is of particular concern with protein-rich, large-volume samples such as plasma that are commonly used in circulating cell-free nucleic acid studies.

PrepMan™ Ultra Sample Preparation Reagent (Applied Biosystems™)

The PrepMan® Ultra Sample Preparation Reagent was developed for the rapid preparation of DNA template from Gram-negative food-borne pathogens for use in PCR amplification reactions. These samples often have high lipid content that can inhibit PCR amplification of the template. Using a simple boil and spin protocol, PrepMan®Ultra Reagent efficiently inactivates PCR inhibitors, significantly reducing the need to repeat the template preparation step.

PrepMan® Ultra reagent has been used to prepare PCR-ready template from a wide variety of different sources, including bacteria, yeast, filamentous fungi, both from a plate or from liquid culture, mammalian tissue smears, human cells (buccal swab), and mammalian whole blood. The resulting template has been used amplified in allelic discrimination assays, genetically modified organism (GMO) detection and quantification, food pathogen detection, microbial identification, and bacterial strain typing.

TRIzol™ Plus RNA Purification Kit and Phasemaker™ Tubes Complete System (Invitrogen™)

This system includes both the TRIzol™ Plus RNA Purification Kit and Invitrogen™ Phasemaker™ Tubes.

The TRIzol Plus RNA Purification Kit combines the strong lysis capability of TRIzol Reagent with the convenient and time-saving silica-cartridge purification protocol of the PureLink™ RNA Mini Kit to provide ultrapure total RNA typically within an hour from a wide variety of sample types.

Phasemaker Tubes contain a thick liquid polymer that separates and completely isolates the upper aqueous phase of the TRIzol Reagent mix from the organic phase underneath, greatly simplifying the removal of the upper phase containing the RNA. This TRIzol Plus RNA Purification Kit and Phasemaker Tubes Complete System is for the extraction of RNA, not DNA or protein. It will accommodate 50–100 mg of tissue. MagMAX™ CORE Lysis Solution (Applied Biosystems™)

This MagMAX CORE Lysis Solution is made available as a supplement to that included in the MagMAX CORE Nucleic Acid Purification Kit. This proprietary buffer solution is used to break open cells in preparation for downstream molecular detection applications such as real time PCR, RT-PCR, and sequencing applications.

Max Bacterial Enhancement Reagent (Invitrogen™)

The Max Bacterial Enhancement Reagent is designed for use with TRIzol® Reagent to improve the isolation of intact total RNA up to 3-fold from Gram-positive and Gram-negative bacteria. The Max Bacterial Enhancement Reagent is a ready-to-use solution composed of chelating agents, detergent, and a buffer, and is used as an efficient pre-treatment buffer for bacterial cells prior to RNA isolation with TRIzol® Reagent. The use of Max Bacterial Enhancement Reagent with TRIzol® inactivates endogenous RNases and promotes protein denaturation, improving the RNA quality and integrity. The Max Bacterial Enhancement Reagent is available either separately or packaged with TRIzol as part of the TRIzol® Max™ Bacterial RNA Isolation Kit (Cat. No. 16096020 or 16096040).

MagMAX™ Viral/Pathogen Elution Buffer (Applied Biosystems™)

Agarase (0.5 U/µL) (Thermo Scientific™)

Thermo Scientific Agarase is an enzyme that specifically digests the agarose polysaccharide core into neoagaro-oligosaccharides. Agarase allows for gentle yet efficient recovery of DNA or RNA fragments from low melting point agarose. The recovered nucleic acids can be directly used for amplification, cloning, sequencing, etc.

MagMAX™ Cell-Free DNA Lysis/Binding Buffer (Applied Biosystems™)

This buffer is included in the MagMAX Cell-Free DNA Isolation Kit (Cat. No. A29319). It is made available separately for applications that require more Lysis/Binding Buffer than is provided in the kit.

Lysis Buffer 1 + RNase for Food ID

This convenient packaging of Lysis Buffer 1 + RNase for Food ID is recommended for use with the PrepSEQ Nucleic Acid Extraction Kit (Cat. No. 4480466) to prepare DNA for meat ID and GMO testing from food and feed samples. The protocol using Lysis Buffer 1 + RNase in conjunction with the PrepSEQ Nucleic Acid Extraction Kit can be readily automated on the Thermo Scientific KingFisher mL Food Protection Purification System platform to reduce hands-on time and increase reproducibility of the DNA extraction procedure. The purified DNA can then be used in PCR or other molecular biology techniques, and is well-suited for use with the following food and feed testing kits:

TRIzol™ Max™ Bacterial RNA Isolation Kit (Invitrogen™)

The TRIzol® Max™ Bacterial RNA Isolation Kit provides a simple and reliable method to improve the isolation of intact total RNA from gram-positive and gram-negative bacteria. The kit utilizes both the Max™ Bacterial Enhancement Reagent and TRIzol® Reagent to inactivate endogenous RNases and promote protein denaturing, improving RNA quality and integrity. Isolation of RNA takes about an hour.

Two Products in One Kit

TRI Reagent™ Solution (Invitrogen™)

Tri Reagent® solution is a complete, ready-to-use reagent for the isolation of total RNA or the simultaneous isolation of RNA, DNA, and protein from a variety of biological samples. Key features:

Productive—obtain higher yields than with traditional guanidine thiocyanate or cesium chloride methods

Using TRI Reagent® solution
TRI Reagent® starting material can be either fresh or frozen tissue, or cell samples of almost any size—the protocol is easily scalable. This RNA isolation method is well-recognized and referenced in numerous publications, a testament to its reliability.

How it works
TRI Reagent® combines phenol and guanidine thiocyanate in a monophase solution to facilitate the immediate inhibition of RNase activity. Biological samples are homogenized or lysed in TRI Reagent®; the subsequent addition of bromochloropropane or chloroform results in the separation of the homogenate into aqueous and organic phases. RNA partitions to the aqueous phase, DNA to the interphase, and proteins to the organic phase. The RNA can then be precipitated from the aqueous phase with the addition of isopropanol. The entire protocol can be completed in approximately 1 hour.

Applications
The isolated RNA is suitable for any downstream application, including RT-PCR, amplification for array analysis, hybridization assays, or in vitro translation. PCR, Southern blotting, and cloning can be performed using DNA isolated with TRI Reagent®. Isolated proteins can be used in SDS-PAGE and western blotting.

Tri Reagent® solution is incorporated into our RiboPure™ and MagMAX™-96 for Microarrays RNA isolation kits.

MagMAX™ Viral/Pathogen Ultra Enzyme Mix (Applied Biosystems™)
