TRIzol® LS Reagent differs from the standard TRIzol® Reagent in concentration, which permits larger samples to be processed. Just as with the standard TRIzol® Reagent, the integrity of resulting RNA preparations is maintained by the highly effective inhibition of RNase activity during sample homogenization. The simplicity of the TRIzol® LS Reagent method allows simultaneous processing of a large number of samples. The entire procedure can be completed in 1 hour. Total RNA isolated by TRIzol® LS Reagent is free of protein and DNA contamination.

Formulated to serve multiple isolations
TRIzol® LS Reagent allows you to perform sequential precipitation of RNA, DNA, and proteins from a single sample. After homogenizing the sample with TRIzol® LS Reagent, chloroform is added, and the homogenate is allowed to separate into a clear upper aqueous layer (containing RNA), an interface, and a red lower organic layer (containing the DNA and proteins). RNA is precipitated from the aqueous layer with isopropanol. DNA is precipitated from the aqueous/organic interface with ethanol. Protein is precipitated from the phenol-ethanol layer by isopropanol precipitation. The precipitated RNA, DNA, or protein is washed to remove impurities, and then resuspended for use in downstream applications.

Purified products are ideal for use with a variety of applications
Isolated RNA can be used in real-time quantitative PCR (qPCR), northern blot analysis, dot blot hybridization, poly-(A)+ selection, In vitro translation, RNase protection assays, and molecular cloning. Go beyond cell-free DNA and get the total picture
Isolating and analyzing circulating cell-free total nucleic acid (cfTNA) from plasma and other cell-free samples serves as a non-invasive method to detect and monitor targets associated with certain disease states such as cancer.

The first step in attaining this valuable information is efficient nucleic acid isolation that specifically recovers the circulating nucleic acid species that contains information related to the disease state. The importance of cell-free DNA (cfDNA) is becoming established, but it may not tell the whole story. Cell-free RNA (cfRNA) sequences serve as critical biomarkers as well in the form of microRNAs and gene fusions. This inclusion of the cfRNA with the cfDNA provides an opportunity for researchers to expand their studies and advance human health.

The MagMAX Cell-Free Total Nucleic Acid kit is developed by the same Thermo Fisher Scientific scientists that brought you the MagMAX Cell-Free DNA Isolation Kit. The difference is the inclusion of all cfRNA species in the eluate, allowing you to increase the amount of information available from a single cell-free sample. Just like the cfDNA Isolation Kit, the cfTNA kit is designed for both manual and automated isolation of nucleic acid from a wide sample volume input range of 1 to 6 mL. The phenol-free, magnetic-bead format makes the kit fully amenable to automated isolation.

Thanks to the scalable format that allows for various sample inputs, it is possible to vary the number of samples processed with a single kit. For example, enough reagents are provided to process 50 samples of 2 mL plasma each. Alternatively, with the same kit it is possible to process 25 samples of 4 mL plasma each. Higher and lower input volumes are possible and the number of samples that can be processed will vary accordingly. This unique product attribute allows you to tailor the product to your specific needs.

Benefits of MagMAX magnetic bead–based purification
Magnetic beads offer many benefits compared to other technologies for isolating nucleic acid. Beads bind the DNA more efficiently than glass-fiber filters, resulting in higher and more consistent yields. Additionally, because filters and vacuum manifolds are not used, there is no risk of filter clogging due to cellular particulates in samples. This clogging issue is of particular concern with protein-rich, large-volume samples such as plasma that are commonly used in circulating cell-free nucleic acid studies.

This binding buffer is included in the MagMAX Viral/Pathogen Nucleic Acid Isolation kits and MagMAX Microbiome Ultra Nucleic Acid Isolation Kit (Cat. Nos. A42352, A42356, A42357, and A42358). It is made available separately for applications that require more binding buffer than is provided in the kit.

MagMAX™ Viral/Pathogen Proteinase K (Applied Biosystems™)

This Proteinase K is included in the MagMAX Viral/Pathogen Nucleic Acid Isolation kits and MagMAX Microbiome Ultra Nucleic Acid Isolation Kit (Cat. Nos. A42352, A42356, A42357, and A42358). It is made available separately for applications that require more Proteinase K than is provided in the kit.

MagMAX™ FFPE DNA/RNA Ultra Kit (Applied Biosystems™)

The MagMAX™ FFPE DNA/RNA Ultra Kit is designed for sequential isolation of DNA and RNA from the same formaldehyde- or paraformaldehyde-fixed paraffin-embedded (FFPE) tissue sample. The DNA and RNA are recovered in separate eluates, and both are compatible with a broad range of applications, including real-time PCR and next-generation sequencing. The isolation of RNA and DNA from the same FFPE sample makes this kit an ideal method of sample preparation for Oncomine™ Comprehensive and Oncomine™ Focus assays.

Features of the MagMAX FFPE DNA/RNA Ultra Kit include:
• Flexible design that permits both manual and automated isolation of RNA and DNA
• Minimal requirement of 5 µm section or curl input
• Capable of processing 40 µm FFPE sections or curls; greater than 40 µm sections can be processed with an alternative protocol
• Compatible with targeted RNA and DNA sequencing panels
• Contains Dynabeads™ MyOne™ Silane for consistent isolation of RNA, microRNA, and DNA
• Alternative protocols for the isolation of RNA or DNA only

Unlocking the information contained in FFPE samples is essential to cancer research. Common nucleic acid isolation methods require you to choose which nucleic acid to recover from a given sample, with loss of the other nucleic acid (and any information that may have resulted from its analysis). Alternatively, the DNA and RNA are co-purified, or the sample is split in half so that each can be purified.

More information from less sample input
The MagMAX FFPE DNA/RNA Ultra Kit protocol addresses these issues by first isolating DNA from the FFPE sample and then recovering the RNA from the DNA-depleted supernatant. The nucleic acids are prepared as separate eluates and are then ready for downstream analysis. This allows for a fuller analysis of precious samples, including investigation of important biomarkers, such as hotspot mutations, CNVs, gene fusions, and indels.

Benefits of MagMAX magnetic bead–based purification
The magnetic bead–based purification format of the kit allows for easy scaling of the number of samples processed either manually with a magnetic stand or with a magnetic particle processor such as the KingFisher™ Duo Prime or KingFisher™ Flex processor.

Magnetic beads offer many benefits compared to other technologies for isolating nucleic acids. Beads bind the RNA and DNA more efficiently than glass fiber filters, resulting in higher and more consistent yields. Additionally, because filters and vacuum manifolds are not used, there is no risk of undigested tissue particulates clogging these items during the extraction process. This clogging issue is of particular concern with fibrous FFPE tissue samples that that are not fully digested during the proteinase K treatment.

Optimized bead surface chemistry
The Dynabeads MyOne Silane utilized in the MagMAX FFPE DNA/RNA Ultra Kit have an optimized silica-like surface chemistry with high specific surface area that allows efficient kinetics and high sensitivity in nucleic acid capture. These aspects make Dynabeads MyOne Silane ideal for the recovery of both RNA and DNA targets found in FFPE samples. Once captured, both of these nucleic acids are then eluted separately in volumes of 50 µL and are ready for downstream applications.
