PSC Definitive Endoderm Induction Kit (Gibco™)

Gibco™ PSC Definitive Endoderm Induction Kit consists of two xeno-free media that enable efficient induction of human pluripotent stem cells (PSCs) to definitive endoderm. Unlike other methods that require multiple components and take five or more days, the PSC Definitive Endoderm Induction Kit can generate ≥90% CXCR4+/PDGFRα- definitive endoderm cells with only two components in just two days. Each medium is supplied as a 1X complete medium, requiring no mixing in of additional components, and the resultant definitive endoderm is capable of differentiating into midgut/hindgut, pancreatic endoderm, or liver bud progenitors that express relevant physiological markers.

Generation of high quality definitive endoderm

StemPro® Adipogenesis Differentiation Kit has been developed for the adipogenic differentiation of mesenchymal stem cells (MSCs) in tissue culture vessels. The kit contains all reagents required for inducing MSCs to be committed to the adipogenesis pathway and generate adipocytes. Using StemPro® Adipogenesis Differentiation Kit in combination with StemPro® MSC SFM or MesenPRO RS™ Medium provides a standardized culture workflow solution for MSC isolation, expansion and differentiation into lipid vesicle-forming adipocytes.

Floor Plate Cell Expansion Kit (Gibco™)

The Floor Plate Cell Expansion Kit is a component of the Gibco™ PSC Dopaminergic Neuron Differentiation Kit that is also offered separately. It enables additional expansion of floor plate progenitor cells generated using complete Floor Plate Cell Specification Medium (part of the Differentiation kit).

Note: Each additional expansion beyond passage 2 will result in an additional 4–6-fold increase in the total number of floor plate cells without compromising the quality of TH neurons (positive for both TH and FOXA2). Further expansion (up to passage 10) results in stable expansion of progenitor cells, but the resulting TH neurons may show decreased FOXA2 expression.

PSC Dopaminergic Neuron Differentiation Kit (Gibco™)

The Gibco™ PSC Dopaminergic Neuron Differentiation Kit is a culture media system that enables the differentiation of human pluripotent stem cells (hPSCs) into functional midbrain dopaminergic neurons. Unlike other methods of differentiating hPSCs into dopaminergic neurons, which can be lengthy, ill-defined, and produce a biologically irrelevant phenotype, the PSC Dopaminergic Neuron Differentiation Kit allows you to differentiate hPSCs into authentic midbrain dopaminergic neurons with increased speed and unprecedented scalability, all while retaining proper midbrain phenotype throughout.

The kit is a three-step system used over 35 days, with components that are sequentially added to specify PSCs to the midbrain floor plate, expand floor plate progenitor cells, and differentiate floor plate progenitor cells into functional midbrain dopaminergic neurons. Each component is performance tested to ensure generation of the correct phenotype, and the midbrain dopaminergic neurons have been functionally tested for the ability to produce spontaneous action potential and release of dopamine upon depolarization.

Step 1: Specification of PSCs into midbrain floor plate cells
During central nervous system development, midbrain dopaminergic neurons are derived from a distinct population of cells termed midbrain floor plate cells, which are formed during days 21–28 of gestation and located along the ventral midline of the developing neural tube. Recent reports have focused on the importance of differentiating hPSCs into cells from this region, rather than using a more generic neural stem cell population, in order to create authentic midbrain dopaminergic neurons. The first step in the creation of dopaminergic neurons using the PSC Dopaminergic Neuron Differentiation Kit is the specification of hPSCs into midbrain-specified floor plate progenitor (FP) cells in complete Floor Plate Specification Medium.

Step 2: Create a bankable population of floor plate progenitor cells
Standard methods of creating midbrain dopaminergic neurons involve lengthy protocols that are continuous and not scalable. Using the PSC Dopaminergic Neuron Differentiation Kit, a workable bank of floor plate progenitor cells that maintain midbrain identity is created.

The ability to bank progenitor cells in this step enables:
• A significant increase in resultant DA neuron purity
• The ability to scale experiments beyond current limits
• The ability to distribute cells among peers or thaw for later use

Step 3: Differentiation of floor plate precursor cells to midbrain dopaminergic neurons
The final step of the PSC Dopaminergic Neuron Differentiation Kit protocol involves differentiation of floor plate progenitor cells into midbrain dopaminergic neurons. Dopaminergic neurons generated during this step have been shown to maintain midbrain identity, exhibit a gene expression profile equivalent to that of neurons generated using published protocols, and exhibit functionality as shown by spontaneous action potential and release of dopamine upon depolarization.

PSC Cardiomyocyte Differentiation Kit (Gibco™)

The Gibco® PSC Cardiomyocyte Differentiation Kit consists of a set of serum-free and xeno-free media that enable efficient differentiation of human pluripotent stem cells (PSCs) to contracting cardiomyocytes in as few as 8 days. Unlike other methods that require multiple components and longer assay duration, the PSC Cardiomyocyte Differentiation Kit can generate cardiomyocytes from pluripotent stem cells in a ready-to-use media format and in less time.

Comprised of three 1X media that require no thawing or mixing, each medium is used consecutively over a total of 14 days, resulting in functional cardiomyocytes that express relevant physiological markers, contract in culture, and can be subsequently maintained in culture for >15 days (maintenance medium also sold separately (Cat. No. A2920801)).

The PSC Cardiomyocyte Differentiation Kit is comprised of 1X media that do not require frozen storage, so no thawing or mixing of reagents. Simply warm the media and add to PSC culture.

The PSC Cardiomyocyte Differentiation Kit is capable of producing contracting cardiomyocytes in as few as 8 days. Differentiated cardiomyocytes can be subsequently maintained in culture for >15 days.

Generation of high quality cardiomyocytes
Cardiomyocytes generated using the PSC Cardiomyocyte Differentiation Kit are functionally relevant, expressing key markers such as TNNT2, Nkx2.5, MYH6, and α-actinin; and contract in culture.

PSC Cardiomyocyte Maintenance Medium (Gibco™)

Gibco™ Cardiomyocyte Maintenance Medium is a serum-free and xeno-free medium that is capable of maintaining cardiomyocytes that have been differentiated using the PSC Cardiomyocyte Differentiation Kit (Cat. No. A2921201). This medium is included in the kit and sold separately for researchers wishing to maintain differentiated cardiomyocytes in culture for extended periods of time.
