Qubit™ RNA HS Assay Kit (Invitrogen™)

The Qubit RNA HS Assay Kit, when used with the Qubit Fluorometer, provides an accurate and selective method for the quantitation of low-abundance RNA samples. The assay is highly selective for RNA and will not quantitate DNA, protein, or free nucleotides. Common contaminants, such as salts, free nucleotides, solvents, detergents, or protein, are well-tolerated in the assay. The assay kit is designed to be accurate for RNA sample concentrations between 250 pg/µL and 100 ng/µL. The kit provides concentrated assay reagent, dilution buffer, and pre-diluted RNA standards. Simply dilute the reagent using the buffer provided, add your sample (any volume between 1 µL and 20 µL is acceptable), and read the concentration using the Qubit Fluorometer.

Qubit™ microRNA Assay Kit (Invitrogen™)

The Qubit microRNA Assay Kit allows easy and accurate quantification of microRNA (miRNA) using the Qubit Fluorometer, even in the presence of common contaminants such as salts, free nucleotides, solvents, detergents, and protein. The assay is highly selective for miRNA over rRNA or large mRNA (>1000 bp). Although the reagent is not exclusively selective for miRNA, we have been able to reproducibly quantify miRNA in pure samples at levels as low as 0.5 ng in the assay tube following the supplied protocol below, even in the presence of mRNA.

Features of Qubit microRNA assay:
• Accurately detects as little as 0.5 ng miRNA in the assay tube
• Core dynamic range of 5–500 ng/mL
• Accurate results for sample concentrations from 50 ng/mL to 100 µg/mL

The Qubit microRNA Assay Kit provides concentrated assay reagent, dilution buffer, and pre-diluted miRNA standards. Simply dilute the reagent using the buffer provided, add your sample (any volume between 1 µL and 20 µL is acceptable), and read the concentration using the Qubit Fluorometer. Because the assay tolerates 1–20 µL of sample in an assay volume of 200 µL, accurate results can be obtained for initial sample concentrations from 50 ng/mL–100 µg/mL.

Common contaminants, such as salts, free nucleotides, solvents, detergents, and protein, are well-tolerated in the assay. The assay kit is designed to be accurate for initial RNA sample concentrations from 1 ng/µL to 8 µg/µL, providing an assay range from 20 ng –8 µg. The kit provides concentrated assay reagent, dilution buffer, and pre-diluted RNA standards. Simply dilute the reagent using the buffer provided, add the sample (any volume between 1 µL and 20 µL is acceptable), and read the concentration using a Qubit 2.0, 3, or 4 fluorometer.

Notes:

Quant-iT™ RNA Assay Kit (Invitrogen™)

The Quant-iT™ RNA Assay Kit makes RNA quantitation easy and accurate. The kit provides concentrated assay reagent, dilution buffer, and pre-diluted RNA standards. The assay is highly selective for RNA over double-stranded DNA, and in the range of 5-100 ng of RNA, the fluorescence signal is linear. Quant-iT™ DNA Assay Kits (Q33120, Q33130) are also available.

Qubit™ RNA XR Standard (Invitrogen™)

The Qubit RNA XR (extended range) Assay RNA standard is designed for use with Qubit RNA XR Assay Kit. This standard can also be used with the Quant-iT RNA XR Assay Kit (Cat. No. Q33225). It is supplied at the same concentration as the highest concentration RNA standard in the Quant-iT assay and is simply diluted into the standard series. This standard cannot be used as a replacement for the large and/or structured standard in the Qubit RNA IQ Assay. Sufficient material is supplied to perform standards with up to 500 samples.

Qubit™ 1X dsDNA HS Assay Lambda Standard (Invitrogen™)

FluoReporter™ Blue Fluorometric dsDNA Quantitation Kit, 200-2,000 assays (Invitrogen™)

The FluoReporter® Blue Fluorometric dsDNA Quantitation Kit provides the protocols and reagents for analyzing cellular NA with the blue-fluorescent Hoechst 33258 nucleic acid stain. With this kit, quantitation of cellular DNA is rapid, and all manipulations can be carried out in microplate wells. The cells are lysed by freezing them in distilled water, the diluted dye solution is then added to the lysed cells, and fluorescence is measured.

RNA Quantification Kit for SYBR Green I and ROX™ Passive Reference Dye (Applied Biosystems™)

Accurate quantification from limited RNA concentrations

Target preparation assay kits such as GeneChip™ WT Pico Kit and GeneChip 3' IVT Pico Kit allow expression profiling using as little as 100 pg of total RNA (approximately 10 cells). These low amounts of RNA for such target preparation assays require accurate measurement of the quantity of input sample RNA for optimal performance. However, at these low RNA amounts, it becomes virtually impossible to accurately quantify the RNA with standard UV absorbance or fluorescent dye–based techniques.

The RNA quantification kit uses Power SYBR Green RNA-to-CT 1-Step Kit to measure levels of the consistently expressed 18S ribosomal RNA (rRNA) in human, mouse, and rat samples. The amount of 18S rRNA target in the unknown samples is then scaled against a Control HeLa RNA standard curve for accurate quantification that fails using conventional techniques. In addition to the Power SYBR Green RNA-to-CT 1-Step Kit, the kit also includes Control HeLa Total RNA for standard curve preparation and an 18S rRNA Primer Mix, providing an assay range of 0.01 pg–1 ng total RNA.

Quant-iT™ PicoGreen™ dsDNA Reagent (Invitrogen™)

Using the PicoGreen® dsDNA quantitation assay, you can selectively detect as little as 25 pg/ml of dsDNA in the presence of ssDNA, RNA, and free nucleotides (1). The assay is linear over three orders of magnitude and has little sequence dependence, allowing you to accurately measure DNA from many sources, including genomic DNA, viral DNA, miniprep DNA, or PCR amplification products. PicoGreen® dsDNA Quantitation Reagents are:

• Orders of magnitude more sensitive than UV absorbance readings, saving on precious sample (Figure 1)
• Specific for dsDNA in the presence of equimolar amounts of RNA (Figure 2)
• Easy to use-just add the dye to sample, wait 5 minutes, and read
• Suitable for use in 96- and 384-well plate formats
• Compatible with most fluorescence-based microplate readers and fluorometers and Fluorescence

Simply dilute the reagent 1:200, load 200 µL into the wells of a microplate, add 1–20 µL of sample, mix, then read the fluorescence. The assay is performed at room temperature, and the signal is stable for approximately 3 hours. Because the assay tolerates 1–20 µL of sample in an assay volume of 200 µL, accurate results can be obtained for initial sample concentrations from 50 ng/mL–100 µg/mL.

Qubit™ 4 System Verification Assay Kit (Invitrogen™)

The Qubit 4 System Verification Assay Kit is a fast, easy-to-use, reagent-based assay that tests the performance of the Qubit 4 Fluorometer. The kit consists of three components: a blank reagent solution, a green fluorescent reagent, and a far-red fluorescent reagent. The green fluorescent reagent is designed to test the blue excitation (470 nm LED; 430-495 nm excitation filter) and green fluorescence detection (510-580 nm emission filter) on the fluorometer, while the far-red fluorescent reagent is designed to test the red excitation (635 nm LED; 600-645 nm excitation filter) and far-red fluorescence detection (665-720 nm emission filter).

The assay kit provides sufficient reagents to perform up to 50 assays.

Note: The Qubit 4 System Verification Assay Kit can only be used with the Qubit 4 Fluorometer.
