The MicroSEQ ID Purification Combo Kit v2.0 includes reagents for post-PCR and post-sequencing clean-up in the MicroSEQ ID microbial identification workflow. Comparability studies demonstrated equivalent results between the original ExoSAP-IT Reagent and the improved ExoSAP-IT Express Reagent coupled with the MicroSEQ ID Ultra Sequencing Clean-Up kit in the MicroSEQ ID workflow (see figure below). Both ExoSAP-IT and ExoSAP-IT Express enzyme-treated PCR samples produced very similar sequencing results and the correct identification result.

How It Works

ChargeSwitch® technology employs a completely novel mechanism that replaces harsh reagents such as chaotropic salts, ethanol, or organic solvents and isopropanol precipitation with a simple ion-exchange procedure. In this method, DNA binds to a surface ligand that is positively charged at low pH, and is eluted when the pH is raised to 8.5 (Figure 1). The ChargeSwitch®-Pro protocol retains the ease and familiarity of common silica spin-column protocols with binding, washing, and elution steps (Figure 2).


Better Downstream Results

Since ChargeSwitch® technology does not require the use of ethanol, chaotropic salts, or organic solvents for nucleic acid purification, the resulting DNA is free of enzymatic inhibitors and thus shows better downstream performance (Figure 3). These compounds are known to inhibit many enzymatic reactions such as PCR, cloning, restriction enzyme digestion, and array CGH labeling. Expect to see reduced variability in your sequencing applications, with reads in excess of 800 bp with Phred scores >20 (Figure 4); clean restriction enzyme digestions (Figure 5); and removal of dNTPs and primers in excess of 98% (Figure 6).

Rapid, Three Step, pH Based Purification Protocol
The ChargeSwitch® gDNA Plant Kit allows purification of genomic DNA from between 50-100 mg of fungi and plant samples, including leaves, tissues and seeds (Figure 1), to yield up to 7 µg of gDNA typically in less than 15 minutes following sample preparation and lysis. The ChargeSwitch® gDNA Plant Kit uses magnetic beads with a unique, ionizable functional group whose affinity for nucleic acids is pH-dependent to facilitate nucleic acid purification. Genomic DNA is isolated without the use of centrifugation or organic solvents, and is of higher purity than silica-prepared DNA, which is typically contaminated with guanidine salts.

Compatible with Single Tube and High-Throughput Protocols
The ChargeSwitch® gDNA Plant Kit is designed for automated processing of both small numbers of samples individually, as well as large numbers of samples using a liquid handling robot or a multi-channel pipette.

For processing up to 24 individual samples at a time the MagnaRack™ two-piece magnetic separation tube rack is required. This rack consists of a magnetic base station and a removable tube rack, which can hold up to 24 microcentrifuge tubes and can be used with most ChargeSwitch® products.

For high-throughput applications utilizing 96 well blocks, the 96-Well Magnetic Separator is required. This rack can hold and separate up to 96 magnetic bead samples in a deep well plate when placed on top of the magnetic base station.

GeneCatcher™ gDNA Blood Kit, 0.3-1 mL (Invitrogen™)

The GeneCatcher™ gDNA Blood Kits allow rapid and efficient extraction of genomic DNA (gDNA) from human blood, including archived or poorly stored blood samples. Genomic DNA is extracted from blood samples using the cost-effective, user-friendly magnetic bead based GeneCatcher™ Technology without the use of centrifugation (Figures 1,2).

GeneJET Gel Extraction Kit (Thermo Scientific™)

Thermo Scientific GeneJET Gel Extraction Kit is designed for rapid and efficient purification of DNA fragments from standard or low-melting point agarose gels run in either TAE or TBE buffer.

The kit utilizes a proprietary silica-based membrane technology in the form of a convenient spin column. The kit can be used to purify DNA fragments from 25 bp to 20 kb in size with recovery rates up to 95%. Each GeneJET purification column has a binding capacity of up to 25 µg of DNA and can process up to 1 g of agarose gel. Special workflow provides more information from less sample input
FFPE samples are precious, which often requires the choosing of one extraction method over another to save sample. With the RecoverAll Multi-Sample RNA/DNA Workflow, recovering RNA and DNA from the same FFPE tissue sample in a single workflow eliminates the need to choose which nucleic acid to recover from a particular FFPE section.

First, FFPE slide-mounted or tissue blocks sections are deparaffinized and digested with protease. The samples are then loaded on a filter cartridge; DNA remains bound to the filter cartridge while RNA flows through. To recover RNA, the flow-through is applied to a new filter cartridge, treated with DNase, then eluted from the filter. To recover DNA, the DNA bound to the filter cartridge is treated with RNase A, then eluted from the filter.

ChargeSwitch® gDNA 3-10 ml Blood Kit facilitates purification of up to 300 µg of gDNA from 3 – 10 ml of blood sample. This is done using magnetic bead-based technology that follows a 3-step protocol: DNA Capture, DNA Purification, and Elution (Figure 1). First, cells are lysed, and crude DNA is captured on magnetic beads, leaving most of the cell debris and protein behind in solution. Any residual protein is digested using the Protease and then washed away to leave pure intact DNA. Finally, the pure DNA is then eluted into a small volume, ready for use in any downstream applications (Figure 2). We recommend using the 50 ml Tube Magnetic Separator to obtain the best results.

PrepMan™ Ultra Sample Preparation Reagent (Applied Biosystems™)

The PrepMan® Ultra Sample Preparation Reagent was developed for the rapid preparation of DNA template from Gram-negative food-borne pathogens for use in PCR amplification reactions. These samples often have high lipid content that can inhibit PCR amplification of the template. Using a simple boil and spin protocol, PrepMan®Ultra Reagent efficiently inactivates PCR inhibitors, significantly reducing the need to repeat the template preparation step.

PrepMan® Ultra reagent has been used to prepare PCR-ready template from a wide variety of different sources, including bacteria, yeast, filamentous fungi, both from a plate or from liquid culture, mammalian tissue smears, human cells (buccal swab), and mammalian whole blood.
