This Elution solution is a component of the MagMAX Saliva gDNA Isolation Kit. It is offered here as a stand-alone reagent for applications that require more Elution solution than is provided in the kit.

Mouse RiboPure™-Blood RNA Isolation Kit with Manual (Invitrogen™)

For the purification of extremely high-quality total RNA (and optionally, microRNA) from mouse or rat blood in about 35 minutes. The Ambion® kit includes sufficient reagents for 25 RNA purifications from whole blood samples ranging from 0.1–0.5 each.

• Includes RNAlater®, predispensed in 2 mL tubes, for RNA stabilization
• Resulting RNA is suitable for stringent downstream applications such as array analysis and qRT-PCR

Due to interference from the enzymatic inhibitors and anticoagulants present in blood samples, RNA purification from whole blood requires a stringent isolation method that yields high-quality RNA depleted of inhibiting agents and contaminants. The kit includes microfuge tubes preloaded with RNAlater® Tissue Collection:RNA Stabilization Solution, an aqueous, nontoxic storage reagent that prevents RNA degradation and preserves RNA profiles within samples. Stabilized samples can be stored at room temperature for up to 3 days or at –20°C for long-term storage. The related Figures show the performance of RNA isolated with the Mouse RiboPure™-Blood Kit in real-time RT-PCR. The kit includes an alternative protocol for isolating total RNA that contains microRNA. Typically, 30–55 µg of total RNA (containing microRNA) is obtained from 0.5 mL mouse blood. Yields from rat blood are considerably more variable, generally 18–40 µg from 0.5 mL blood.

TaqMan™ miRNA ABC Purification Kit - Human Panel B (Applied Biosystems™)

Use the TaqMan® miRNA ABC Purification Kit to purify high-quality miRNA from blood-related or other difficult sample types without hazardous chemicals.
• Simple and fast workflow using oligonucleotide-conjugated Dynal® magnetic beads
• Small sample input: typical volumes range from 10 µL for blood samples to 50 µL for serum and plasma samples
• RT-PCR inhibitors commonly found in blood-related samples are easily washed away

Simple and Fast miRNA Purification
The TaqMan® miRNA ABC (anti-miRNA bead capture) purification kit contains buffers and magnetic beads for single-tube isolation of specific microRNA (miRNA) from small inputs of all human sample types, including blood, serum, plasma, FFPE samples, solid tissues, cultured cells, and saliva in 75 minutes (see workflow, Figure 1).

Human Panel A and Human Panel B
The TaqMan® miRNA ABC Purification Kit is offered in two configurations: Human Panel A and Human Panel B. The beads in each kit are superparamagnetic Dynabeads® covalently bound to a unique set of 377 anti-miRNA oligonucleotides for each panel that correspond to the assays contained in the TaqMan® Array Human MicroRNA A and B cards. The miRNA isolation process relies on hybridization of miRNAs to the corresponding anti-miRNA oligonucleotides attached to the beads. Complete lists of miRNAs isolated by the panels can be found in the TaqMan® miRNA ABC Purification Kit protocol (below).

miRNA Analysis
The miRNAs isolated using the TaqMan® miRNA ABC Purification Kit are ready for conversion into cDNA and downstream analysis by quantitative PCR using either single tube (individual) TaqMan® microRNA assays or the TaqMan® Array Human microRNA A and B cards. Figure 2 gives examples of the data that can be obtained from miRNA isolated from various sample types using the TaqMan® miRNA ABC Purification Kit.

PureLink™ RNA Mini Kit (Invitrogen™)

The PureLink RNA Mini Kit is a column-based kit used to isolate high-quality total RNA from a wide variety of sample types in 20 minutes using standard laboratory equipment. The kit includes RNase-free lysis and wash solutions that protect RNA from RNases while liberating the RNA from DNA, proteins, and other cellular debris. The fast spin-column workflow is ideal for processing low to mid-throughput batch sizes. The advanced PureLink RNA Mini spin column design allows for maximum sample input (200 mg of tissue) and RNA recovery (up to 1000 µg). This means that users can process both small and large sample sizes with the same RNA isolation kit.

• Process sample inputs from 5–200 mg of animal/plant tissue or from 0.5 x 106 to 108 tissue culture cells with one spin column in 20 minutes
• Isolate more RNA than competitor mini spin columns: up to 1000 µg of total RNA from one column

Extra-large binding capacity enables rapid RNA purification using standard laboratory equipment
The PureLink RNA Mini Kit provides rapid purification of total RNA from a wide range of cells and tissue types to yield up to 1000 µg of purified RNA from a single extraction (see Figure 1). High-quality total RNA can be obtained from mini- to midi-prep amounts of starting material with just trace amounts of residual genomic DNA contamination. The extra-large binding capacity enables one kit to handle most RNA isolation needs in a quick 20 minute protocol without the need for special sample processing instruments.

Easy, optional, on-column DNase treatment for sensitive applications
The PureLink RNA Mini Kit columns are highly efficient for isolating high quality total RNA while removing the majority of genomic DNA. In general, most applications requiring RNA from animal tissue or mammalian cell lines do not require additional DNase treatment. However, some applications such as gene expression analysis by qRT-PCR without intron-spanning primers or working with samples from organisms with very small or no introns may require more complete removal of residual contaminating DNA. The PureLink DNase Set allows for convenient on-column digestion of DNA during the RNA isolation protocol. Treating with DNase while “on-column” is easier and allows higher RNA recovery than treating with DNase after the RNA has been isolated. The PureLink DNase Set can also be used to remove residual DNA from RNA that has been previously purified. Both the “on-column” and post-RNA purification workflows are options available with the PureLink DNase Set.

Simplified, non-toxic RNA purification
The PureLink RNA Mini Kit utilizes non-toxic guanidine-isothiocyanate lysis buffer to protect the RNA during the isolation steps. Special RNase-free reagents combined with certified RNase-free silica membranes in a unique column configuration allow for a safe and easy procedure that can typically be completed in less than 20 minutes without the need of hazardous phenol/chloroform extraction, CsCl centrifugation, or LiCl or alcohol precipitation.

The PureLink RNA Mini Kit is recommended for use with the Homogenizer (Cat. No. 12183026), designed to homogenize cell or tissue lysates via centrifugation, prior to nucleic acid purification. The Homogenizer is especially effective for clarifying particulates from plant tissues.

For plant tissues rich in polyphenolics or starch (e.g., pine needles, potato tubers), we recommend using PureLink Plant RNA Reagent (Cat. No. 12322012) together with the PureLink RNA Mini Kit.

Cells-to-CT™ Bulk Fast Advanced RT Reagents (Invitrogen™)

Invitrogen Fast Advanced Cells-to-CT Bulk RT Reagents are separately-available, large-quantity versions of the reagents used for reverse transcription (RT) in the TaqMan Fast Advanced Cells-to-CT Kit and the SYBR Green Fast Advanced Cells-to-CT kit (20X RT enzyme mix and 20X RT buffer). These reagents are sufficient for 2500 RT reactions. When combined with the Cells-to- CT Bulk Lysis Reagents, these reagents can be easily incorporated into automated, high-throughput applications.

BigDye XTerminator™ Purification Kit (Applied Biosystems™)

The BigDye XTerminator® Purification Kit is a fast, simple purification method for DNA sequencing reactions that removes unincorporated BigDye® terminators and salts. No more dye blobs! Cleanup is complete in under 40 minutes and requires less than 10 minutes of labor.

Simple Purification Process
Traditional purification methods, such as ethanol precipitation, require the addition of multiple reagents along with decanting and centrifuging steps. The BigDye XTerminator® Purification Kit requires the addition of only two reagents which can be added sequentially or premixed:

• XTerminator™ Solution – Scavenges unincorporated dye terminators and free salts from the post-sequencing reaction.
• SAM™ Solution – Enhances the performance of the XTerminator™ Solution and stabilizes the post-purification reactions.

The protocol is very simple (see figure 1):
1. Perform cycle sequencing with BigDye® terminators.
2. After cycle sequencing, centrifuge the reaction plate briefly, and then pipette the correct volume of SAM™ Solution into each plate well.
3. Pipette the correct volume of XTerminator™ Solution into each well.
4. Vortex the XTerminator™ Solution bulk container briefly.
5. Using a wide-bore pipette tip (recommended wide-bore tips), pipette the correct volume of liquid into each plate well
6. Seal the plate using a heat seal or Clear Adhesive Film (P⁄N 4306311). Vortex for 30 minutes (recommended 96-well microplate vortexers and 384-well microplage vortexers), then centrifuge the reaction plate briefly.
7. Place the reaction plate in the Applied Biosystems® DNA analyzer. Select run module and run plate.

Excellent Long Fragment Recovery
You won’t sacrifice sequence length when you use the BigDye XTerminator® Purification Kit. Figure 2 shows data from the DNA Technology Unit, Plant Biotechnology Institute (NRC-PBI) Saskatoon, Canada. The Q20 base read=1044 bp.

Improved Short Fragment Recovery
Use the BigDye XTerminator® Purification Kit to obtain higher quality data. Figure 3 shows a comparison between the BigDye XTerminator® Purification Kit and ethanol precipitation-based purification performed at the DNA Technology Unit, Plant Biotechnology Institute (NRC-PBI) Saskatoon, Canada. The BigDye XTerminator® Purification Kit results in superior short fragment recovery. Ethanol purification produces sequence data marred by dye blobs.

Increased Stability
Sealed reaction plates are stable for 48 hours at room temperature or 10 days at 4°C You can re-run the same sample in case of a power interruption, computer malfunction, etc. without having to repeat the experiment. This increases your flexibility when loading a reaction plate on a CE instrument.

Run Modules Available
Applied Biosystems provides download-able run modules for use with the BigDye XTerminator® Purification Kit and Data Collection Software. These modules are designed for the 3100, 3100–Avant (with Data Collection v2.0 or above), 3130⁄3130xl, and 3730⁄3730xl Analyzers. MagMAX™ Microbiome Bead Tubes (Applied Biosystems™)

These bead tubes are included in the MagMAX Microbiome Ultra Nucleic Acid Isolation Kit (Cat. No. A42358). They are made available separately for applications that require more tubes than are provided in the kit.

MagMAX™ FFPE DNA/RNA Ultra Kit (Applied Biosystems™)

The MagMAX™ FFPE DNA/RNA Ultra Kit is designed for sequential isolation of DNA and RNA from the same formaldehyde- or paraformaldehyde-fixed paraffin-embedded (FFPE) tissue sample. The DNA and RNA are recovered in separate eluates, and both are compatible with a broad range of applications, including real-time PCR and next-generation sequencing. The isolation of RNA and DNA from the same FFPE sample makes this kit an ideal method of sample preparation for Oncomine™ Comprehensive and Oncomine™ Focus assays.

Unlocking the information contained in FFPE samples is essential to cancer research. Common nucleic acid isolation methods require you to choose which nucleic acid to recover from a given sample, with loss of the other nucleic acid (and any information that may have resulted from its analysis). Alternatively, the DNA and RNA are co-purified, or the sample is split in half so that each can be purified.

More information from less sample input
The MagMAX FFPE DNA/RNA Ultra Kit protocol addresses these issues by first isolating DNA from the FFPE sample and then recovering the RNA from the DNA-depleted supernatant. The nucleic acids are prepared as separate eluates and are then ready for downstream analysis. This allows for a fuller analysis of precious samples, including investigation of important biomarkers, such as hotspot mutations, CNVs, gene fusions, and indels.

Benefits of MagMAX magnetic bead–based purification
The magnetic bead–based purification format of the kit allows for easy scaling of the number of samples processed either manually with a magnetic stand or with a magnetic particle processor such as the KingFisher™ Duo Prime or KingFisher™ Flex processor.

Magnetic beads offer many benefits compared to other technologies for isolating nucleic acids. Beads bind the RNA and DNA more efficiently than glass fiber filters, resulting in higher and more consistent yields. Additionally, because filters and vacuum manifolds are not used, there is no risk of undigested tissue particulates clogging these items during the extraction process. This clogging issue is of particular concern with fibrous FFPE tissue samples that that are not fully digested during the proteinase K treatment.

Optimized bead surface chemistry
The Dynabeads MyOne Silane utilized in the MagMAX FFPE DNA/RNA Ultra Kit have an optimized silica-like surface chemistry with high specific surface area that allows efficient kinetics and high sensitivity in nucleic acid capture. These aspects make Dynabeads MyOne Silane ideal for the recovery of both RNA and DNA targets found in FFPE samples. Once captured, both of these nucleic acids are then eluted separately in volumes of 50 µL and are ready for downstream applications.

TRIzol™ LS Reagent and Phasemaker™ Tubes Complete System (Invitrogen™)

This system includes both TRIzol™ LS Reagent and Invitrogen™ Phasemaker™ Tubes.

TRIzol LS Reagent is a complete, ready-to-use reagent for the isolation of high-quality total RNA from a variety of liquid biological samples such as serum and virus preparations.

Phasemaker Tubes contain a thick liquid polymer that separates and completely isolates the upper aqueous phase of the TRIzol Reagent mix from the organic phase underneath, greatly simplifying removal of the upper phase containing the RNA. This TRIzol LS Reagent and Phasemaker Tubes Complete System is for the extraction of RNA, not DNA or protein. It will accommodate approximately 0.25 mL of sample.

MicroSEQ™ ID Sequencing Clean-up Cartridges (Applied Biosystems™)

The MicroSEQ® ID Sequencing Clean-up Cartridges are used for post-sequencing clean-up in the MicroSEQ® ID workflow. The gel filtration matrix used in the cartridges provides a convenient and simple way to remove dye terminators, dNTPs, and salts, as well as residual primers from cycle sequencing reactions.

The MicroSEQ® ID Sequencing Clean-up Cartridges can be used downstream of the following sequencing kits:

The optimized reagents included in the MagMAX Viral/Pathogen kit allow you to maximize the amount of sample input, thereby increasing the amount of RNA and/or DNA recovered. Up to 400 µL of sample can be processed per well in a KingFisher 96-deep well plate. For larger sample volumes, protocols are available to process up to 2 mL of sample per well in a KingFisher 24-deep well plate.

The Chargeswitch® -Pro Filter Plasmid Mini Kit includes 10 plasmid minipreps and provides high yield and high purity plasmid with the fastest and the simplest protocol.
The ChargeSwitch®-Pro Plasmid Filter Mini, Midi, Maxi Kits generates high-quality plasmid DNA without the use of harsh or potentially interfering reagents, for better success in downstream applications. The system is based upon a novel dual⁄nested column design which facilitate the fastest and simplest protocol for high quality plasmid. The system is composed of two columns. How it works
The ChargeSwitch® technology replaces the use of ethanol, chaotropic salts, or organic solvents for nucleic acid purification with a simple ion-exchange mechanism. At low pH values (<6.5) the ChargeSwitch® surface ligand is positively charged and binds plasmid DNA. Raising the pH (>8.5) neutralizes the charge and elutes the bound nucleic acid. The ChargeSwitch® protocol retains the ease and familiarity of the common silica spin column protocol with a binding, washing, and elution step, making adapting to the ChargeSwitch® method effortless.

MagMAX™ Pathogen RNA/DNA Kit (Applied Biosystems™)

The Applied Biosystems® MagMAX™ Pathogen RNA/DNA Kit enables purification of RNA and DNA from viruses and easy-to-lyse bacteria and parasites, using magnetic particle technology in a 96-well format. PCR inhibitors are effectively removed in the process, making the resulting nucleic acid ideal for real-time PCR and RT-PCR applications.

The MagMAX™ Pathogen RNA/DNA Kit is Easy to Automate
Purify RNA/DNA from 96 samples in approximately 45 minutes (Fig. 2) using the MagMAX™ Express-96 Deep Well Magnetic Particle Processor. Low and high sample input volumes can be used for most sample types, and multiple sample types can be processed at the same time on the same plate after sample-specific pre-processing, saving time and plastics.

Reduce false-negative results due to the effective PCR inhibitor removal achieved by the MagMAX™ Pathogen RNA/DNA Kit. Magnetic particles bind RNA/DNA more efficiently and are more thoroughly washed than glass fiber filters, resulting in higher yields and purity of RNA/DNA.
