PureLink™ Pro Quick96 Plasmid Purification Kit (Invitrogen™)

The PureLink® Pro Quick96 Plasmid Kit combines advanced silica plate extraction chemistry with an optimized 96-well plate design for manual or automated processing of plasmid DNA from E. coli in as little as 45 minutes (Table 1). Using the PureLink® Pro Quick96 Plasmid Kit, you will:

How it works
The PureLink® Pro Quick96 Plasmid Kit uses two 96-well plates, a filtration plate (clear O-ring), and a binding plate (red O-ring) to purify plasmid DNA. Bacterial cells are subjected to alkaline lysis and are pelleted by centrifugation. The bacterial lysate is then applied to the Quick96 Filtration Plate for clearing and collection in the Quick96 Binding Plate, where the plasmid DNA binds to the silica membrane. After washing steps to remove contaminants, purified DNA is eluted and ready for use in downstream applications. The entire procedure can be completed in 45 minutes (Figure 1).

Improved plate design
The PureLink® Pro Quick96 Plasmid Kit provides optimized semiskirted 96-well plates for improved performance and results (Figure 2). The Quick96 Filtration Plate is designed for fast and efficient clearing of the bacterial lysates. The Quick96 Binding Plate offers an increased binding capacity of up to 20 µg plasmid DNA. In addition, the plate nozzles with annular collar prevent cross-contamination of samples and improve drying of the silica membrane to prevent ethanol carryover. In addition, sample processing can be performed manually using a benchtop vacuum manifold, by centrifugation, or with an automated liquid handling platform. The semiskirted plate design is compatible with most vacuum manifolds on robotic workstations.

Higher yields and purity
The PureLink® Pro Quick96 Plasmid Kit allows rapid and reproducible purification of up to 20 µg of plasmid DNA from E. coli strains grown in up to 5 ml LB, 3 ml 2xYT, or 3 ml TB. In an automated platform using 1.5 ml E. coli culture grown overnight in a square-well block, typical yields are 5-15 µg plasmid DNA, with an average of 9.4 µg (Figure 3). The resulting plasmid DNA is supercoiled with no detectable genomic DNA or RNA (Figure 4). The PureLink® Pro Quick96 Plasmid Kit provides higher yields of high-quality plasmid DNA for use in common downstream applications such as automated sequencing, PCR, restriction enzyme digestion, and cloning.

Buccal swabs represent a simple, noninvasive, and widely used method for collecting DNA. Isolating DNA from swabs is now easier than ever, thanks to the combined power of the the iPrep™ ChargeSwitch® Buccal Cell Kit and the iPrep™ Purification Instrument. This combination provides for fast lysis before automated processing-purified DNA can be obtained from cell samples in as little as 40 minutes, including a 20 minute lysis step. Additionally, youll experience less inhibition of downstream processes such as STR (Figure 1) and multiplex analysis, with higher yields and purity (Table 1).

One Kit for a Diverse Set of Sample Types
Why purchase multiple purification kits to isolate gDNA from different sample types? The MagMAX™ DNA Multi-Sample Ultra Kit eliminates the headache of ordering these extra kits because it has been optimized for use with a variety of biological samples. Benefits of MagMAX™ Magnetic Bead–based Purification
Magnetic beads offer many benefits compared to other technologies for isolating gDNA. Beads bind gDNA more efficiently than glass fiber filters, resulting in higher and more consistent gDNA yields. Additionally, because filters are not used, there is no risk of filter clogging due to cellular particulates in samples.

Optimized components and protocols
The PureLink® Genomic DNA Mini Kit includes parts and protocols that improve upon the easy and familiar spin-column methods currently in use (see figure). Columns are newly configured to bind and release DNA more efficiently (see figure). Each sample source uses a specialized lysis procedure and an optimized lysis buffer formulated to enhance proteinase K activity and eliminate protein contamination. A chaotropic salt buffer promotes stable binding of DNA to the column resin, and powerful wash buffers remove all traces of protein and salt. DNA is eluted in a low-salt buffer to allow for pH stabilization of the DNA in storage.

Higher yields and purity
With PureLink® Genomic DNA Kits you can expect high yields of high-purity gDNA (determined by A260/A280 measurements), no matter the sample type, from bacteria to tissues to blood to cultured cells (see figures). Benefits of MagMAX magnetic bead–based purification
The magnetic bead–based purification format of the kit allows for easy scaling of the number of samples processed either manually with a magnetic stand or with a magnetic particle processor such as the KingFisher Duo Prime or KingFisher Flex processors.

Magnetic beads offer many benefits compared to other technologies for isolating nucleic acids. Beads bind the RNA and DNA more efficiently than glass fiber filters, resulting in higher and more consistent yields. Additionally, because filters and vacuum manifolds are not used, there is no risk of undigested tissue particulates clogging these items during the extraction process. S.N.A.P.™ UV-Free Gel Purification Kit (Invitrogen™)

The S.N.A.P.™ UV-Free Gel Purification Kit uses non-toxic reagents to rapidly purify intact DNA fragments from agarose gels. The key to isolating undamaged DNA is the unique crystal violet reagent, which allows visualization of DNA (>=200 ng) in the gel under ambient light. This eliminates the need for ethidium bromide and exposure to UV light, which can nick and damage DNA. Nucleic acids up to 7 kb are efficiently purified from the gel using the convenient S.N.A.P.™ resin, provided ready to use in a spin column format. The resin eliminates organic extractions, which generate hazardous waste. In addition, there are no time-consuming ethanol precipitations.

Easy to Use
The S.N.A.P.™ UV-Free Gel Purification Kit yields pure, intact DNA in an easy-to-perform, rapid procedure. Simply:
1. Prepare gel with crystal violet reagent and load samples in crystal violet loading buffer.
2. Visualize DNA band under ambient light and excise from agarose gel.
3. Add NaI solution (provided) and melt the agarose at 42-50°C.
4. Load sample onto the S.N.A.P.™ spin column.
5. Wash away contaminants.
6. Elute DNA in water.

iPrep™ PureLink™ gDNA Blood Kit (Invitrogen™)

The iPrep™ PureLink® gDNA Blood Kit allows rapid and automated extraction of genomic DNA (gDNA) from human blood including fresh or frozen blood samples, and concentrated leukocyte preparations (buffy coat). Genomic DNA is extracted from blood samples using the Dynabeads® MyOne™ SILANE and iPrep™ Purification Instrument within 30 minutes without the use of centrifugation. The purified genomic DNA is suitable for use in downstream applications, including PCR. Advantages of the iPrep™ PureLink® gDNA Blood Kit:

GeneJET PCR Purification Kit (Thermo Scientific™)

Thermo Scientific GeneJET PCR Purification Kit utilizes a proprietary silica-based membrane technology in the form of a convenient spin column, eliminating the need for tedious resin manipulations or toxic phenol-chloroform extractions. It effectively removes primers, dNTPs, unincorporated labeled nucleotides, enzymes, and salts from PCR and other reaction mixtures.

The kit can be used for purification of DNA fragments from 25 bp to 20 kb with recovery rates up to 100%. Each GeneJET purification column has a total binding capacity of up to 25 µg of DNA, and the entire procedure takes 5 minutes. The purified DNA can be used in common downstream applications, such as sequencing, restriction digestion, labeling, ligation, cloning, in vitro transcription, blotting or in situ hybridization.

mirVana™ miRNA Isolation Kit, without phenol (Invitrogen™)

The mirVana™ miRNA Isolation Kit uses a rapid procedure to isolate small RNAs from tissue and cells using an efficient glass fiber filter (GFF)-based method. The method isolates total RNA ranging in size from kilobases down to 10-mers. Each kit contains sufficient reagents and consumables for either 40 isolations of total RNA (including small RNAs), or 20 separate large and small RNA fractions. Phenol must be purchased separately. Features of this kit:

Efficient recovery of small RNA
Current RNA purification procedures rely on organic extraction, followed by alcohol precipitation or adsorption of nucleic acid molecules on a GFF or silicate matrix. Unfortunately, the former procedure requires alcohol precipitation, which is time consuming and inefficiently recovers small RNAs. The latter procedure typically results in the loss of substantial amounts of small RNA. The mirVana™ miRNA Isolation Kit uses organic extraction followed by purification on a GFF under specialized binding and wash conditions. As a result, the kit effectively recovers all RNA—from large mRNA and ribosomal RNA down to 10-mers—from virtually all cell and tissue types. The mirVana™ miRNA Isolation Kit enables the isolation of small RNA-containing total RNA from samples consisting of 102–107 cultured cells or 0.5–250 mg tissue.

Enrich for small RNA
Although the mirVana™ miRNA Isolation Kit efficiently purifies all RNA larger than 10 nt, it also includes procedures for isolating RNA fractions, specifically enriched or depleted in small RNA species. Through the enrichment procedure included in the mirVana™ miRNA Isolation Kit, RNA molecules of ~200 nt and less can be efficiently purified away from the larger RNA species. The resulting RNA preparation is highly enriched for miRNAs, siRNAs, and/or snRNAs. Small RNA enrichment using the mirVana™ miRNA Isolation Kit procedure allows more sensitive small RNA detection with less background as compared to the same assay used with total RNA.

MagMAX™ DNA Multi-Sample Kits are designed for rapid purification of high-quality genomic DNA from a variety of sample types including blood and blood cards, tissue, mouse tails, buccal swabs, buffy coats, and cultured cells. These purification kits use MagMAX™ magnetic bead-based nucleic acid isolation technology to produce high yields of purified DNA, free from inhibitors that may affect downstream reactions such as genotyping assays, CE and next-generation sequencing, and high resolution melting analysis. The 50 prep kit is designed for manual microfuge tube processing.

The demands for DNA yield and purity have become more stringent with the development of new analytical methods and technologies, such as high-throughput genotyping, and next-generation DNA sequencing platforms. MagMAX™ DNA Multi-Sample Kits meet the challenge by delivering highly pure DNA that is free of nucleases, proteins, and other inhibitors of downstream enzymatic reactions.
As shown in Table 1, DNA with high yield and purity can be obtained from a variety of materials using the MagMAX™ DNA Multi-Sample Kits. This is in contrast to filter membrane–based methods that often require different kit configurations for various input materials. For applications requiring maximal yields, for small batch sizes, or for DNA archival purposes MagMAX™ DNA Multi-Sample Kit (tube format, manual processing) may be desirable because larger sample inputs are accommodated. DNA yields suitable for most routine applications may be obtained using the MagMAX™-96 DNA Multi-Sample Kits (96-well plate format, compatible with automated handling) (Table 1).

The MagMAX™ DNA Multi-Sample Kit also delivers high yields of DNA from lipid or glycogen rich samples, such as brain and liver tissues, respectively. Figure 1 shows more than twice as much DNA was purified from tissues with the MagMAX™ DNA Multi-Sample Kit than with the leading competitor’s filter membrane–based method.

Most enzymatic molecular applications are prone to inhibition by analytes carried over from the sample matrix (e.g., heme, heparin, salts, protein) or the purification method itself (phenol, salts, alcohol). Figure 2 illustrates the effect of DNA purity on sensitive molecular applications, such as quantitative PCR (qPCR). DNA was extracted from blood using the MagMAX™-96 DNA Multi-Sample Kit, or a competitor’s filter membrane–based kit and subjected to real-time PCR analysis. The cycle threshold CT values obtained with the DNA purified using the MagMAX™ DNA Multi-Sample Kit were lower than those obtained with the filter membrane kit–purified DNA, indicating the MagMAX™ kit yields higher-purity DNA.

MagMAX™ DNA Multi-Sample Kits provide purified DNA that is suitable for a variety of Applied Biosystems application workflows (Figure 3), including SNP genotyping and DNA sequencing analysis (see Figure 4). The Cells-to-Ct method features a unique lysis reagent that lyses cells and protects RNA. DNA removal occurs simultaneously in the 5-minute lysis step to save time and improve ease of use. Lysis is then stopped by the addition of a “Stop Solution” that irreversibly binds to the lysis reagents.

The SYBR Green Fast Advanced Cells-to-CT Kit contains Cells-to-Ct Fast Advanced reverse transcription (RT) reagents for cDNA synthesis and PowerUp SYBR Green Master Mix for real-time PCR analysis. Primer sets are provided by the user.

Simple 7-minute sample preparation
The SYBR Green Fast Advanced Cells-to-CT Kit is designed for 10–100,000 cultured cells/sample. Cells are washed in PBS and lysed in lysis solution for five minutes at room temperature; DNase treatment can be performed simultaneously. Lysis is terminated at room temperature by a 2-minute incubation with Stop Solution. The lysates are now ready for reverse transcription or storage at –20°C for a short term or -80C for a long term.

Maximize sensitivity of detection
Unlike some competitor kits that limit the amount of lysate in the RT reaction to 10%, the SYBR Green Fast Advanced Cells-to-CT Kit can accommodate 45% of the total RT reaction volume as cell lysate. Additionally, cDNA can comprise up to 30% of the real-time PCR reaction volume. The large lysate volume in the optimized RT reaction, along with the large cDNA volume in the subsequent real-time PCR using the PowerUP SYBR Green Master Mix, lead to maximum sensitivity. The master mix amplifies the target precisely and accurately, enabling the detection of small quantities of target, such as transcripts expressed at low levels.

Reduced sample loss and transfer error
Because samples can be processed directly in culture wells (96 or 384 wells), sample handling and the potential for sample loss or transfer error are minimized, facilitating rapid, high-throughput processing. Unlike old-fashioned, multi-step RNA isolation protocols, no heating, washing, or centrifugation steps are required. The kit greatly simplifies a laborious 30–60 minute process and reduces it to 7 minutes.

A simple and reliable technology for everyday RNA purification
PureLink® technology combines guanidine-isothiocyanate lysis with the speed, purity, and ease-of-use of silica-membrane purification. The safe and easy procedure can be completed in typically less than 15 minutes with >85% recovery from the first elution. The kit can be used for clean-up of purified RNA from enzymatic reactions, desalting of RNA samples after phenol extraction or ethanol precipitation, and concentration of RNA. A simple and rapid procedure
Homogenize your sample in lysis buffer, add 70% ethanol, and mix. Deposit the resulting solution into a PureLink® Micro kit column with collection tube, and centrifuge for one minute. Nucleic acids bind to the PureLink® silica membrane in the column, and after subsequent washes and on-column digestion with DNase, purified RNA can be eluted with RNase-free water.
