PureLink™ HiPure Precipitator Module (Invitrogen™)

The PureLink™ HiPure Precipitator allows fast, simple, and efficient desalting and concentration of plasmid DNA isolated using anion-exchange chromatography. The traditional method for DNA precipitation of anion exchange isolated plasmid DNA involves isopropanol precipitation followed by centrifugation and drying of DNA which is time consuming and labor intensive. The use of the PureLink™ HiPure Precipitator allows plasmid DNA concentration within 5 minutes, eliminates the need for centrifugation, and reduces the risk of losing the DNA pellet during supernatant removal. The recovery is >80% and resulting plasmid DNA is ready to use in the most challenging applications such as transfection.

PureLink™ Quick Gel Extraction Kit (Invitrogen™)

The PureLink® Quick Gel Extraction Kit allows you to rapidly and efficiently purify DNA fragments from TAE or TBE agarose gels of various percentages. Unique method eliminates the need to isolate RNA from cultured cells
The Cells-to-Ct method features a unique lysis reagent that lyses cells and protects RNA. DNA removal occurs simultaneously in the 5-minute lysis step to save time and improve ease of use. Lysis is then stopped by the addition of a “Stop Solution” that irreversibly binds to the lysis reagents.

The TaqMan Fast Advanced Cells-to-CT Kit contains reverse transcription (RT) reagents for cDNA synthesis and TaqMan Fast Advanced Master Mix for real-time PCR analysis. TaqMan primer/probe sets are sold separately.

Simple 7-minute sample preparation
The TaqMan Fast Advanced Cells-to-CT Kit is designed for 10–100,000 cultured cells/sample. Cells are washed in PBS and lysed in solution for five minutes at room temperature; DNase treatment can be performed simultaneously. Lysis is terminated at room temperature by a 2-minute incubation with Stop Solution. The lysates are now ready for reverse transcription or storage at –20°C for a short term or -80°C for a long term.

Maximize sensitivity of detection
Unlike some competitor kits that limit the amount of lysate in the RT reaction to 10%, the TaqMan Fast Advanced Cells-to-CT Kit can accommodate 45% of the total RT reaction volume as cell lysate. Additionally, cDNA can comprise up to 45% of the real-time PCR reaction volume. The large lysate volume in the optimized RT reaction, along with the large cDNA volume in the subsequent real-time PCR using the TaqMan Fast Advanced Master Mix, lead to maximum sensitivity. The master mix amplifies the target precisely and accurately, enabling the detection of small quantities of target, such as transcripts expressed at low levels.

Because samples can be processed directly in culture wells (96 or 384 wells), sample handling and the potential for sample loss or transfer error are minimized, facilitating rapid, high-throughput processing. PureLink™ HiPure Plasmid FP (Filter and Precipitator) Maxiprep Kit (Invitrogen™)

The PureLink™ HiPure Plasmid FP (Filter and Precipitator) Maxiprep Kit isolates plasmid DNA, from cell pellet to ultrapure DNA, in less than one hour. Using the HiPure Filter Columns and HiPure Precipitators supplied in the kit, you can purify plasmid DNA without the centrifugation steps required in traditional isolation protocols. The quality of the purified plasmid DNA is equivalent to that obtained by two passes through cesium chloride gradients - the most rigorous method for DNA purification - making it suitable for transfection and other applications requiring high-purity plasmid DNA. The PureLink™ HiPure Plasmid FP Maxiprep Kit (Table 1) provides:

How it works
The PureLink™ HiPure Plasmid FP Maxiprep Kit employs an alkaline lysis method to prepare and rapidly clear bacterial lysates without the need for centrifugation (Figure 1). The unique HiPure Filter Column includes a lysate filtration cartridge integrated into the anion-exchange column for single-step lysate clarification and plasmid DNA binding. Impurities such as RNA, proteins, and other contaminants are removed with a single wash, and ultrapure plasmid DNA is eluted with a high-salt buffer. To precipitate and desalt the DNA, isopropanol is added to the eluted DNA and the mixture is then applied to the HiPure Precipitator using a large syringe. After a subsequent washing and drying step, the plasmid DNA is easily eluted from the HiPure Precipitator with either TE buffer or water. The use of the precipitator eliminates the need for centrifugation, reducing the risk of losing the DNA pellet during supernatant removal, and saving time. The resulting DNA is ready to use in the most challenging applications. The entire protocol - from E. coli cell pellet to ultrapure transfection-grade plasmid DNA - can be completed in about an hour.

Highest yields and ready-to-use concentrations
The HiPure Precipitator has been optimized to allow for lower elution volumes (down to 500 µl) to produce highly concentrated DNA without a significant decrease in yields. In fact, yields at higher concentrations are consistently higher than the competitor kits (Figure 2). Yields and concentration can be adjusted to meet the needs of your downstream application.

RiboMinus™ Human/Mouse Transcriptome Isolation Kit (Invitrogen™)

The RiboMinus™ Human/Mouse Transcriptome Isolation Kit constitutes a novel purification system that enriches the whole spectrum of mRNA species by depleting large ribosomal RNA molecules in a total RNA sample. The kit removes >95% of 18S and 28S rRNA molecules from human or mouse total RNA (Figure 1) without relying on current polyA+ purification methods. This increases the representation of mRNA species, including non-polyA-tailed, fragmented mRNA or other regulatory RNAs that cannot be purified via polyA-selection methods. Purified RNA is suitable for several downstream applications, including, library construction, RT-PCR, and microarray (Figure 2).

RiboMinus™ Transcriptome Isolation Kit, human/mouse (Invitrogen™)

RiboMinus™ Transcriptome Isolation Kits are novel purification systems that enrich the whole spectrum of RNA transcripts by depleting large ribosomal RNA molecules in a total RNA sample. These kits use a 5µ biotin-labeled, Locked Nucleic Acid (LNA) probe specific for large rRNA to remove 95-98% of rRNA molecules from human, mouse, bacteria, or yeast total RNA (Figure 1). This removal increases the representation of RNA transcript species, including non-poly(A) tailed, pre-processed RNA, small tRNAs, siRNA, snRNA, microRNA, fragmented mRNA, and other regulatory RNAs that cannot be purified via poly(A) selection methods.

The RiboMinus™ Transcriptome Isolation Kits enable:

Thermo Scientific GeneJET Plant RNA Purification Mini Kit is designed for rapid and efficient purification of high quality total RNA from wide variety of plant species and tissue types.

The kit utilizes silica-based membrane technology in the form of a convenient spin column, eliminating the need for expensive resins, toxic phenol-chloroform extractions, or time-consuming alcohol precipitation. The standard procedure takes less than 20 minutes following cell lysis. Purified high quality RNA can be used in a wide range of downstream applications, such as RT-PCR, RT-qPCR, Northern blotting and other RNA-based analysis. Process overview
Bacterial cells are harvested and resuspended in ResuspensionBuffer with RNase. The cells are lysed using an alkaline/SDScell lysis procedure. The lysate is then neutralized andconditions are adjusted for subsequent binding. After lysateclarification using the PureLink HQ 96 Clarification Plate, thelysate is processed through the PureLink HQ 96 BindingPlate. The DNA binds to the silica-based membrane in theplate and impurities are removed by washing with WashBuffer. The DNA is then eluted in low salt Elution Buffer orwater.

Dynabeads™ DNA DIRECT™ Universal Kit (Invitrogen™)

The Dynabeads® protocol consist of only a few steps of pipetting and magnetic separation, with all operations performed in a single tube. Contaminating PCR inhibitors are eliminated without any centrifugation steps or use of phenol/chloroform. The kit is particularly useful for isolation of DNA from bacteria and cultured cells, as well as from clinical specimens and tissues from various species.

Magnetic beads offer many benefits compared to other technologies for isolating RNA. Beads bind RNA more efficiently than glass fiber filters, resulting in higher and more consistent RNA yields. Additionally, because filters are not used, there is no risk of filter clogging due to cellular particulates in samples. Since only a small volume of magnetic beads is needed for high-efficiency binding, the bound RNA can be eluted in just 20–50 µL of nuclease-free water, concentrating RNA from large, dilute samples.

Optimized for Isolation of Virus from Biofluids:
The Applied Biosystems® MagMAX™ Viral RNA Isolation Kit has been optimized for use with biological fluids and cell-free samples such as serum, plasma, swabs, and cell culture media. These samples often present unique challenges when purifying RNA, including low viral titers, large volumes to be processed, and sample complexity. Without the need for organic solvents or RNA precipitation, the MagMAX™ Viral RNA isolation kit eliminates the problems often encountered with other RNA isolation methods such filter clogging, large elution volumes, and inconsistent yields.

Get the Same Amount of RNA from 10-fold Less Sample
The sensitivity of viral RNA detection was compared using RNA isolated with Applied Biosystems® MagMAX™ protocol or the current National Veterinary Services Laboratories (NVSL) validated viral testing protocol. Even at one-tenth the sample input, viral RNA isolated using the MagMAX™ kit could be detected at equal or greater levels than RNA isolated using the NVSL protocol and recommended competitor's kit (see data). Accessory Products:
Carrier RNA (SKU#4382878), Lysis/Binding Solution Concentrate (SKU#AM8500), Wash Solution 1 Concentrate (SKU#AM8504), and Wash Solution 2 Concentrate (SKU#AM8640) are components of the MagMAX™ Viral RNA Isolation Kits that may also be purchased separately. A 96-well magnetic ring stand is required to use this kit. JetQuick™ Blood and Cell Culture DNA Maxiprep Kit (Invitrogen™)

The JetQuick™ Blood and Cell Culture DNA Maxiprep Kit is designed for rapid and efficient purification of genomic DNA (gDNA) from large volumes of blood, body fluids (amniotic fluid, saliva, sperm, lymph), and mammalian cells. It utilizes silica membrane-based technology that allows for isolation of 20-50 kb DNA that is suitable for a wide range of downstream applications, including PCR, restriction enzyme digestion, and Southern blotting.

RiboMinus™ Transcriptome Isolation Kits are novel purification systems that enrich the whole spectrum of RNA transcripts by depleting large ribosomal RNA molecules in a total RNA sample. These kits use a 5µ biotin-labeled, Locked Nucleic Acid (LNA) probe specific for large rRNA to remove 95-98% of rRNA molecules from human, mouse, bacteria, or yeast total RNA (Figure 1). This removal increases the representation of RNA transcript species, including non-poly(A) tailed, pre-processed RNA, small tRNAs, siRNA, snRNA, microRNA, fragmented mRNA, and other regulatory RNAs that cannot be purified via poly(A) selection methods. The RiboMinus™ Transcriptome Isolation Kits enable:

• Comprehensive analysis of the whole transcriptome without contamination or interference from rRNA
• More complete collection of transcribed elements of the genome—no reliance on poly(A) purification methods

• Cleaner, clearer microarray analysis due to higher signal-to-background ratios (Figure 2)

Purified RNA is concentrated and suitable for numerous downstream applications, including microarray analysis (Figure 2), library construction, and qRT-PCR.
