GeneJET Plant Genomic DNA Purification Kit (Thermo Scientific™)

Thermo Scientific GeneJET Plant Genomic DNA Purification Mini Kit is designed for rapid and efficient purification of high quality genomic DNA from a wide variety of plant species and tissue types. The standard procedure takes less than 30 minutes following cell lysis and yields purified DNA greater than 30 kb in size. DNA yields vary between different species and tissues depending on genome size, ploidy, cell number, and the age of the tissue sample. The typical yield from a standard tissue source (e. g. young wheat leaves) is 30 to 32 µg from 100 mg of tissue. Isolated DNA can be used directly in PCR, qPCR, southern blotting, and enzymatic reactions.

The mirVana™ miRNA Isolation Kit uses a rapid procedure to isolate small RNAs from tissue and cells using an efficient glass fiber filter (GFF)-based method. The method isolates total RNA ranging in size from kilobases down to 10-mers. Each kit contains sufficient reagents and consumables for either 40 isolations of total RNA (including small RNAs), or 20 separate large and small RNA fractions.

Efficient recovery of small RNA
Current RNA purification procedures rely on organic extraction, followed by alcohol precipitation or adsorption of nucleic acid molecules on a GFF or silicate matrix. Unfortunately, the former procedure requires alcohol precipitation, which is time consuming and inefficiently recovers small RNAs. The latter procedure typically results in the loss of substantial amounts of small RNA. The mirVana™ miRNA Isolation Kit uses organic extraction followed by purification on a GFF under specialized binding and wash conditions. As a result, the kit effectively recovers all RNA—from large mRNA and ribosomal RNA down to 10-mers—from virtually all cell and tissue types. The mirVana™ miRNA Isolation Kit enables the isolation of small RNA-containing total RNA from samples consisting of 102–107 cultured cells or 0.5–250 mg tissue.

Enrich for small RNA
Although the mirVana™ miRNA Isolation Kit efficiently purifies all RNA larger than 10 nt, it also includes procedures for isolating RNA fractions, specifically enriched or depleted in small RNA species. Through the enrichment procedure included in the mirVana™ miRNA Isolation Kit, RNA molecules of ~200 nt and less can be efficiently purified away from the larger RNA species. The resulting RNA preparation is highly enriched for miRNAs, siRNAs, and/or snRNAs. Small RNA enrichment using the mirVana™ miRNA Isolation Kit procedure allows more sensitive small RNA detection with less background as compared to the same assay used with total RNA.

PureLink™ RNA Mini Kit (Invitrogen™)

The PureLink RNA Mini Kit is a column-based kit used to isolate high-quality total RNA from a wide variety of sample types in 20 minutes using standard laboratory equipment. The kit includes RNase-free lysis and wash solutions that protect RNA from RNases while liberating the RNA from DNA, proteins, and other cellular debris. The fast spin-column workflow is ideal for processing low to mid-throughput batch sizes. The advanced PureLink RNA Mini spin column design allows for maximum sample input (200 mg of tissue) and RNA recovery (up to 1000 µg). This means that users can process both small and large sample sizes with the same RNA isolation kit.

• Process sample inputs from 5–200 mg of animal/plant tissue or from 0.5 x 106 to 108 tissue culture cells with one spin column in 20 minutes

Extra-large binding capacity enables rapid RNA purification using standard laboratory equipment
The PureLink RNA Mini Kit provides rapid purification of total RNA from a wide range of cells and tissue types to yield up to 1000 µg of purified RNA from a single extraction (see Figure 1). High-quality total RNA can be obtained from mini- to midi-prep amounts of starting material with just trace amounts of residual genomic DNA contamination. The extra-large binding capacity enables one kit to handle most RNA isolation needs in a quick 20 minute protocol without the need for special sample processing instruments.

Easy, optional, on-column DNase treatment for sensitive applications
The PureLink RNA Mini Kit columns are highly efficient for isolating high quality total RNA while removing the majority of genomic DNA. In general, most applications requiring RNA from animal tissue or mammalian cell lines do not require additional DNase treatment. However, some applications such as gene expression analysis by qRT-PCR without intron-spanning primers or working with samples from organisms with very small or no introns may require more complete removal of residual contaminating DNA. The PureLink DNase Set allows for convenient on-column digestion of DNA during the RNA isolation protocol. Treating with DNase while “on-column” is easier and allows higher RNA recovery than treating with DNase after the RNA has been isolated. The PureLink DNase Set can also be used to remove residual DNA from RNA that has been previously purified. Both the “on-column” and post-RNA purification workflows are options available with the PureLink DNase Set.

Simplified, non-toxic RNA purification
The PureLink RNA Mini Kit utilizes non-toxic guanidine-isothiocyanate lysis buffer to protect the RNA during the isolation steps. Special RNase-free reagents combined with certified RNase-free silica membranes in a unique column configuration allow for a safe and easy procedure that can typically be completed in less than 20 minutes without the need of hazardous phenol/chloroform extraction, CsCl centrifugation, or LiCl or alcohol precipitation.

The PureLink RNA Mini Kit is recommended for use with the Homogenizer (Cat. No. 12183026), designed to homogenize cell or tissue lysates via centrifugation, prior to nucleic acid purification. The Homogenizer is especially effective for clarifying particulates from plant tissues.

RNAqueous™-96 Automated Kit (Invitrogen™)

The Ambion® RNAqueous®-96 Automated Kit is used for automated, phenol-free total RNA isolation from 100-500,000 cells or 0.1–1 mg tissue using a guanidinium-based lysis/denaturant and glass fiber filter separation technology. The kit contains enough reagents for 384 (4 x 96) RNA purifications. The RNAqueous®-96 Automated Kit uses an RNA-binding glass-fiber filter to provide high, reproducible yields of intact RNA. An optional on-the-filter DNase treatment can be performed to ensure removal of genomic DNA for RT-PCR applications. RiboMinus™ Human/Mouse Transcriptome Isolation Kit (Invitrogen™)

The RiboMinus™ Human/Mouse Transcriptome Isolation Kit constitutes a novel purification system that enriches the whole spectrum of mRNA species by depleting large ribosomal RNA molecules in a total RNA sample. The kit removes >95% of 18S and 28S rRNA molecules from human or mouse total RNA (Figure 1) without relying on current polyA+ purification methods. This increases the representation of mRNA species, including non-polyA-tailed, fragmented mRNA or other regulatory RNAs that cannot be purified via polyA-selection methods. Purified RNA is suitable for several downstream applications, including, library construction, RT-PCR, and microarray (Figure 2).

PureLink™ PCR Micro Kit (Invitrogen™)

PureLink® PCR Micro Kits are ideal for purifying and concentrating DNA from low-yielding PCR or restriction digestion reactions. These highly purified DNA fragments and amplicons provide excellent results in downstream applications such as DNA sequencing, restriction digestion, cloning, PCR, and labeling. PureLink® PCR Micro Kits provide a simple and rapid method for purifying and concentrating DNA fragments ranging in size from 100 bp-12 Kb. The unique micro-nozzle design allows up to 95% recovery of DNA fragments with as little as a 5 µl elution volume. With less than a 10% coefficient of variance in the elution volume you can be sure that no ethanol is carried over from the wash steps giving you the best chance of success in downstream applications. Removal of primers, dNTPs, enzymes, and salts exceeds 98.5%. This kit can also efficiently remove unincorporated dye-labeled nucleotides from a cDNA labeling reaction.

Easy-DNA™ gDNA Purification Kit (Invitrogen™)

The Easy-DNA™ Kit is a fast and easy way to isolate high-quality, high-molecular weight genomic DNA (gDNA) from a wide range of cells and tissue types. Sample sizes can range from a single hair follicle up to 1 gram of mammalian tissue. Other samples tested include mouse tails, plant leaves, yeast, and E. coli cells. These samples yield high-quality DNA with an average size between 100 kb and 200 kb, which is suitable for PCR, DNA hybridization, genomic DNA library construction, and other applications. Advantages of the Easy-DNA™ Kit include:

Reliability—high-quality genomic DNA from large or small samples of cells, tissue, plant, yeast, E. coli, blood, and even hair follicles

Simple workflow, quality results
Optimized protocols for a variety of cell and tissue types are included in the instruction manual supplied with the kit. Generally, small-scale gDNA preparations are performed in microfuge tubes, in which samples are mixed with Lysis Solution and incubated at 65°C for 10 minutes. After extraction with chloroform (not included in kit), precipitation, and reconstitution of the DNA pellet, the preparation is complete. The kit contains a protein degradation reagent (for use with high-protein–containing samples such as connective tissue), mussel glycogen for use as a co-precipitant for dilute DNA samples, and an RNase solution.

MicroSEQ™ ID Ultra Sequencing Clean-up 8-Strips Refill Kit (Applied Biosystems™)

The MicroSEQ™ ID Ultra Sequencing Clean-up 8-Strips Refill Kit is used for post-sequencing clean-up in the MicroSEQ™ ID workflow. It includes the 8-well strips that are part of the MicroSEQ™ ID Ultra Sequencing Clean-up 8-Strips Kit (Cat. No. A33246) but does not include the 96-well receiver plate. Dynabeads™ mRNA DIRECT™ Purification Kit (Invitrogen™)

The Dynabeads® mRNA DIRECT™ Kit is designed for simple and rapid isolation of pure, intact polyadenylated (polyA) mRNA directly from the crude lysate of animal and plant cells and tissues. The isolated mRNA is suitable for use in all downstream applications. Advantages of the Dynabeads® mRNA DIRECT™ Kit:

System overview
The isolation protocol relies on base pairing between the polyA residues at the 3' end of most mRNA, and the oligo (dT)25 residues covalently coupled to the surface of the Dynabeads® . Other RNA species lacking a polyA tail will not hybridize to the beads and are readily washed away. Ribosomal RNA, DNA, proteins, and small RNA molecules (such as transfer RNA, micro RNA, and small nucleolar RNA) do not bind to the beads and are discarded. RNase inhibiting agents in the Lysis/Binding Buffer together with stringent hybridization and washing conditions ensure the isolation of pure, intact mRNA from crude samples rich in RNase, without the use of strong chaotropic agents. The mRNA purification beads specifically target and capture the mRNA transcriptome from an extremely wide variety of crude starting samples (see protocol). 1 mg of Dynabeads® Oligo (dT)25 beads (200 µL) binds up to 2 µg of mRNA. A typical mammalian cell contains about 10–30 pg of total RNA, from which 1–5% is mRNA.

MicroSEQ™ ID Purification Combo Kit v2.0, with cartridges (Applied Biosystems™)

The MicroSEQ ID Purification Combo Kit v2.0 includes reagents for post-PCR and post-sequencing clean-up in the MicroSEQ ID microbial identification workflow. This version contains one tube of ExoSAP-IT Express Reagent for post-PCR clean-up and MicroSEQ ID Sequencing Clean-up Cartridges for post-sequencing clean-up.

ExoSAP-IT Express Reagent removes unincorporated dNTPs and primers from the post-amplification PCR mix in 5 minutes compared to 30 minutes with the original ExoSAP-IT Reagent while producing equivalent results. The MicroSEQ ID Sequencing Clean-up Cartridges provide a convenient and simple way to remove dye terminators, dNTPs, salts, and residual primers from cycle sequencing reactions. The end products can be used directly for sequencing on any Applied Biosystems genetic analyzer.

Comparability studies demonstrated equivalent results between the original ExoSAP-IT Reagent and the improved ExoSAP-IT Express Reagent coupled with the MicroSEQ ID Sequencing Clean-Up kit in the MicroSEQ ID workflow (see figure 1). Both ExoSAP-IT and ExoSAP-IT Express enzyme-treated PCR samples produced very similar sequencing results and the correct identification result.

Max Bacterial Enhancement Reagent (Invitrogen™)

The Max Bacterial Enhancement Reagent is designed for use with TRIzol® Reagent to improve the isolation of intact total RNA up to 3-fold from Gram-positive and Gram-negative bacteria. The Max Bacterial Enhancement Reagent is a ready-to-use solution composed of chelating agents, detergent, and a buffer, and is used as an efficient pre-treatment buffer for bacterial cells prior to RNA isolation with TRIzol® Reagent. The use of Max Bacterial Enhancement Reagent with TRIzol® inactivates endogenous RNases and promotes protein denaturation, improving the RNA quality and integrity. The Max Bacterial Enhancement Reagent is available either separately or packaged with TRIzol as part of the TRIzol® Max™ Bacterial RNA Isolation Kit (Cat. No. 16096020 or 16096040).

MagMAX™ Saliva gDNA Isolation Kit (Applied Biosystems™)

Isolate DNA from stabilized and freshly collected saliva
The MagMAX Saliva gDNA Isolation Kit isolates gDNA from 200 µL–2 mL of freshly collected saliva or stabilized saliva collected using a commercially available saliva collection device (see figure below). MagMAX Saliva gDNA isolation chemistry is compatible with the Oragene® saliva collection devices from DNA Genotek, the SalivaGard® saliva collection device from Biomatrica, and the GeneFiX™ Saliva DNA Collection Device from IsoHelix.

The MagMAX Saliva gDNA Isolation Kit maximizes gDNA yields by treating samples with an innovative lysis/binding solution to simultaneously lyse cells, denature proteins, and capture gDNA on high-capacity magnetic binding beads. The Saliva gDNA Wash I Solution removes proteins, detergents, and other impurities from the bound nucleic acid. The gDNA is then released from the beads using a low-TE elution solution.

Two products in one

Simple lysis to column purification protocol

How it works
The PureLink® Pro Quick96 Plasmid Kit uses two 96-well plates, a filtration plate (clear O-ring), and a binding plate (red O-ring) to purify plasmid DNA. Bacterial cells are subjected to alkaline lysis and are pelleted by centrifugation. The bacterial lysate is then applied to the Quick96 Filtration Plate for clearing and collection in the Quick96 Binding Plate, where the plasmid DNA binds to the silica membrane. After washing steps to remove contaminants, purified DNA is eluted and ready for use in downstream applications. The entire procedure can be completed in 45 minutes (Figure 1).

Improved plate design
The PureLink® Pro Quick96 Plasmid Kit provides optimized semiskirted 96-well plates for improved performance and results (Figure 2). The Quick96 Filtration Plate is designed for fast and efficient clearing of the bacterial lysates. The Quick96 Binding Plate offers an increased binding capacity of up to 20 µg plasmid DNA. In addition, the plate nozzles with annular collar prevent cross-contamination of samples and improve drying of the silica membrane to prevent ethanol carryover. In addition, sample processing can be performed manually using a benchtop vacuum manifold, by centrifugation, or with an automated liquid handling platform. The semiskirted plate design is compatible with most vacuum manifolds on robotic workstations.

Higher yields and purity
The PureLink® Pro Quick96 Plasmid Kit allows rapid and reproducible purification of up to 20 µg of plasmid DNA from E. coli strains grown in up to 5 ml LB, 3 ml 2xYT, or 3 ml TB. In an automated platform using 1.5 ml E. coli culture grown overnight in a square-well block, typical yields are 5-15 µg plasmid DNA, with an average of 9.4 µg (Figure 3). The resulting plasmid DNA is supercoiled with no detectable genomic DNA or RNA (Figure 4). RiboMinus™ Transcriptome Isolation Kit, human/mouse (Invitrogen™)

RiboMinus™ Transcriptome Isolation Kits are novel purification systems that enrich the whole spectrum of RNA transcripts by depleting large ribosomal RNA molecules in a total RNA sample. These kits use a 5µ biotin-labeled, Locked Nucleic Acid (LNA) probe specific for large rRNA to remove 95-98% of rRNA molecules from human, mouse, bacteria, or yeast total RNA (Figure 1). How it works
The PureLink™ HiPure Plasmid FP Maxiprep Kit employs an alkaline lysis method to prepare and rapidly clear bacterial lysates without the need for centrifugation (Figure 1). The unique HiPure Filter Column includes a lysate filtration cartridge integrated into the anion-exchange column for single-step lysate clarification and plasmid DNA binding. Impurities such as RNA, proteins, and other contaminants are removed with a single wash, and ultrapure plasmid DNA is eluted with a high-salt buffer. To precipitate and desalt the DNA, isopropanol is added to the eluted DNA and the mixture is then applied to the HiPure Precipitator using a large syringe. After a subsequent washing and drying step, the plasmid DNA is easily eluted from the HiPure Precipitator with either TE buffer or water. The use of the precipitator eliminates the need for centrifugation, reducing the risk of losing the DNA pellet during supernatant removal, and saving time. The resulting DNA is ready to use in the most challenging applications. The entire protocol - from E. coli cell pellet to ultrapure transfection-grade plasmid DNA - can be completed in about an hour.

Highest yields and ready-to-use concentrations
The HiPure Precipitator has been optimized to allow for lower elution volumes (down to 500 µl) to produce highly concentrated DNA without a significant decrease in yields. Simple Workflow Without Sacrificing Sensitivity & Specificity
The process consists of pelleting and washing a 500uL aliquot from the blood tube, followed by a unique digestion⁄DNase step which preserves RNA integrity. Principle

A bacterial culture is harvested and lysed. The lysate is then cleared by centrifugation and applied on the silica column to selectively bind DNA molecules at a high salt concentration. How it works

The ChargeSwitch® technology replaces the use of ethanol, chaotropic salts, or organic solvents for nucleic acid purification with a simple ion-exchange mechanism. At low pH values (<6.5) the ChargeSwitch® surface ligand is positively charged and binds plasmid DNA. Raising the pH (>8.5) neutralizes the charge and elutes the bound nucleic acid. RiboMinus™ Transcriptome Isolation Kit, yeast (Invitrogen™)

RiboMinus™ Transcriptome Isolation Kits are novel purification systems that enrich the whole spectrum of RNA transcripts by depleting large ribosomal RNA molecules in a total RNA sample. These kits use a 5µ biotin-labeled, Locked Nucleic Acid (LNA) probe specific for large rRNA to remove 95-98% of rRNA molecules from human, mouse, bacteria, or yeast total RNA (Figure 1). This removal increases the representation of RNA transcript species, including non-poly(A) tailed, pre-processed RNA, small tRNAs, siRNA, snRNA, microRNA, fragmented mRNA, and other regulatory RNAs that cannot be purified via poly(A) selection methods. The RiboMinus™ Transcriptome Isolation Kits enable:

It has traditionally been difficult to isolate high-quality RNA from yeast because of problems disrupting the cells. Many protocols use enzymatic pretreatments that may affect gene expression patterns. The RiboPure™-Yeast RNA Isolation Kit uses no enzymatic pretreatments and thus does not alter expression profiles. It combines a simple and efficient glass bead/one step phenol–mediated disruption using Ambion's Vortex Bead Beater Adapter (see Accessory Products) followed by a glass fiber filter– based RNA purification step. After DNase treatment with the included DNA-free™ Reagents, the resulting RNA is exceptionally pure containing little to no genomic DNA (less than 0.05%). The kit has been used extensively at Ambion to isolate high-quality RNA from Saccharomyces cerevisiae, Schizosaccharomyces pombe, Pichia pastoris, and Candida albicans (see Figure). The kit provides the highest yield of any method tested from each of these species (see Figure).

Accessory Products:
The Kit requires the use of the Vortex Adapter (SKU #AM10024), which turns a standard laboratory vortex into a bead beater. This adapter fits the Vortex Genie® 2 and holds 12 tubes.

Thermo Scientific GeneJET Gel Extraction and DNA Cleanup Micro Kit is developed as 3 in 1 kit designed for rapid and efficient purification of DNA from PCR, enzymatic reaction mixtures, and DNA extraction from standard or low-melting point agarose gels run in either Tris acetate (TAE) or Tris borate (TBE) buffer. The kit combines the convenience of spin column technology with the selective binding properties of a silica membrane, eliminating the need for tedious resin manipulations or toxic phenol-chloroform extractions.

The GeneJET Gel Extraction and DNA Cleanup Micro Kit effectively removes primers, primer dimers, dNTPs, unincorporated labeled nucleotides, enzymes and salts from PCR and other reaction mixtures. The kit can be used for purification of DNA fragments from 100 bp to 20 kb. The recovery rates are 90 to 100% for 100 bp to 3 kb DNA fragment size range. Process overview
Bacterial cells are pelleted, resuspended, and lysed. Thelysate is then neutralized and conditions are adjusted forsubsequent binding. After clarification by centrifugation, thelysate is processed through the PureLink spin column. TheDNA binds to the silica-based membrane in the column, andimpurities are removed by a single wash step. The DNA isthen eluted in Elution Buffer or water.

iPrep™ ChargeSwitch™ gDNA Tissue Kit (Invitrogen™)

The iPrep™ ChargeSwitch® gDNA Tissue Kit allows rapid and efficient purification of DNA from a wide range of tissue samples types. Designed specifically for use with the iPrep™ Purification Instrument, this kit yields larger amounts of highly pure DNA than can be achieved with competing systems (Table 1)-with many tissues, up to 2-fold greater yields are typical. The resulting DNA is also highly pure (Figure 1), and is ideal for use in PCR (Figure 2).

The iPrep™ ChargeSwitch® gDNA Tissue Kit takes advantage of the powerful ChargeSwitch® technology (Figure 3), a major advance in DNA purification. MICROBEnrich™ Kit (Invitrogen™)

For the separation and isolation of bacterial RNA from mixed samples. The Ambion® kit includes sufficient reagents for depleting 500 µg of host cell RNA (20 reactions x 25 µg RNA) from a complex host-bacterial RNA mixture. Gene expression analysis following host-bacterial interactions is revolutionizing our understanding of the host response to bacterial infection. Numerous studies have utilized microarray analysis to follow host cell transcriptome alterations in response to interactions with bacteria. However, because no method has existed to isolate bacterial RNA away from host cell RNA, the corresponding analyses of bacterial transcriptomes has been impossible. The MICROBEnrich™ Kit enables whole genome microarray expression analyses of bacterial RNA after host-bacterial interactions.

Start With Any Total RNA Sample
For microarray analysis and other sensitive applications, a DNase treatment is recommended to remove contaminating genomic DNA from the total RNA prior to the MICROBEnrich™ Kit procedure.

Remove >90% of Mammalian RNA from Complex Total RNA Mixtures
The MICROBEnrichKituses hybridization capture technology (patent pending) to remove human, mouse, and rat RNA (both mRNA and rRNA) from complex host-bacterial RNA populations, leaving behind enriched microbial total RNA. In the first step of the procedure, host-bacterial total RNA is incubated with a mixture of capture oligonucleotides that bind the mammalian 18S and 28S rRNAs and polyadenylated RNAs. Next, the rRNA/oligo nucleotide hybrids and all polyadenylated mRNAs are removed from the mixture with oligonucleotide-derivatized magnetic beads. After magnetic separation, the bacterial RNA remaining in the supernatant and can be precipitated with ethanol. An Agilent 2100 bioanalyzer electropherogram (see Figure) shows nearly complete removal (>95%) of the host 18S and 28S rRNAs from a mixed sample. Bacterial mRNAs purified with the MICROBEnrich™ Kit are better templates than total RNA for downstream applications and therefore deliver dramatic increases in sensitivity when used in applications such as array analysis (see Figure).

The MICROBEnrich™ Kit Will Work With Any Bacterial Species
The MICROBEnrich Kit was optimized using E. coli RNA in a background of human, mouse, and rat RNA. However, the kit should perform well with RNA from any bacterial species. Although many mammalian host species may be compatible with the MICROBEnrichKit, only human, mouse, and rat rRNA sequences were used for the design and optimization of the capture oligonucleotides.

Accessory Products:
A magnetic stand is required for use this kit. Unique method eliminates the need to isolate RNA from cultured cells
The Cells-to-Ct method features a unique lysis reagent that lyses cells and protects RNA. DNA removal occurs simultaneously in the 5-minute lysis step to save time and improve ease of use. Lysis is then stopped by the addition of a “Stop Solution” that irreversibly binds to the lysis reagents.

The TaqMan Fast Advanced Cells-to-CT Kit contains reverse transcription (RT) reagents for cDNA synthesis and TaqMan Fast Advanced Master Mix for real-time PCR analysis. TaqMan primer/probe sets are sold separately.

Simple 7-minute sample preparation
The TaqMan Fast Advanced Cells-to-CT Kit is designed for 10–100,000 cultured cells/sample. Cells are washed in PBS and lysed in solution for five minutes at room temperature; DNase treatment can be performed simultaneously. Lysis is terminated at room temperature by a 2-minute incubation with Stop Solution. The lysates are now ready for reverse transcription or storage at –20°C for a short term or -80°C for a long term.

Maximize sensitivity of detection
Unlike some competitor kits that limit the amount of lysate in the RT reaction to 10%, the TaqMan Fast Advanced Cells-to-CT Kit can accommodate 45% of the total RT reaction volume as cell lysate. Additionally, cDNA can comprise up to 45% of the real-time PCR reaction volume. The large lysate volume in the optimized RT reaction, along with the large cDNA volume in the subsequent real-time PCR using the TaqMan Fast Advanced Master Mix, lead to maximum sensitivity. The master mix amplifies the target precisely and accurately, enabling the detection of small quantities of target, such as transcripts expressed at low levels.

Reduced sample loss and transfer error
Because samples can be processed directly in culture wells (96 or 384 wells), sample handling and the potential for sample loss or transfer error are minimized, facilitating rapid, high-throughput processing. Unlike old-fashioned, multi-step RNA isolation protocols, no heating, washing, or centrifugation steps are required. The kit greatly simplifies a laborious 30–60 minute process and reduces it to 7 minutes.

Gene expression profiling and analysis of whole blood is becoming an increasingly important life science tool. Accurately measuring the molecular profile of blood-based RNA transcripts is a critical need in areas of research such as disease association studies, biomarker discovery and validation, and monitoring of drug treatment regimes.

Accessory Product
The highly unstable nature of RNA transcripts ex vivo creates an even greater challenge for accurate expression analysis. Ongoing metabolic changes can drastically affect gene expression profiles in as little as one hour in standard blood collection tubes (see Figure). Use of the Applied Biosystems® Tempus™ Blood RNA Tubes (SKU #4342792) is therefore recommended. These tubes are designed for direct collection of 3 mL of blood into a plastic, evacuated blood collection tube containing RNA Stabilizing Reagent. The RNA, and therefore the expression profile, is stable in the collection tube for up to five days at room temperature and minimally 7 days at 4°C (see Figure).

Performance with Ease
The ability of the Tempus™ system (Tempus™ Blood RNA Tubes and Tempus™ Spin RNA Isolation Kit) to stabilize RNA expression profiles has been tested over a range of storage conditions (see Figure). Rapid Purification Protocol
The ChargeSwitch® gDNA Normalized Buccal Cell Kit allows purification of genomic DNA from human buccal cell swabs or pelleted cells from a mouthwash in less than 15 minutes after lysates preparation. ChargeSwitch® gDNA Normalized Buccal Cell Kit uses magnetic beads with a unique, ionizable functional group whose affinity for nucleic acids is pH-dependent to facilitate nucleic acid purification. High-quality genomic DNA is purified at a normalized concentration of 1-3 ng/ µl in a total volume of 150 µl (Figure 1) by eliminating the need for lengthy quantitation and dilution steps. After employing the ChargeSwitch® gDNA Normalized Buccal Cell Kit, purified gDNA is suitable for use in STR analysis of poor or environmentally degraded samples, and for downstream applications such as PCR.

Compatible with Single Tube and High-Throughput Protocols
The ChargeSwitch® gDNA Normalized Buccal Cell Kit is designed for automated processing of both small numbers of samples individually, as well as large numbers of samples using a liquid handling robot or a multi-channel pipette.

For processing up to 24 individual samples at a time the MagnaRack™ two-piece magnetic separation tube rack is required. This rack consists of a magnetic base station and a removable tube rack, which can hold up to 24 microcentrifuge tubes and can be used with most ChargeSwitch® products.

For high-throughput applications utilizing 96 well blocks, the 96-Well Magnetic Separator is required. This rack can hold and separate up to 96 magnetic bead samples in a deep well plate when placed on top of the magnetic base station.
