Rapid Purification Protocol
The ChargeSwitch® gDNA Normalized Buccal Cell Kit allows purification of genomic DNA from human buccal cell swabs or pelleted cells from a mouthwash in less than 15 minutes after lysates preparation. ChargeSwitch® gDNA Normalized Buccal Cell Kit uses magnetic beads with a unique, ionizable functional group whose affinity for nucleic acids is pH-dependent to facilitate nucleic acid purification. High-quality genomic DNA is purified at a normalized concentration of 1-3 ng/ µl in a total volume of 150 µl (Figure 1) by eliminating the need for lengthy quantitation and dilution steps. Samples can be stored for up to 2 weeks at 4°C before processing without a noticeable loss in DNA yield or quality.

Note: DNA yield varies and is dependent on several factors including the technique of the person taking the swab, whether the donor is a high or low shedder, and the type of swab used.

Validated for Single Tandem Repeat (STR) Profiling
After employing the ChargeSwitch® gDNA Normalized Buccal Cell Kit, purified gDNA is suitable for use in STR analysis of poor or environmentally degraded samples, and for downstream applications such as PCR.

Compatible with Single Tube and High-Throughput Protocols
The ChargeSwitch® gDNA Normalized Buccal Cell Kit is designed for automated processing of both small numbers of samples individually, as well as large numbers of samples using a liquid handling robot or a multi-channel pipette.

For processing up to 24 individual samples at a time the MagnaRack™ two-piece magnetic separation tube rack is required. This rack consists of a magnetic base station and a removable tube rack, which can hold up to 24 microcentrifuge tubes and can be used with most ChargeSwitch® products.

For high-throughput applications utilizing 96 well blocks, the 96-Well Magnetic Separator is required. This rack can hold and separate up to 96 magnetic bead samples in a deep well plate when placed on top of the magnetic base station.

Simple Workflow Without Sacrificing Sensitivity & Specificity
The process consists of pelleting and washing a 500uL aliquot from the blood tube, followed by a unique digestion⁄DNase step which preserves RNA integrity. The preparation takes place entirely at room temperature unlike other products that require temperature-specific incubation steps. Up to 96 samples can be prepared in less than an hour, half the time it takes for competitive spin column purification to process only 12 samples, with less hands-on effort. The resulting lysate can go directly into one-step or two-step qRT-PCR reactions or stored for later use (1 hour at 4°C or 5 months frozen). This straightforward procedure takes place in the same sample processing well, from start to finish, meaning no sample transfers. Maximize Yields of Total DNA
High yield of DNA and minimal DNA degradation is achieved by using young plant samples such as leaves and by freezing the sample in liquid nitrogen immediately after collection. Total DNA is eluted in 2 aliquots of 100 µl each to obtain greater DNA yield, with the first elution generating between 65-80% DNA recovery and the second elution > 90%. Typically, DNA isolated using the PureLink® Genomic Plant DNA Purification Kit has an A260/A280 > 1.80 when samples are diluted in Tris-HCl (pH 7.5), indicating that the DNA is reasonably free of proteins that could interfere with downstream applications.

How It Works

ChargeSwitch® technology employs a completely novel mechanism that replaces harsh reagents such as chaotropic salts, ethanol, or organic solvents and isopropanol precipitation with a simple ion-exchange procedure. In this method, DNA binds to a surface ligand that is positively charged at low pH, and is eluted when the pH is raised to 8.5 (Figure 1). The ChargeSwitch®-Pro protocol retains the ease and familiarity of common silica spin-column protocols with binding, washing, and elution steps (Figure 2).


Better Downstream Results

Since ChargeSwitch® technology does not require the use of ethanol, chaotropic salts, or organic solvents for nucleic acid purification, the resulting DNA is free of enzymatic inhibitors and thus shows better downstream performance (Figure 3). These compounds are known to inhibit many enzymatic reactions such as PCR, cloning, restriction enzyme digestion, and array CGH labeling. Expect to see reduced variability in your sequencing applications, with reads in excess of 800 bp with Phred scores >20 (Figure 4); clean restriction enzyme digestions (Figure 5); and removal of dNTPs and primers in excess of 98% (Figure 6).

MagMAX™ Saliva gDNA Binding Beads (Applied Biosystems™)

These magnetic binding beads are a component of the MagMAX Saliva gDNA Isolation Kit. They are offered here as a stand-alone reagent for applications that require more magnetic binding beads than is provided in the kit.

Benefits of MagMAX™ Magnetic Bead-Based Purification:
Magnetic beads offer many benefits compared to other technologies for isolating RNA. Beads bind RNA more efficiently than glass fiber filters, resulting in higher and more consistent RNA yields. Additionally, because filters are not used, there is no risk of filter clogging due to cellular particulates in samples. Since only a small volume of magnetic beads is needed for high-efficiency binding, the bound RNA can be eluted in just 20–50 µL of nuclease-free water, concentrating RNA from large, dilute samples.

Optimized for Isolation of Virus from Biofluids:
The Applied Biosystems® MagMAX™ Viral RNA Isolation Kit has been optimized for use with biological fluids and cell-free samples such as serum, plasma, swabs, and cell culture media. These samples often present unique challenges when purifying RNA, including low viral titers, large volumes to be processed, and sample complexity. Without the need for organic solvents or RNA precipitation, the MagMAX™ Viral RNA isolation kit eliminates the problems often encountered with other RNA isolation methods such filter clogging, large elution volumes, and inconsistent yields.

Get the Same Amount of RNA from 10-fold Less Sample
The sensitivity of viral RNA detection was compared using RNA isolated with Applied Biosystems® MagMAX™ protocol or the current National Veterinary Services Laboratories (NVSL) validated viral testing protocol. Even at one-tenth the sample input, viral RNA isolated using the MagMAX™ kit could be detected at equal or greater levels than RNA isolated using the NVSL protocol and recommended competitor's kit (see data). Accessory Products:

Principle

A bacterial culture is harvested and lysed. The lysate is then cleared by centrifugation and applied on the silica column to selectively bind DNA molecules at a high salt concentration. The adsorbed DNA is washed to remove contaminants, and the pure plasmid DNA is eluted in a small volume of elution buffer or water. The purified DNA is ready for immediate use in all molecular biology procedures, such as fast and conventional digestion with restriction enzymes, PCR, in vitro transcription, transformation, and automated sequencing. MagMAX™ Plant DNA Isolation Kit (Applied Biosystems™)

The MagMAX™ Plant DNA Isolation Kit is designed for automated high-throughput or manual purification of DNA from a wide variety of plant species. The kit uses MagMAX magnetic bead technology, eliminating the need for phenol/chloroform extraction or alcohol precipitation. The procedures are optimized for the isolation of DNA from 10–100 mg plant samples. DNA yields vary between different species and tissues depending on genome size, ploidy, cell number, age of tissue sample, and storage conditions.

The MagMAX Plant DNA Isolation Kit relies on magnetic bead technology, enabling reproducible yields and robust performance. Benefits of MagMAX magnetic bead–based purification
The magnetic bead–based purification format of the kit allows for easy scaling of the number of samples processed either manually with a magnetic stand or with a magnetic particle processor such as the KingFisher™ Duo Prime or KingFisher™ Flex processors.

Magnetic beads offer many benefits compared to other technologies for isolating nucleic acids. Beads bind the RNA and DNA more efficiently than glass fiber filters, resulting in higher and more consistent yields. Additionally, because filters and vacuum manifolds are not used, there is no risk of undigested tissue particulates clogging these items during the extraction process. This clogging issue is of particular concern with fibrous plant tissue samples that that are not fully lysed during the disruption and homogenization steps.

How it works

The ChargeSwitch® technology replaces the use of ethanol, chaotropic salts, or organic solvents for nucleic acid purification with a simple ion-exchange mechanism. At low pH values (<6.5) the ChargeSwitch® surface ligand is positively charged and binds plasmid DNA. Raising the pH (>8.5) neutralizes the charge and elutes the bound nucleic acid. The ChargeSwitch® protocol retains the ease and familiarity of the common silica spin column protocol with a binding, washing, and elution step (Figure 1, Table 1), making adapting to the ChargeSwitch® method effortless.

DNA purified using the ChargeSwitch®-Pro Plasmid Miniprep Kit lacks potentially inhibitory compounds, improving DNA integrity for better downstream PCR performance (Figure 2). Expect to see improved cloning due to higher transformation efficiencies (Figure 3), faster restriction enzyme digestions (Figure 4), sequencing reads in excess 800 bp with Phred scores >20 (Figure 5), and a higher degree of supercoiling (Figure 6).

RiboMinus™ Transcriptome Isolation Kit, bacteria (Invitrogen™)

RiboMinus™ Transcriptome Isolation Kits are novel purification systems that enrich the whole spectrum of RNA transcripts by depleting large ribosomal RNA molecules in a total RNA sample. These kits use a 5µ biotin-labeled, Locked Nucleic Acid (LNA) probe specific for large rRNA to remove 95-98% of rRNA molecules from human, mouse, bacteria, or yeast total RNA (Figure 1). This removal increases the representation of RNA transcript species, including non-poly(A) tailed, pre-processed RNA, small tRNAs, siRNA, snRNA, microRNA, fragmented mRNA, and other regulatory RNAs that cannot be purified via poly(A) selection methods. The RiboMinus™ Transcriptome Isolation Kits enable:

• Comprehensive analysis of the whole transcriptome without contamination or interference from rRNA
• More complete collection of transcribed elements of the genome—no reliance on poly(A) purification methods

• Cleaner, clearer microarray analysis due to higher signal-to-background ratios (Figure 2)

Purified RNA is concentrated and suitable for numerous downstream applications, including microarray analysis (Figure 2), library construction, and qRT-PCR.

RiboPure™ RNA Purification Kit, blood (Invitrogen™)

The RiboPure™-Blood Kit is for the isolation of high-quality RNA directly from whole blood. The kit combines two methods of RNA purification to efficiently remove blood components (protein, heme, genomic DNA, and RNases) that can compromise downstream procedures. The kit includes sufficient reagents for 40 RNA purifications from whole blood samples ranging from 0.3–0.5 mL each. Advantages of the RiboPure™-Blood Kit:

• Purify total RNA directly from whole blood—without prior recovery of WBCs
• Simple, fast procedure—from blood to RNA in less than 30 minutes
• Includes RNAlater® Solution—store blood samples 3 days or more at room temperature
• Includes DNA-free™ Reagents—to ready RNA for RT-PCR and array analysis

Applications
The RiboPure™-Blood Kit produces high yields of pure RNA directly from whole blood, eliminating the need to isolate WBCs prior to RNA purification. RNA recovered using the RiboPure™-Blood Kit can be used in the most challenging applications, including real-time RT-PCR and array analysis. Average yields of RNA are between 2–4 µg/0.5 mL of whole blood (variation is due to differences in WBC numbers between samples).

Stabilize RNA in whole blood
When collecting blood samples for RNA analysis, storage of the samples prior to RNA isolation is often required. The RiboPure™-Blood Kit contains RNAlater® Solution for the stabilization of RNA in whole blood. It also "freezes" the gene expression profile of the cells. Treated samples can be safely stored at ambient temperature for extended periods of time (up to three days or more). Blood samples stored in RNAlater® Solution yield RNA of comparable quality to blood samples processed directly (see figure).

Simple procedure yields highly pure RNA
The RiboPure™-Blood procedure consists of three steps: 1) addition of Lysis Solution directly to RNAlater®-treated or fresh whole blood, 2) initial RNA purification by phenol/chloroform extraction, and 3) final RNA purification on a glass fiber filter. An additional DNase treatment can then be performed with the DNA-free™ Reagents included in the kit. The entire procedure, including DNase treatment, can be completed in less than 1 hour.

Accessory products
The RNA can subsequently be enriched for white blood cell (WBC) mRNA using our GLOBINclear™ Kits to selectively remove globin transcripts. The Mouse RiboPure™-Blood RNA Isolation Kit (Cat. No. AM0951) is also available for the recovery of total RNA—and optionally microRNA—directly from mouse or rat blood. Mouse RiboPure™-Blood includes sample tubes preloaded with RNAlater® Solution to prevent RNA degradation and preserve RNA profiles.
