Simple and Fast miRNA Purification
The TaqMan® miRNA ABC (anti-miRNA bead capture) purification kit contains buffers and magnetic beads for single-tube isolation of specific microRNA (miRNA) from small inputs of all human sample types, including blood, serum, plasma, FFPE samples, solid tissues, cultured cells, and saliva in 75 minutes (see workflow, Figure 1).

Human Panel A and Human Panel B
The TaqMan® miRNA ABC Purification Kit is offered in two configurations: Human Panel A and Human Panel B. The beads in each kit are superparamagnetic Dynabeads® covalently bound to a unique set of 377 anti-miRNA oligonucleotides for each panel that correspond to the assays contained in the TaqMan® Array Human MicroRNA A and B cards. The miRNA isolation process relies on hybridization of miRNAs to the corresponding anti-miRNA oligonucleotides attached to the beads. Complete lists of miRNAs isolated by the panels can be found in the TaqMan® miRNA ABC Purification Kit protocol (below).

miRNA Analysis
The miRNAs isolated using the TaqMan® miRNA ABC Purification Kit are ready for conversion into cDNA and downstream analysis by quantitative PCR using either single tube (individual) TaqMan® microRNA assays or the TaqMan® Array Human microRNA A and B cards. Figure 2 gives examples of the data that can be obtained from miRNA isolated from various sample types using the TaqMan® miRNA ABC Purification Kit.

PureLink™ miRNA Isolation Kit (Invitrogen™)

The Ambion® PureLink® miRNA Isolation Kit provides a simple, reliable, and rapid column-based method for isolating high-quality total microRNA (miRNA) from a wide variety of sources without the need for hazardous reagents such as phenol. The kit provides a rapid and convenient method for purifying small RNA molecules from animal and plant cells, as well as from bacteria and yeast samples.
• Purify small RNA molecules with a novel enrich/bind/elute procedure

Rapid and Convenient Column Purification Procedure for small RNAs
The Ambion® PureLink® miRNA Isolation Kit is specifically designed to purify small RNA molecules, including miRNAs and siRNAs, from a variety of samples typically in less than 15 minutes (see Table 1). The kit utilizes a silica-based two-column system to enrich exclusively for small RNAs (see Figure 1). RNAs greater than approximately 200 nucleotides are bound to the first column. Ethanol is then added to the flow-through in order to increase the binding affinity of small RNAs to the glass fiber membrane in the second column. Once bound to the second column, small RNA molecules are then eluted, ready-for-use, and free of large RNA molecules such as 28S rRNA, 18S rRNA, and mRNA, allowing for focused expression analysis studies of small RNA molecules.

Purified RNA is Suitable for Use with a Variety of Downstream Applications and Products:
The miRNA purified using the Ambion® PureLink® miRNA Isolation Kit is suitable for downstream applications including RNA sequencing, microarray analysis, real-time PCR (qPCR), reverse transcription real-time PCR (RT-qPCR), and northern blotting.

mirVana™ miRNA Isolation Kit, with phenol (Invitrogen™)

The mirVana™ miRNA Isolation Kit uses a rapid procedure to isolate small RNAs from tissue and cells using an efficient glass fiber filter (GFF)-based method. The method isolates total RNA ranging in size from kilobases down to 10-mers. Each kit contains sufficient reagents and consumables for either 40 isolations of total RNA (including small RNAs), or 20 separate large and small RNA fractions.

Features of this kit include:

• Efficient isolation of small RNA-containing total RNA
• Enrich for small RNA (<200 nt) to increase sensitivity in downstream analyses

Efficient recovery of small RNA
Current RNA purification procedures rely on organic extraction, followed by alcohol precipitation or adsorption of nucleic acid molecules on a GFF or silicate matrix. The latter procedure typically results in the loss of substantial amounts of small RNA. The mirVana™ miRNA Isolation Kit uses organic extraction followed by purification on a GFF under specialized binding and wash conditions. As a result, the kit effectively recovers all RNA—from large mRNA and ribosomal RNA down to 10-mers—from virtually all cell and tissue types. The mirVana™ miRNA Isolation Kit enables the isolation of small RNA-containing total RNA from samples consisting of 102–107 cultured cells or 0.5–250 mg tissue.

Enrich for small RNA
Although the mirVana™ miRNA Isolation Kit efficiently purifies all RNA larger than 10 nt, it also includes procedures for isolating RNA fractions, specifically enriched or depleted in small RNA species. Through the enrichment procedure included in the mirVana™ miRNA Isolation Kit, RNA molecules of ~200 nt and less can be efficiently purified away from the larger RNA species. The resulting RNA preparation is highly enriched for miRNAs, siRNAs, and/or snRNAs. Small RNA enrichment using the mirVana™ miRNA Isolation Kit procedure allows more sensitive small RNA detection with less background as compared to the same assay used with total RNA.
