PureLink™ HiPure Plasmid FP (Filter and Precipitator) Maxiprep Kit Invitrogen™

The PureLink™ HiPure Plasmid FP (Filter and Precipitator) Maxiprep Kit isolates plasmid DNA, from cell pellet to ultrapure DNA, in less than one hour. Using the HiPure Filter Columns and HiPure Precipitators supplied in the kit, you can purify plasmid DNA without the centrifugation steps required in traditional isolation protocols. The quality of the purified plasmid DNA is equivalent to that obtained by two passes through cesium chloride gradients - the most rigorous method for DNA purification - making it suitable for transfection and other applications requiring high-purity plasmid DNA. How it works
The PureLink™ HiPure Plasmid FP Maxiprep Kit employs an alkaline lysis method to prepare and rapidly clear bacterial lysates without the need for centrifugation (Figure 1). The unique HiPure Filter Column includes a lysate filtration cartridge integrated into the anion-exchange column for single-step lysate clarification and plasmid DNA binding. Impurities such as RNA, proteins, and other contaminants are removed with a single wash, and ultrapure plasmid DNA is eluted with a high-salt buffer. To precipitate and desalt the DNA, isopropanol is added to the eluted DNA and the mixture is then applied to the HiPure Precipitator using a large syringe. After a subsequent washing and drying step, the plasmid DNA is easily eluted from the HiPure Precipitator with either TE buffer or water. The use of the precipitator eliminates the need for centrifugation, reducing the risk of losing the DNA pellet during supernatant removal, and saving time. The resulting DNA is ready to use in the most challenging applications. The entire protocol - from E. coli cell pellet to ultrapure transfection-grade plasmid DNA - can be completed in about an hour.

Highest yields and ready-to-use concentrations
The HiPure Precipitator has been optimized to allow for lower elution volumes (down to 500 µl) to produce highly concentrated DNA without a significant decrease in yields. In fact, yields at higher concentrations are consistently higher than the competitor kits (Figure 2). Yields and concentration can be adjusted to meet the needs of your downstream application.

How it works
The ChargeSwitch® technology replaces the use of ethanol, chaotropic salts, or organic solvents for nucleic acid purification with a simple ion-exchange mechanism. At low pH values (<6.5) the ChargeSwitch® surface ligand is positively charged and binds plasmid DNA. Raising the pH (>8.5) neutralizes the charge and elutes the bound nucleic acid. The ChargeSwitch® protocol retains the ease and familiarity of the common silica spin column protocol with a binding, washing, and elution step, making adapting to the ChargeSwitch® method effortless.

The PureLink® HiPure Plasmid Filter Maxiprep Kit is designed to isolate plasmid DNA of the highest purity from 100 to 500 mL of an overnight bacterial culture. How it works
The addition of a HiPure Filter Column to the PureLink® HiPure Plasmid Kits provides rapid clearing of the bacterial lysate without the need for centrifugation (see figure). The lysate filtration cartridge is integrated into the DNA-binding column for a combined step of lysate clarification and plasmid DNA binding directly to the anion-exchange resin. A single column wash removes impurities such as RNA, proteins, metabolites, and other low molecular weight molecules; the ultrapure plasmid DNA is then eluted with a high-salt buffer. The DNA is then desalted and concentrated with an isopropanol precipitation step, then collected by centrifugation. The entire protocol can be completed in about two hours.

GeneJET Plasmid Midiprep Kit Thermo Scientific™

Thermo Scientific GeneJET Plasmid Midiprep Kit, sufficient for 100 preps, is designed for large scale isolation of high quality plasmid DNA from recombinant E. coli cultures. The PureLink™ HiPure Precipitator allows fast, simple, and efficient desalting and concentration of plasmid DNA isolated using anion-exchange chromatography. The traditional method for DNA precipitation of anion exchange isolated plasmid DNA involves isopropanol precipitation followed by centrifugation and drying of DNA,which is time consuming and labor intensive. The use of the PureLink™ HiPure Precipitator allows plasmid DNA concentration within 5 minutes, eliminates the need for centrifugation, and reduces the risk of losing the DNA pellet during supernatant removal. The recovery is >80% and resulting plasmid DNA is ready to use in the most challenging applications such as transfection.

Contaminant-free DNA
Unlike cesium chloride gradient protocols, PureLink® HiPure Plasmid Purification Systems do not use organic solvents, ethidium bromide, or cesium chloride, all of which are troublesome to work with and to dispose of. Typically, plasmid DNA prepared using the PureLink® HiPure Plasmid Midiprep Kit will have an A260/A280 ratio of >1.80, indicating that the DNA is reasonably free of proteins that might interfere with downstream applications. Endotoxin levels are typically within 0.1–1 EU/µg, making this plasmid DNA ideal for mammalian cell transfection.

PureLink™ HiPure Expi Plasmid Gigaprep Kit Invitrogen™

The PureLink™ HiPure Expi Plasmid Gigaprep Kit enables isolation of the highest levels of transfection-grade plasmid DNA from a single E.coli giga prep using an enhanced anion exchange resin. The PureLink™ HiPure Plasmid Gigaprep Kit also includes vacuum filtration columns that provide bacterial filtration without centrifugation, enabling extremely fast process times.
