Isolate genomic DNA from multiple sample types with a single sample prep kit
The MagMAX DNA Multi-Sample Ultra 2.0 Kit isolates gDNA from 50 µL–2 mL of whole blood (figure below), fresh and stabilized saliva (figure below), and EDTA/Streck cfDNA tube buffy coat (figure below) samples. Additionally, the same kit can be used to isolate gDNA from buccal swabs and other biological sample types. The MagMAX DNA Multi-Sample Ultra 2.0 Kit maximizes gDNA yields by treating samples with proteinase K (PK) and PK-enhancer followed by an innovative lysis/binding solution to simultaneously lyse cells, denature proteins, and capture gDNA on the high-binding capacity Ultra 2.0 gDNA magnetic binding beads. The Ultra 2.0 Wash I solution removes proteins, detergents, and other impurities from the bound nucleic acid. The gDNA is then released from the beads using a low-TE elution solution.

The MagMAX DNA Multi-Sample Ultra 2.0 Kit is optimized for use on the moderate-throughput KingFisher Duo Prime (Cat. No. 5400110) and the higher-throughput KingFisher Flex (Cat. No. 5400630) purification systems. Users can process up to 400 µL of sample volume in high-throughput SBS 96-well deep-well microtiter plates using the Flex system. For smaller sample batches, users can process 12 samples using the Duo Prime system. Larger sample input volumes (1–2 mL), which are more challenging to process, are purified in SBS 24-well deep-well microtiter plates for either 24 samples on the Flex or 6 samples on the Duo Prime system. ChargeSwitch™ gDNA Rendered Meat Purification Kit (Invitrogen™)

The ChargeSwitch® gDNA Rendered Meat Purification Kit allows for rapid and efficient purification of genomic DNA (gDNA) from animal feed, animal meal, and heparin products for downstream molecular analysis. The kit is listed in the U.S. Food and Drug Administration Center for Veterinary Medicine’s protocols as a sample preparation method suitable for monitoring crude heparin quality and testing animal feed for banned materials such as specific animal proteins.

Note: The number of reactions possible with this kit is dependent upon the starting sample type: animal feed & meal samples = 50 preparations, heparin products = 25 preparations.

The ChargeSwitch® gDNA Rendered Meat Purification Kit protocol starts with prepared lysates and takes approximately 15 minutes to complete without centrifugation, vacuum, or organic solvents (for batches of up to 5 samples per operator—larger batch sizes will require longer processing times). For highly predictable and consistent extraction and isolation of genomic DNA from human whole blood, following a simple separation protocol. Isolates linear DNA yields relative to the number of white blood cells (WBC) present in the sample. The 1 µm Dynabeads® have an optimized silica-like surface chemistry and a high specific surface area, providing efficient kinetics and a high capacity.

PureLink™ Genomic DNA Mini Kit (Invitrogen™)

The PureLink® Genomic DNA Mini Kit enables high-yield, high-purity, genomic DNA (gDNA) extractions from a wide variety of sample types. Use this single kit for genomic DNA purification from blood, tissues, cells, bacteria, swabs, and blood spots, with a familiar silica-based, microcentrifuge spin-column format. The PureLink® Genomic DNA Mini Kit can be used with many different sample types, each with its own optimized protocol outlined in the manual.

Optimized components and protocols
The PureLink® Genomic DNA Mini Kit includes parts and protocols that improve upon the easy and familiar spin-column methods currently in use (see figure). Columns are newly configured to bind and release DNA more efficiently (see figure). Each sample source uses a specialized lysis procedure and an optimized lysis buffer formulated to enhance proteinase K activity and eliminate protein contamination. A chaotropic salt buffer promotes stable binding of DNA to the column resin, and powerful wash buffers remove all traces of protein and salt. DNA is eluted in a low-salt buffer to allow for pH stabilization of the DNA in storage.

Higher yields and purity
With PureLink® Genomic DNA Kits you can expect high yields of high-purity gDNA (determined by A260/A280 measurements), no matter the sample type, from bacteria to tissues to blood to cultured cells (see figures). The enhanced PureLink® Lysis and Wash Buffers help to consistently deliver purified gDNA samples with absorbance reading ratios of ~1.9 for both A260/A280 and A260/A230, indicating no detectable contamination from carryover of protein or guanidine (see figure).

MagMAX™ DNA Multi-Sample Ultra 2.0 Proteinase K (Applied Biosystems™)

This Proteinase K solution is included in the MagMAX DNA Multi-Sample Ultra 2.0 Kit. It is available separately for applications that require more Proteinase K solution than is provided in the kit.
