Dynabeads™ SILANE Genomic DNA Kit (Invitrogen™)

For highly predictable and consistent extraction and isolation of genomic DNA from human whole blood, following a simple separation protocol. Isolates linear DNA yields relative to the number of white blood cells (WBC) present in the sample. The 1 µm Dynabeads® have an optimized silica-like surface chemistry and a high specific surface area, providing efficient kinetics and a high capacity.

Their uniform size and surface area ensure highly reproducible results and an improved performance compared to magnetic particles from alternative suppliers. Other particles often have a random size range distribution, surface area and binding capacity. This could compromise the reproducibility of your results. GeneJET FFPE DNA Purification Kit (Thermo Scientific™)

Thermo Scientific GeneJET FFPE DNA Purification Kit is designed for fast and convenient purification of DNA from various amounts of archived FFPE tissue. Up to 8 sections (10 µm thickness) of FFPE sample from various tissues can be used for genomic DNA extraction without overnight incubation. Elimination of toxic reagents commonly used for deparaffinization (i.e., xylene) allows a safe DNA purification procedure with high DNA yields and excellent quality. Each preparation recovers up to 8 µg of genomic DNA that can be eluted in volumes from 20 µL allowing for concentrated samples. Isolate genomic DNA from multiple sample types with a single sample prep kit
The MagMAX DNA Multi-Sample Ultra 2.0 Kit isolates gDNA from 50 µL–2 mL of whole blood (figure below), fresh and stabilized saliva (figure below), and EDTA/Streck cfDNA tube buffy coat (figure below) samples. Additionally, the same kit can be used to isolate gDNA from buccal swabs and other biological sample types. The MagMAX DNA Multi-Sample Ultra 2.0 Kit maximizes gDNA yields by treating samples with proteinase K (PK) and PK-enhancer followed by an innovative lysis/binding solution to simultaneously lyse cells, denature proteins, and capture gDNA on the high-binding capacity Ultra 2.0 gDNA magnetic binding beads. The Ultra 2.0 Wash I solution removes proteins, detergents, and other impurities from the bound nucleic acid. The gDNA is then released from the beads using a low-TE elution solution.

The MagMAX DNA Multi-Sample Ultra 2.0 Kit is optimized for use on the moderate-throughput KingFisher Duo Prime (Cat. No. 5400110) and the higher-throughput KingFisher Flex (Cat. No. 5400630) purification systems. Users can process up to 400 µL of sample volume in high-throughput SBS 96-well deep-well microtiter plates using the Flex system. For smaller sample batches, users can process 12 samples using the Duo Prime system. Larger sample input volumes (1–2 mL), which are more challenging to process, are purified in SBS 24-well deep-well microtiter plates for either 24 samples on the Flex or 6 samples on the Duo Prime system. KingFisher instrument BindIt software scripts for all sample types and both Flex and Duo Prime systems can be downloaded below and installed onto the KingFisher instruments.

iPrep™ PureLink™ gDNA Blood Kit (Invitrogen™)

The iPrep™ PureLink® gDNA Blood Kit allows rapid and automated extraction of genomic DNA (gDNA) from human blood including fresh or frozen blood samples, and concentrated leukocyte preparations (buffy coat). Genomic DNA is extracted from blood samples using the Dynabeads® MyOne™ SILANE and iPrep™ Purification Instrument within 30 minutes without the use of centrifugation. The purified genomic DNA is suitable for use in downstream applications, including PCR. Advantages of the iPrep™ PureLink® gDNA Blood Kit:

iPrep™ Purification Instrument
The iPrep™ PureLink® gDNA Blood Kit is designed for use with the iPrep™ Purification Instrument. The iPrep™ Purification Instrument is a benchtop, automated nucleic acid purification instrument with integrated Magnetic and Syringe Unit capable of purifying nucleic acids from up to 13 samples (12 samples + 1 positive control) using magnetic bead-based technology.

Using the iPrep™ PureLink® gDNA Blood Kit
With A260/A280 ratios of 1.8, the resulting DNA is highly pure (see figure), ideal for use in sensitive multiplex assays (see table) or for long-term storage. This kit is optimized to isolate highly-pure DNA from 150 µL to 350 µL of fresh or frozen blood in less than 30 minutes (see figure). It is highly tolerant of anticoagulants such as citrate and EDTA, and yields high molecular weight DNA. The yield is highly reproducible (see figure), and there is no cross-contamination between samples (see figure). Furthermore, the kit can also be used with cultured cells. The table summarizes the performance of the iPrep™ PureLink® gDNA Blood Kit. The iPrep™ PureLink® gDNA Blood Kit comes with all reagents (supplied in prefilled cartridges), tubes, and tips needed to isolate DNA from blood samples. All you need to do is add the cartridges, tips, and your sample tubes to the instrument, select your preferred elution volume, and come back 30 minutes later to collect your pure gDNA in individual screwcap elution tubes.

ChargeSwitch® gDNA Mini Tissue Kit uses magnetic beads with a unique, ionizable functional group whose affinity for nucleic acids is pH-dependent to facilitate nucleic acid purification. Genomic DNA is isolated without the need for hazardous chemicals, centrifugation, or vacuum manifolds to yield up to 30 µg of gDNA from 0.5 cm mouse tail tips or approximately 25 mg of tissue. For spleen tissue, reduce tissue size to 10 mg.

We recommend using the MagnaRack™ Magnetic Rack, which is a two-piece magnetic separation rack that consists of a magnetic base station and a removable tube rack. The tube rack fits onto the magnetic base station in two different positions associating the row of 12 neodymium magnets with a single row of 12 tubes for simple and easy "on the magnet" and "off the magnet" sample processing.

The GeneCatcher™ gDNA Blood Kits allow rapid and efficient extraction of genomic DNA (gDNA) from human blood, including archived or poorly stored blood samples. Genomic DNA is extracted from blood samples using the cost-effective, user-friendly magnetic bead based GeneCatcher™ Technology without the use of centrifugation (Figures 1,2).

Designed specifically for use with the iPrep™ Purification Instrument, the iPrep™ ChargeSwitch® Forensic Kit takes advantage of the powerful ChargeSwitch® technology, a major advance in DNA purification. The ChargeSwitch® Direct 96 gDNA Kit is a solid-phase, high-throughput genomic DNA (gDNA) purification system designed for rapid and efficient isolation of gDNA from fresh or frozen samples. This kit utilizes 96-well plates coated with the ChargeSwitch® nucleic acid-binding surface, whose ionic charge can be switched by changing the pH of the surrounding buffer. At low pH, the positive charge of the ChargeSwitch® surface binds the negatively charged nucleic acids. Proteins and other contaminants that do not bind are washed away, leaving pure DNA ready for downstream applications. This kit is designed for research use only (Limited Use Label License 5).

Rapid, Three Step, pH Based Purification Protocol
The ChargeSwitch® gDNA Plant Kit allows purification of genomic DNA from between 50-100 mg of fungi and plant samples, including leaves, tissues and seeds (Figure 1), to yield up to 7 µg of gDNA typically in less than 15 minutes following sample preparation and lysis. The ChargeSwitch® gDNA Plant Kit uses magnetic beads with a unique, ionizable functional group whose affinity for nucleic acids is pH-dependent to facilitate nucleic acid purification. Genomic DNA is isolated without the use of centrifugation or organic solvents, and is of higher purity than silica-prepared DNA, which is typically contaminated with guanidine salts.

Compatible with Single Tube and High-Throughput Protocols
The ChargeSwitch® gDNA Plant Kit is designed for automated processing of both small numbers of samples individually, as well as large numbers of samples using a liquid handling robot or a multi-channel pipette.

For processing up to 24 individual samples at a time the MagnaRack™ two-piece magnetic separation tube rack is required. This rack consists of a magnetic base station and a removable tube rack, which can hold up to 24 microcentrifuge tubes and can be used with most ChargeSwitch® products.

For high-throughput applications utilizing 96 well blocks, the 96-Well Magnetic Separator is required. This rack can hold and separate up to 96 magnetic bead samples in a deep well plate when placed on top of the magnetic base station.

PrepFiler™ BTA Forensic DNA Extraction Kit (Applied Biosystems™)

The PrepFiler BTA Forensic DNA Extraction Kit has been developed for the extraction of DNA from calcified tissues (bone, tooth), as well as certain adhesive-containing samples such as cigarette butts, tape lifts, and envelope flaps. The kit enables DNA yields and purity comparable to or better than conventional phenol-chloroform-based methods, with an approximate processing time of only 2 to 3 hours.

Optimized for use with bone and tooth, as well as other adhesive-containing samples, and allows you to obtain DNA yields and purity comparable to or better than conventional phenol-chloroform-based methods

• Enables fewer tube transfers and greater overall ease of use than conventional methods, thus minimizing the opportunity for sample mix-ups or contamination
• Utilizes a simple, robust protocol that facilitates ease of implementation and training, and is more “tolerant" to a range of experience levels

Improved Packaging and Configuration
Several packaging and configuration improvements have been implemented for the updated PrepFiler kits:

• An additional wash buffer, PrepFiler Wash Buffer B, has now been incorporated into the kits to minimize the potential for detergent carryover from PrepFiler Wash Buffer A, which can inhibit downstream PCR applications when the maximum volume of DNA extract is used for subsequent PCR amplification.
• The kit packaging has been updated to include all reagents, including magnetic particles, in the same kit box, with a single expiration date.
• Shipping will occur at ambient temperature, and the kits can also be stored upon receipt at ambient temperature.

All plastics consumables for use with the PrepFiler kits will now only be sold separately, to provide the ordering flexibility to select only the plastics consumables needed. Those plastics include:

• PrepFiler Filter Columns for substrate removal and PrepFiler Spin Tubes for sample processing in tube format
• 2 mL Microcentrifuge Tubes and Caps for lysis steps using bone, tooth, or tape

The PicoPure® DNA Extraction Kit offers an easy, streamlined genomic extraction procedure that produces PCR-ready DNA. Extract and amplify DNA in the same tube, without organic extraction or spin columns. Recover DNA from as few as ten cells procured by Laser Capture Microdissection (LCM) or from milligrams of tissue. The PicoPure® DNA Extraction Kit provides conveniently packaged, stable Proteinase K, PCR-compatible DNA Reconstitution Buffer and a complete user guide. Each of ten lyophilized Proteinase K aliquots can be freshly reconstituted to make 150 µl of PicoPure® DNA Extraction Solution-enough for 150 x 10 µl CapSure® HS LCM Cap extractions or 30 x 50 µl CapSure® Macro LCM Cap extractions (See Figure 1).

Maximize DNA Recovery
Because DNA is not lost due to organic extractions or spin columns, the PicoPure® Kit enables reliable and reproducible DNA recovery from as few as ten cells. PCR studies illustrate the advantages of the PicoPure® Kit. Equal amounts of DNA were transferred to PCR, either directly in the PicoPure Kit Buffer or after purification using a DNA Extraction Kit from a leading manufacturer. Quantitative Real-time PCR (QRT-PCR) demonstrates that the PicoPure® Kit samples amplify earlier, suggesting more DNA present, and amplify more consistently, indicating less sample variation, than column-purified samples. The PicoPure Kit's reliably high DNA recovery enables increased detection sensitivity, for example, when amplifying single-copy genes from small samples in mutation analysis or genotyping assays. This high DNA recovery makes the Kit ideal for small samples, such as those prepared by LCM (See Figure 2).

Extract DNA for Any PCR Application
DNA extracted with the PicoPure Kit is compatible with endpoint PCR-performed in the same 0.5 ml tube used for DNA extraction. For gene copy number quantitation, DNA from very small samples can be directly amplified by qRT-PCR without further purification, using platforms such as the StepOnePlus™ Real-time PCR System. For highly sensitive mutation and genotype analysis, DNA can be extracted and directly amplified from small samples, and gene fragments can be separated by dHPLC (denaturing High-Performance Liquid Chromatography), using platforms such as the WAVE® System (Transgenomics). (See Figures 3 & 4)

Compatible with Most Tissue and Cell Preparation Procedures
The PicoPure® DNA Extraction Kit enables successful recovery of genomic DNA from animal tissue sections and cell samples prepared using a wide range of methods*. Superior results are obtained from formalin-fixed paraffin-embedded (FFPE) tissue sections, frozen tissue sections, ethanol fixed cells, and cytological smears. (See Figure 5.)
