ChargeSwitch® PCR Clean-Up Kit is the fastest and most effective method of removing primers, unincorporated dNTPs, thermostable polymerases and salts (Figure 1). This kit is scalable for use on liquid-handling robots. This kit includes all the necessary reagents for the number of preps listed and requires the use of a magnetic accessory. PureLink™ Pro 96 PCR Purification Kit (Invitrogen™)

The PureLink® Pro 96 PCR Purification Kit combines advanced silica plate extraction chemistry with an optimized 96-well plate design for manual or automated processing of PCR cleanup in as little as 20 minutes. The PureLink® Pro 96 PCR Purification Kits provide rapid and efficient removal of short primers, dNTPs, enzymes, short-failed PCR products, and salts from PCR products. The simple PureLink® bind⁄wash⁄elute procedure facilitates routine purifications of double-stranded DNA fragments from 100 bp to 12 kb.

Improved plate design
The PureLink® Pro 96 PCR Purification Kit provides optimized semi-skirted 96-well plates(Figure 1) for improved performance and results. In addition, the plate nozzles with annular collar prevent cross-contamination of samples and improve drying of the silica membrane to prevent ethanol carryover. In addition, sample processing can be performed manually using a benchtop vacuum manifold, by centrifugation, or with an automated liquid handling platform. The semi-skirted plate design is compatible with most vacuum manifolds on robotic workstations.

PCR products purified using the PureLink® Pro 96 PCR Purification Kit demonstrate outstanding performance in downstream applications such as cloning automated fluorescent DNA sequencing, PCR, restriction mapping, and labeling.

PureLink™ PCR Micro Kit (Invitrogen™)

PureLink® PCR Micro Kits are ideal for purifying and concentrating DNA from low-yielding PCR or restriction digestion reactions. These highly purified DNA fragments and amplicons provide excellent results in downstream applications such as DNA sequencing, restriction digestion, cloning, PCR, and labeling. PureLink® PCR Micro Kits provide a simple and rapid method for purifying and concentrating DNA fragments ranging in size from 100 bp-12 Kb. The unique micro-nozzle design allows up to 95% recovery of DNA fragments with as little as a 5 µl elution volume. With less than a 10% coefficient of variance in the elution volume you can be sure that no ethanol is carried over from the wash steps giving you the best chance of success in downstream applications. Removal of primers, dNTPs, enzymes, and salts exceeds 98.5%. This kit can also efficiently remove unincorporated dye-labeled nucleotides from a cDNA labeling reaction.

GeneJET PCR Purification Kit (Thermo Scientific™)

Thermo Scientific GeneJET PCR Purification Kit utilizes a proprietary silica-based membrane technology in the form of a convenient spin column, eliminating the need for tedious resin manipulations or toxic phenol-chloroform extractions. It effectively removes primers, dNTPs, unincorporated labeled nucleotides, enzymes, and salts from PCR and other reaction mixtures.

The kit can be used for purification of DNA fragments from 25 bp to 20 kb with recovery rates up to 100%. Each GeneJET purification column has a total binding capacity of up to 25 µg of DNA, and the entire procedure takes 5 minutes. The purified DNA can be used in common downstream applications, such as sequencing, restriction digestion, labeling, ligation, cloning, in vitro transcription, blotting or in situ hybridization.

The PureLink PCR Purification Kit provides a more convenient, rapid, and high-yielding purification process compared to other companies' kits. After PCR amplification, purification of the PCR amplicons is required for many common downstream applications, including enzyme digestion, ligation, sequencing, and labeling experiments, to prevent enzymes (polymerase, restriction enzyme) and reaction components (dNTPs, primers, buffers, salts) from carrying over, contaminating, and negatively influencing the downstream experiments.

The PureLink PCR Purification Kit is based on the selective binding of dsDNA to silica-based membrane in the presence of chaotropic salts. Simply mix the PCR product with Binding Buffer and apply to the PureLink Spin Column. The dsDNA binds to the silica-based membrane in the column. Remove impurities by thorough washing with Wash Buffer. To purify the DNA, elute the dsDNA in low-salt Elution Buffer or water.The purified PCR product is suitable for automated fluorescent DNA sequencing, restriction enzyme digestion, and cloning.

How It Works

ChargeSwitch® technology employs a completely novel mechanism that replaces harsh reagents such as chaotropic salts, ethanol, or organic solvents and isopropanol precipitation with a simple ion-exchange procedure. In this method, DNA binds to a surface ligand that is positively charged at low pH, and is eluted when the pH is raised to 8.5 (Figure 1). The ChargeSwitch®-Pro protocol retains the ease and familiarity of common silica spin-column protocols with binding, washing, and elution steps (Figure 2).

Better Downstream Results

Since ChargeSwitch® technology does not require the use of ethanol, chaotropic salts, or organic solvents for nucleic acid purification, the resulting DNA is free of enzymatic inhibitors and thus shows better downstream performance (Figure 3). These compounds are known to inhibit many enzymatic reactions such as PCR, cloning, restriction enzyme digestion, and array CGH labeling. Expect to see reduced variability in your sequencing applications, with reads in excess of 800 bp with Phred scores >20 (Figure 4); clean restriction enzyme digestions (Figure 5); and removal of dNTPs and primers in excess of 98% (Figure 6).

DNAclear™ Purification Kit (Invitrogen™)

The Ambion® DNAclear™ kit is designed to purify double-stranded DNA (dsDNA) generated by enzymatic reactions such as cDNA synthesis and PCR. Includes sufficient reagents for 20 purifications.

The DNAclear™ cDNA Purification Kit process is simple and fast. DNA amounts ranging from 1 ng to 3 µg can be efficiently purified and concentrated. The DNAclear™ procedure consists of three steps: 1) DNA is bound to the Microfilter Cartridge in the presence of the DNA Binding Buffer, 2) the DNA is washed with the cDNA Wash Buffer, and 3) the DNA is eluted with preheated nuclease-free water. The DNAclear™ Kit can be used to remove free nucleotides, oligonucleotides, proteins, and salts from dsDNA samples. The DNA that is recovered from the Microfilter Cartridge is ready for any application that requires highly pure DNA including in vitro transcription reactions.
