GeneJET Gel Extraction and DNA Cleanup Micro Kit (Thermo Scientific™)

Thermo Scientific GeneJET Gel Extraction and DNA Cleanup Micro Kit is developed as 3 in 1 kit designed for rapid and efficient purification of DNA from PCR, enzymatic reaction mixtures, and DNA extraction from standard or low-melting point agarose gels run in either Tris acetate (TAE) or Tris borate (TBE) buffer. The kit combines the convenience of spin column technology with the selective binding properties of a silica membrane, eliminating the need for tedious resin manipulations or toxic phenol-chloroform extractions.

The GeneJET Gel Extraction and DNA Cleanup Micro Kit effectively removes primers, primer dimers, dNTPs, unincorporated labeled nucleotides, enzymes and salts from PCR and other reaction mixtures. The kit can be used for purification of DNA fragments from 100 bp to 20 kb. The recovery rates are 90 to 100% for 100 bp to 3 kb DNA fragment size range. Each GeneJET DNA Purification Column has a total binding capacity of up to 10 µg of DNA, and the entire procedure takes approximately 3.5 minutes for DNA cleanup

S.N.A.P.™ UV-Free Gel Purification Kit (Invitrogen™)

The S.N.A.P.™ UV-Free Gel Purification Kit uses non-toxic reagents to rapidly purify intact DNA fragments from agarose gels. The key to isolating undamaged DNA is the unique crystal violet reagent, which allows visualization of DNA (>=200 ng) in the gel under ambient light. This eliminates the need for ethidium bromide and exposure to UV light, which can nick and damage DNA. Nucleic acids up to 7 kb are efficiently purified from the gel using the convenient S.N.A.P.™ resin, provided ready to use in a spin column format. The resin eliminates organic extractions, which generate hazardous waste. In addition, there are no time-consuming ethanol precipitations.

Easy to Use
The S.N.A.P.™ UV-Free Gel Purification Kit yields pure, intact DNA in an easy-to-perform, rapid procedure. Simply:
1. Prepare gel with crystal violet reagent and load samples in crystal violet loading buffer.
2. Visualize DNA band under ambient light and excise from agarose gel.
3. Add NaI solution (provided) and melt the agarose at 42-50°C.
4. Load sample onto the S.N.A.P.™ spin column.
5. Wash away contaminants.
6. Elute DNA in water.

The eluted DNA is highly pure, intact, and ready to use for cloning, sequencing, transformation, or transfection.

GeneJET Gel Extraction Kit (Thermo Scientific™)

Thermo Scientific GeneJET Gel Extraction Kit is designed for rapid and efficient purification of DNA fragments from standard or low-melting point agarose gels run in either TAE or TBE buffer.

The kit utilizes a proprietary silica-based membrane technology in the form of a convenient spin column. The kit can be used to purify DNA fragments from 25 bp to 20 kb in size with recovery rates up to 95%. Each GeneJET purification column has a binding capacity of up to 25 µg of DNA and can process up to 1 g of agarose gel. The entire procedure takes just 15 minutes, and the isolated DNA is ready to use in all common downstream applications, including ligation, restriction digestion, PCR, sequencing, and labeling.

The PureLink® Quick Gel Extraction Kit allows you to rapidly and efficiently purify DNA fragments from TAE or TBE agarose gels of various percentages. DNA can be extracted and purified from agarose gels with different melting points in ~30 minutes using PureLink® silica membrane-based quick gel extraction columns. For your convenience, purification protocols are provided for centrifugation and for vacuum.

The PureLink™ Quick Gel Extraction and PCR Purification Combo Kit is designed to purify DNA fragments from agarose gels. The simple procedure uses a silica-based spin cartridge to purify DNA fragments from 40 bp to 10 kb in < 30 minutes. Silica Bead DNA Gel Extraction Kit (Thermo Scientific™)

Thermo Scientific Silica Bead DNA Gel Extraction Kit is a simple and efficient system for DNA extraction from agarose gels and reaction mixtures. The kit utilizes the modified protocol of Vogelstein and Gillespie, employing solubilization of the agarose gel and selective adsorption of nucleic acids on specially prepared silica particles at high salt concentration. Silica particles bound with DNA are washed to remove contaminants and pure DNA is eluted with TE buffer or water.

This method requires few manipulations, and is both faster and easier to perform than other organic-based extraction methods. Compared to column based gel-extraction kits, the DNA Gel Extraction Kit offers flexibility for scaling reaction and elution volumes up or down. The purified DNA is suitable for all common molecular biology procedures, including restriction digestion, cloning, sequencing, etc.

Care should be taken to avoid DNA damage with UV light when the gel-purified DNA fragment will be used in downstream cloning reactions. Always use a long wavelength UV (360 nm) light-box during excision of the gel slice. If only a short-wavelength UV (254 to 312 nm) light-box is available, minimize the UV exposure to a few seconds or keep the gel on a glass or plastic plate. Alternatively, visible dyes can be included in standard agarose gels to allow for visualization of the DNA bands in ambient light (2, 3).
