For sensitive, highly predictable and consistent extraction and isolation of viral DNA and RNA from human serum/plasma samples, following a simple separation protocol. The 1 µm Dynabeads have an optimized silica-like surface chemistry, and a high specific surface area, providing efficient kinetics and a high sensitivity. A higher sensitivity and improved performance is achieved with Dynabeads® compared to magnetic particles from alternative suppliers. PureLink™ Pro 96 Viral RNA/DNA Purification Kit (Invitrogen™)

The PureLink® Pro 96 Viral RNA⁄DNA Purification Kit is the only 96-well silica plate system that can extract both viral RNA and DNA from 200-µl cell-free fluids which uses a low-depth 96-well centrifuge bucket for ease-of- use, all without any loss in sensitivity. These kits provide:

The PureLink® Pro 96 Viral RNA⁄DNA Purification Kit is based on conventional silica plate extraction chemistry. Cell-free samples are lysed in guanidine containing buffers that have been reformulated to increase the recovery and sensitivity of purified nucleic acids, while carrier RNA is used to protect samples from RNase degradation. Binding of lysates to the Viral Filter Plate can be performed with either a vacuum manifold or centrifugation using a 96-well rotor with buckets having a depth of only 5 cm. After washes with ethanol-based buffers, the viral RNA or DNA is eluted in 100 µl of RNase-free water. The RNA or DNA is ready to use in one-step or two-step RT-PCR and qRT-PCR, qPCR, or other enzymatic amplification procedures.

The PureLink® Pro 96 Viral RNA⁄DNA Purification Kit shows excellent well-to-well consistency
A critical requirement for any 96-well purification system is well-to-well consistency. When using a technique as sensitive as qRT-PCR for viral nucleic acid detection, which may exist as low viral titers from patient samples, consistency between samples becomes even more important. When well-to-well recovery of lentiviral RNA was analyzed using the PureLink® Pro 96 Viral RNA⁄DNA Purification Kit, consistent Ct values were obtained in qRT-PCR analyses while plasma controls run on the same plate were negative, showing no cross contamination (Figure 2).

Get the same results in a 96-plate extraction as you would with a spin column
To prove consistency of our novel purification chemistry in the both high- and low-throughout formats, Armored RNA® HCV virus was spiked in human plasma and 200 µl were used for viral nucleic acid purification with either the PureLink® Viral RNA⁄DNA Mini Kit (spin-column based purification) or with the PureLink® Pro 96 Viral RNA⁄DNA Purification Kit. The purified samples were analyzed for performance comparison using qRT-PCR. Both purification methods successfully demonstrated viral purification and detection as both purification profiles exhibited scalable qRT-PCR detection analysis profiles (Figure 3). The PureLink® Pro 96 Viral RNA⁄DNA Purification Kit provides sensitive linear scalable viral RNA⁄DNA purification with a sensitivity and efficiency equal to that of the spin column purification format.

Improved plate design
The PureLink® Pro Viral RNA⁄DNA purification kit provides optimized semi-skirted 96-well plates (Figure 4) for improved performance and results. In addition, the plate nozzles with annular collar prevent cross-contamination of samples and improve drying of the silica membrane to prevent ethanol carryover. In addition, sample processing can be performed manually using a benchtop vacuum manifold, by centrifugation (at 2250 x g), or with an automated liquid handling platform. The semi-skirted plate design is compatible with most vacuum manifolds on robotic workstations.

Get the Same Amount of RNA from 10-fold Less Sample
The sensitivity of viral RNA detection was compared using RNA isolated with Applied Biosystems® MagMAX™ protocol or the current National Veterinary Services Laboratories (NVSL) validated viral testing protocol. Even at one-tenth the sample input, viral RNA isolated using the MagMAX™ kit could be detected at equal or greater levels than RNA isolated using the NVSL protocol and recommended competitor's kit (see data). MagMAX™ Viral/Pathogen Ultra Nucleic Acid Isolation Kit (Applied Biosystems™)

The MagMAX Viral/Pathogen Ultra Nucleic Acid Isolation Kit contains the same reagents as the standard MagMAX Viral/Pathogen Nucleic Acid Isolation Kit (Cat. No. A42352) and so, like that kit, recovers nucleic acid from viral particles and gram negative bacteria. However, the MagMAX Viral/Pathogen Ultra kit also includes a proprietary enzyme mix that enables lysis and nucleic acid recovery from the toughest pathogens, such as gram-positive bacteria, yeast, and fungi. With this enzyme mix, used in conjunction with optimized reagents, the MagMAX Viral/Pathogen Ultra kit makes it possible to achieve comprehensive pathogen lysis and recovery of RNA and DNA.

The improved version of the MagMAX™ Viral RNA Isolation Kit provides better viral particle lysis and more effective recovery of both viral RNA and DNA from a variety of cell-free biological samples, including plasma, sera, and swabs. The results of these changes are improved yields and reproducibility.

The MagMAX™ Viral RNA Isolation Kit can efficiently isolate viral RNA and DNA from samples as large as 400 µl. RNA and DNA recovery is typically greater than 75%, but may vary depending on sample type. The RNA recovered with the kit is of high quality and purity and is suitable for real-time RT-PCR.

Thermo Scientific GeneJET Viral DNA and RNA Purification Kit is designed for rapid and efficient purification of high quality viral nucleic acids from various human and mammalian liquid samples, and is compatible with common sample preservatives like EDTA, citrate, and heparin.

The kit utilizes a silica-based membrane technology in the form of a convenient spin column. Viral nucleic acids from lysed samples bind to the column membrane while impurities are effectively removed during subsequent washing and centrifugation steps. Ready-to-use nucleic acids are eluted from the column.
