mirVana™ qRT-PCR miRNA Detection Kit (Invitrogen™)

The mirVana™ qRT-PCR Detection Kit has been optimized to amplify specific small RNA using qRT-PCR primer sets (not included), enabling accurate quantitation of miRNA expression levels. The novel detection system can be used to characterize miRNA expression via traditional endpoint RT-PCR or real-time RT-PCR using SYBR® Green I.

• Quantitative results in less than 3 hours
• Detection from as little as 25 pg of total RNA
• Suitable for either real-time or end-point RT-PCR
• Ideal for validating expression profiling data and high-throughput applications.

MicroRNAs (miRNAs) are evolutionarily conserved, small, non-coding RNA molecules that are estimated to regulate at least 30% of all protein encoding genes. Currently more than 2,900 miRNAs have been annotated from a wide range of organisms including plants, worms, flies, mammals, and viruses. To date, miRNAs have been implicated in development, differentiation, apoptosis, fat metabolism, viral infection, and cancer.

Due to their small size (~22 nt), miRNAs cannot be analyzed by traditional PCR approaches. Designed for use in conjunction with qRT-PCR Primer Sets (not included), the mirVana™ qRT-PCR Detection Kit has been optimized to amplify specific small RNAs, enabling accurate quantitation of miRNA expression levels (Figure 1). This novel detection system can be used to characterize miRNA expression via traditional endpoint RT-PCR or real-time RT-PCR using SYBR® Green I.

Since miRNAs represent only a minute fraction of total RNA, detection of these molecules can be difficult. However, the mirVana™ qRT-PCR miRNA Detection Kit is extremely sensitive, detecting miRNA from as little as 25 pg of total RNA. The kit also specifically detects mature miRNAs and not miRNA precursors (Figure 2). Furthermore, the mirVana™ qRT-PCR miRNA Detection Kit exhibits linearity over at least 5 logs (Figure 3) and can be used with total RNA, miRNA-enriched RNA fractions, or even degraded RNA samples.

Each kit contains sufficient reagents for 200 reactions, as well as a control primer set to amplify the ubiquitously expressed miR-24 and human heart total RNA. Note: Thermostable DNA polymerase is not supplied with the kit. For analysis of individual miRNAs, the mirVana™ qRT-PCR miRNA Detection Kit is unparalleled in sensitivity and speed!

CTS™ PureQuant™ Treg Assay (Applied Biosystems™)

The Applied Biosystems CTS PureQuant assays are a family of assays that accurately identify and quantify specific immune cell types in a mixed population of cells. These assays are TaqMan qPCR-based and measure genomic DNA methylation at specific sites that serve as a unique identifier. The assays include standards and controls and each 96-well plate can accommodate up to seven test samples. Results are provided as a percentage of target cell type out of total input cells using a free analysis tool.

The CTS PureQuant Treg Assay utilizes a specific demethylated site in the transcription factor FOXP3 gene to positively identify regulatory T-cells. This specific site allows for discrimination of regulatory T-cells from activated CD4+ FOXP3+ conventional T-cells (see https://onlinelibrary.wiley.com/doi/full/10.1002/eji.200737594).

Benefits include:
Proven performance―core technology has been used for identification and quantification of immune cell types for over a decade
Easily standardized―qPCR method enables consistent results across multiple users, instruments, and sites
Facilitates regulatory filings―regulatory support file provided upon request; established performance specifications following ICHQ2(R1) guidelines
Scheduling flexibility―assay can be performed using fresh or frozen cells/gDNA

The assays include all reagents, standards, and controls required to run the assay. Specific components include:
Calibrator: corrects for variation in bisulfite conversion efficiency. Plasmid-based calibrator harbors the genomic target regions in equimolar ratios and includes GAPDH. Subjected to bisulfite conversion followed by copy number estimation via qPCR standard curve. Allows estimation of assay-specific bisulfite efficiency factor: the ratio between TpG/GAPDH copy numbers measured from the same plasmid.
Reference: checks assay performance. Consists of genomic DNA extracted from human blood of pooled healthy donors. The reference values represent the expected range of the target cell type. For example, CD8+ cells are generally more abundant than Treg or Th17 cells.
Standards: determine the unknown copy number. Consist of six dilutions spanning 4-log linear dynamic range (30-31250 copies). Absolute copy numbers can reliably be determined using the slope and intercept of the respective standard curves.

This product is manufactured at a site with an ISO9001-certified quality management system.

Efficiency across a broad range of input concentrations
Maximum flexibility in RNA template input quantity requires optimal real-time PCR efficiency across a broad range of template quantities. Figure 1 shows the excellent amplification efficiency across six orders of magnitude. The result is comparable to that of the TaqMan RNA-to-CT 2-Step Kit, and is a significant improvement in dynamic range and sensitivity over the TaqMan One-Step RT-PCR Master Mix Reagents.

High sensitivity at low target concentration
The sensitivity of TaqMan RNA-to-CT 1-Step Kit was validated using a synthetic RNA template for which copy number is precisely known. Significant sampling error occurs when measuring low quantities of target, so statistical analysis is required for proper evaluation using multiple replicates. Figure 2 shows the expected quantity of target and corresponding mean CT values. Statistical analysis indicates high confidence of sample quantification based on a T-test (Table I), consistent with as few as 10 copies of target*.

*Results are dependent on a variety of factors, including assay design.
