TaqMan™ PreAmp Master Mix (Applied Biosystems™)

TaqMan® PreAmp Master Mix pre-amplifies small amounts of cDNA without introducing amplification bias to the sample. Preamplification enables you to stretch your precious sample into many more real-time PCR reactions.

TaqMan® PreAmp Master Mix lets you:

• Analyze mRNA from precious samples such as laser capture microdissections, needle biopsies, and formalin-fixed paraffin-embedded (FFPE) tissues
• Stretch as little as 1 ng of cDNA into 200 real-time PCR reactions for gene expression analysis using TaqMan® Gene Expression assays
• Analyze up to 100 gene expression targets with minimal hands-on time

Preserves Equilibrium of Targets Without Preamplification Bias
In the past, preamplification kits and techniques often resulted in uneven amplification of targets and incorrect or biased data (as shown by low correlation coefficients between preamplified and unamplified samples). TaqMan® PreAmp Master Mix amplifies with no bias and provides extremely high correlation between amplified and unamplified cDNA.

Easy Workflow
With TaqMan® PreAmp Master Mix, preamplification setup takes only a few minutes. After a one-time pooling of the TaqMan® assays , preamplification setup (mixing the pooled assays with cDNA sample and TaqMan® PreAmp Master Mix) takes only five additional minutes. Cycling takes 1.5 hours using a GeneAmp® 9700 thermal cycler.

CTS™ PureQuant™ Th17 Assay (Applied Biosystems™)

The Applied Biosystems CTS PureQuant assays are a family of assays that accurately identify and quantify specific immune cell types in a mixed population of cells. These assays are TaqMan qPCR-based and measure genomic DNA methylation at specific sites that serve as a unique identifier. The assays include standards and controls and each 96-well plate can accommodate up to seven test samples. Results are provided as a percentage of target cell type out of total input cells using a free analysis tool.

CTS PureQuant Th17 Assay utilizes a specific demethylated site in the pro-inflammatory cytokine interleukin 17A (IL17). This cytokine is exclusively produced by T helper 17 cells.

Benefits include:
Proven performance―core technology has been used for identification and quantification of immune cell types for over a decade
Easily standardized―qPCR method enables consistent results across multiple users, instruments, and sites
Facilitates regulatory filings―regulatory support file provided upon request; established performance specifications following ICHQ2(R1) guidelines
Scheduling flexibility―assay can be performed using fresh or frozen cells/gDNA

The assays include all reagents, standards, and controls required to run the assay. Specific components include:
Calibrator: corrects for variation in bisulfite conversion efficiency. Plasmid-based calibrator harbors the genomic target regions in equimolar ratios and includes GAPDH. Subjected to bisulfite conversion followed by copy number estimation via qPCR standard curve. Allows estimation of assay-specific bisulfite efficiency factor: the ratio between TpG/GAPDH copy numbers measured from the same plasmid.
Reference: checks assay performance. Consists of genomic DNA extracted from human blood of pooled healthy donors. The reference values represent the expected range of the target cell type. For example, CD8+ cells are generally more abundant than Treg or Th17 cells.
Standards: determine the unknown copy number. Consist of six dilutions spanning 4-log linear dynamic range (30-31250 copies). Absolute copy numbers can reliably be determined using the slope and intercept of the respective standard curves.

This product is manufactured at a site with an ISO9001-certified quality management system.

SYBR™ Green PCR Master Mix (Applied Biosystems™)

Everything you need for SYBR® Green dye–based PCR amplification and detection in a convenient, single-tube format. Applied Biosystems® SYBR® Green PCR Master Mix combines SYBR® Green I dye, AmpliTaq Gold® DNA Polymerase, dNTPs with dUTP, Passive Reference 1, and optimized buffer in the convenience of a single vial.

• Premixed components stored at 2–8°C significantly reduce assay setup time
• SYBR® Green I dye detects double-stranded DNA, so specific probes are not required
• AmpliTaq Gold® DNA polymerase minimizes nonspecific product formation to achieve superior performance
• dUTP significantly reduces carryover contamination when used in conjunction with uracil-DNA glycosylase
• Proprietary buffer enhancements ensure performance and reliability

Maximum Flexibility and Convenience
Applied Biosystems® SYBR® Green PCR Master Mix provides maximum flexibility at reduced cost because no target-specific TaqMan® probes are required. SYBR® Green I dye is a double-stranded DNA binding dye that detects any double-stranded DNA generated during PCR. The hot-start enzyme AmpliTaq Gold® DNA Polymerase minimizes nonspecific product formation (including primer-dimers), yielding superior performance and sensitivity. Passive Internal Reference 1 is provided to normalize non-PCR–related fluorescence fluctuations. This minimizes well-to-well variability that can result from a variety of causes, such as pipetting error or sample evaporation. SYBR® Green I dye is ideal for target identification (screening assays) or when a limited number of assays is needed.

• Broad instrument compatibility

Optimized for Multiplexing
Confidence is paramount when it comes to your results. TaqMan® Multiplex Master Mix is designed to deliver accurate results for multplex reactions and ensure excellent concordance of results in both singleplex and multiplex reactions.

Best-in-class Performance
TaqMan® Multiplex Master Mix is designed for performance equal to or better than other multiplex master mixes on the market. TaqMan® Multiplex Master Mix has been benchmarked against the leading suppliers' multiplex master mixes to help ensure that it succeeds in providing best-in-class sensitivity, accuracy, dynamic range, and specificity.

Reduced Run Times on Standard Instrumentation
TaqMan® Multiplex Master Mix has been optimized for use with both fast and standard instrumentation, enabling researchers who currently own standard instruments to also reap the performance benefits and time savings this mix provides.

Carry-over Contamination Control
TaqMan® Multiplex Master Mix contains heat-labile uracil-DNA glycosylase (UDG) for worry-free carryover contamination control. The dUTP in the mix ensures that any amplified DNA will contain uracil, while UDG removes uracil residues from single- or double-stranded DNA, preventing dU-containing DNA from serving as template in future PCRs. A UDG incubation step before PCR cycling destroys any contaminating dU-containing product from previous reactions. UDG is then inactivated by the high temperatures during normal PCR cycling, thereby allowing the amplification of genuine target sequences.

Instrument Compatibility
TaqMan® Multiplex Master Mix can be used in fast cycling mode on both standard and fast qPCR instruments, such as: 7500 Real-Time PCR System, 7500 Fast Real-Time PCR System, ViiA™ 7 Real-Time PCR System, and QuantStudio™ 6 Flex, 7 Flex, and 12K Flex Real-Time PCR systems.
