Coating Matrix Kit Protein (Gibco™)

A convenient combination of Coating Matrix and Dilution Medium for treatment of tissue culture labware for keratinocyte culture. Contains enough material to coat approximately thirty T-25 flasks. Use of the Coating Matrix significantly enhances the attachment, growth, and population doubling potential of human keratinocytes, especially when used in conjunction with Supplement S7 (S-017-5) or EDGS (S-012-5).

Fibronectin Bovine Protein, Plasma (Gibco™)

Fibronectin is a large, cell surface and plasma protein that exhibits structural and adhesive properties in cell-associated fibrillar matrices. Fibronectin is one of the primary cell adhesion molecules. Bovine Plasma Fibronectin is purified from plasma by affinity chromatography on gelatin-Sepharose and further purified by ion exchange chromatography to remove glycosaminoglycans. Bovine Fibronectin is filtered (0,2 µm) and supplied lyophilized.

Vitronectin (VTN-N) Recombinant Human Protein, Truncated (Gibco™)

Vitronectin (VTN-N) is a recombinant human protein that provides a defined surface for feeder-free culture of human pluripotent stem cells (PSCs). When used with Essential 8™ Medium (A1517001), vitronectin (VTN-N) has been proven to maintain pluripotency and normal growth characteristics in multiple PSC lines. In addition, vitronectin (VTN-N) has been shown to support PSC growth for >50 passages without any signs of karyotypic abnormalities and to maintain the ability of PSCs to differentiate into all three germ line lineages. As published by Guokai Chen et al. (1) in the laboratory of James Thomson, the VTN-N variant of vitronectin supports human pluripotent stem cell attachment and survival better than wild-type vitronectin when used in conjunction with Essential 8™ Medium.

Optimized and Defined Surface
Unlike standard basement membrane extracts (BMEs), vitronectin (VTN-N) was designed in the laboratory of James Thomson for use with Essential 8™ Medium and has been shown to support human pluripotent stem cell attachment and survival better than wild type vitronectin. Vitronectin (VTN-N) provides an optimal environment for feeder-free pluripotent stem cell culture.

Reduce Variability
Vitronectin (VTN-N) is a defined, recombinant human protein that reduces variability in your PSC cultures compared to human-plasma derived vitronectin and standard basement membrane extracts (BMEs).

Attachment Factor Protein (1X) (Gibco™)

A sterile solution (1X) containing gelatin as an attachment factor. When used to coat culture surfaces, AF enhances the growth of microvascular endothelial cells.

Collagen I Rat Protein, Tail (Gibco™)

Collagen I, Rat Tail is the most common fibrillar collagen (90%) found in skin, bone, tendons, and other connective tissues. It is supplied at 3 mg/ml in a 20 ml vial providing the flexibility to dilute to lower concentrations. The detailed gelling procedure forms a firm, clear gel for 3D applications.

Laminin Mouse Protein, Natural (Gibco™)

Laminin is the major glycoprotein component of basement membranes, and has a molecular weight of 850,000 daltons. This preparation of mouse Laminin was isolated from the Engelbreth-Holm-Swarm (EHS) sarcoma and is supplied at a concentration of 0.5-2.0 mg⁄ml.

Fibronectin is a large, cell surface and plasma protein that exhibits structural and adhesive properties in cell-associated fibrillar matrices. Fibronectin is one of the primary cell adhesion molecules. Human Plasma Fibronectin is isolated from human plasma by affinity chromtography. Fibronectin has a molecular weight of 440,000 and is made up of two 220,000 dalton polypeptides. Human Fibronectin is filtered (0,2UI) and supplied lyophilized.

Performance and Quality Testing
The purity is determined by SDS-PAGE. Bovine and human fibronectins are assayed for their ability to support cell attachment and spreading, and screened to confirm absence of mycoplasma or bacterial contamination.
