BSA, acetylated (20 mg/mL) (Invitrogen™)

Ambion® Acetylated BSA is used as a carrier protein and as an enzyme stabilizer in which the absence of contaminants such as nucleases and proteases is essential. Six tubes of 25 mg are provided, at a concentration of 20 mg/mL. The acetylation process inactivates any trace nuclease activity that might be present. While the acetylation process does change BSA's binding characteristics and makes it inhibitory in PCR reactions, it is routinely added to restriction digests. The molecular weight is ~66 kD. Acetylated BSA is rigorously tested for contaminating nonspecific endonuclease, exonuclease, protease, and RNase activity.

UltraPure™ BSA (50 mg/mL) (Invitrogen™)

UltraPure BSA is a non-acetylated bovine serum albumin (BSA) pure enough to use when DNA or RNA integrity is critical. One tube containing 50 mg is provided, at a concentration of 50 mg/mL. BSA has many uses as a carrier protein and as a stabilizing agent in enzymatic reactions. In northern, Southern, and dot blot hybridizations, BSA is also used as a blocking agent. It is recommended in buffers for nick translation, polymerase reactions, and ligations. BSA is also a common additive for PCR amplifications, footprinting, and gel shift assays. In restriction digests, BSA has been shown to enhance enzyme activity. The molecular weight is 68 kD. UltraPure BSA is rigorously tested for DNase, RNase, endonuclease, and protease activity.

Pierce™ 3x DYKDDDDK Peptide (Thermo Scientific™)

Pierce 3x DYKDDDDK Peptide is a lyophilized peptide with the sequence Met-Asp-Tyr-Lys-Asp-His-Asp-Gly-Asp-Tyr-Lys-Asp-His-Asp-Ile-Asp-Tyr-Lys-Asp-Asp-Asp-Asp-Lys (MDYKDHDGDYKDHDIDYKDDDDK). It has a molecular weight of 2861.9 g/mol. For use, reconstitute to 1.5 mg/mL in PBS.

This tricosa-peptide can be used to competitively elute DYKDDDDK-tagged fusion proteins from immobilized anti-DYKDDDDK affinity supports, especially when a DYKDDDDK-tagged protein is sensitive to low pH. Note that Pierce 3x DYKDDDDK Peptide may interfere with protein determination assays and absorbance at 280 nm. However, the peptide can be removed from the eluate by desalting (e.g., with Zeba Spin Desalting Columns, Cat. No. 89891).

HA Synthetic Peptide (Thermo Scientific™)

Lyophilized peptide with the sequence N-Tyr-Pro-Tyr-Asp-Val-Pro-Asp-Tyr-Ala-C. Molecular weight: 1102.2 g/mol

26184 can be used to competitively elute HA-tagged fusion proteins from immobilized anti-HA affinity resin. Use 4 X 1 column volumes of 1 mg/ml HA peptide solution to elute HA-tagged proteins. Elution yields will increase by incubating the peptide on the column for 10-15 minutes at 30-37ºC for each elution fraction.

Reconstitute to 1 mg/ml using TBS or distilled water.

The HA peptide, NH2-YPYDVPDYA-COOH, is a synthetic peptide used to competitively elute fusion proteins containing the influenza hemagglutinin (HA) tag bound to immobilized anti-HA affinity resins.

GFP Recombinant Aequorea victoria Protein, N-His Tag

Green Fluorescent Protein (GFP) recombinant Aequorea victoria protein is supplied as a lyophilized powder. In general, recombinant proteins can be used as protein structure analysis and in cell biology research applications.

This recombinant protein is expressed from a DNA sequence encoding the Aequorea victoria GFP (AAB65663) (Ser 2-Lys 238) fused to a polyhistidine tag at the N-terminus.

N-terminal Sequence Analysis: Glu 99.

1. Mesured by emission analysis.
2. Measure excitation and emission spectra of GFP at wavelengths of 395 nm and 510 nm respectively.

Formulation: Lyophilized from sterile PBS, pH 7.5.

Reconstitution: Dissolve the protein in sterile double-distilled water to a concentration of 0.2 mg/ml or lower. It is recommended that the protein be aliquoted and used as soon as possible. Store aliquots under sterile conditions at -20°C. Avoid repeated freeze-thaw cycles.

The green fluorescent protein (GFP) is a protein that exhibit bright green fluorescence when exposed to blue light. The green-fluorescent protein (GFP) of the jellyfish Aequorea victoria has long been used as a universal reporter in a broad range of heterologous living cells and organisms. Poly (Adp-Ribose) Polymerase 1 (PARP1), recombinant human protein is supplied in frozen format. In general, recombinant proteins can also be used as an immunogen, as a protein standard, or in cell biology research applications.

This recombinant protein was expressed from The amino acids corresponding to the full length of human PARP1 (NP_001609.2) (Met 1-Trp 1014) fused to a polyhistidine tag at the C-terminus.

Activity: Measured by its binding ability in a functional ELISA. Immobilized human PARP1 at 10 µg/ml (100 µl/well) can bind to biotinylated human HSP70. The EC50 of biotinylated human HSP70 is 0.035 µg/ml.

Formulation: 50 mM Tris, 100 mM NaCl, pH 8, 2 mM EDTA, 2 mM PMSF, 0.5 mM TCEP, 10% glycerol.

Reconstitution: No reconstitution needed. Store at -80°C. Avoid repeated freeze-thaw cycles.
