Frizzled 4 Recombinant Human Protein (Gibco™)

The equilibrium binding constant, KD, of Human Frizzled 7 was measured at less than 10nM using mouse Wnt3a as the ligand. Measurements were made using the ForteBio Octet optical biosensor instrument.

UltraPure™ BSA (50 mg/mL) (Invitrogen™)

UltraPure BSA is a non-acetylated bovine serum albumin (BSA) pure enough to use when DNA or RNA integrity is critical. One tube containing 50 mg is provided, at a concentration of 50 mg/mL. BSA has many uses as a carrier protein and as a stabilizing agent in enzymatic reactions. In northern, Southern, and dot blot hybridizations, BSA is also used as a blocking agent. It is recommended in buffers for nick translation, polymerase reactions, and ligations. BSA is also a common additive for PCR amplifications, footprinting, and gel shift assays. In restriction digests, BSA has been shown to enhance enzyme activity. The molecular weight is 68 kD. UltraPure BSA is rigorously tested for DNase, RNase, endonuclease, and protease activity.

Pierce 3x DYKDDDDK Peptide is a lyophilized peptide with the sequence Met-Asp-Tyr-Lys-Asp-His-Asp-Gly-Asp-Tyr-Lys-Asp-His-Asp-Ile-Asp-Tyr-Lys-Asp-Asp-Asp-Asp-Lys (MDYKDHDGDYKDHDIDYKDDDDK). It has a molecular weight of 2861.9 g/mol. For use, reconstitute to 1.5 mg/mL in PBS.

This tricosa-peptide can be used to competitively elute DYKDDDDK-tagged fusion proteins from immobilized anti-DYKDDDDK affinity supports, especially when a DYKDDDDK-tagged protein is sensitive to low pH. Note that Pierce 3x DYKDDDDK Peptide may interfere with protein determination assays and absorbance at 280 nm. However, the peptide can be removed from the eluate by desalting (e.g., with Zeba Spin Desalting Columns, Cat. No. 89891).

S100B Recombinant Human Protein, N-His Tag

Beta-Lactamase (TEM-1) Protein

TEM-1 beta-lactamase is the most widespread enzyme to confer beta-lactam antibiotic resistance to gram-negative bacteria. It is a monomeric protein that requires no metals or cofactors for activity. TEM-1 beta-lactamase efficiently hydrolyzes most penicillin and cephalosporin substrates, including nitrocefin and fluorescent derivates. This protein is intended to be used as a positive control for enzyme-dependent substrate hydrolysis and for use on immunoblots. Other uses include kinetic studies with novel substrates or identification of inhibitors of beta-lactamase as part of an antimicrobial program.

DLL4 Recombinant Human Protein, hIgG1-Fc Tag

Delta-like 4 (Drosophila) (DLL4) recombinant human protein is supplied as a lyophilized powder. Background
Delta-like protein 4 (DLL4) is a type I membrane-bound Notch ligandcharacterized by an extracellular region containing several EGF-likedomains and a DSL domain required for receptor binding, and issuggested to play a key role in vascular development and tumorangiogenesis. The Notch pathway is an evolutionary conservedintercellular signaling pathway involved in numerous biological processesincluding cell fate determination, cellular differentiation, proliferation, survival, and apoptosis. In mammalian cells, five Notch ligands (Jagged1, 2, DLL1, 3, 4) and four Notch receptors (Notch1-4) have been identified, and ligand-receptor interactions results in proteolysis and translocation of the Notch intracellular domain. As a ligand for Notch1 and Notch 4, DLL4 is selectively expressed in the developing endothelium and in some tumor endothelium, and is induced by vascular endothelial growth factor (VEGF)-A and hypoxia. In recent studies, it has been revealed that this DLL4 inhibition may paradoxically lead to increased angiogenesis but poor tumor growth because of the non-functional neovascularization, and constitutive expression of DLL4 may leads to a lethal lymphoproliferative disease. Accordingly, this may provide a new therapeutic approach for certain carcinomas.

Background
The green fluorescent protein (GFP) is a protein that exhibit bright green fluorescence when exposed to blue light. Noggin Recombinant Human Protein (Gibco™)

Recombinant Human Noggin is a bioactive protein intended for use in cell culture applications. Noggin binds and inactivates members of the transforming growth factor-beta (TGF-β) superfamily signaling proteins, such as bone morphogenetic protein-4 (BMP4). By diffusing through extracellular matrices more efficiently than members of the TGF-β superfamily, this protein may have a principal role in creating morphogenic gradients. The protein appears to have pleiotropic effect, both early in development as well as in later stages. It was originally isolated from Xenopus based on its ability to restore normal dorsal-ventral body axis in embryos that had been artificially ventralized by UV treatment. The results of the mouse knockout of the ortholog suggest that it is involved in numerous developmental processes, such as neural tube fusion and joint formation.

Bovine Serum Albumin (Invitrogen™)

Bovine Serum Albumin is suitable for use as an enzyme stabilizer during purification or for dilution of restriction endonucleases and nucleic acid modifying enzymes. BSA is also commonly used in DNA and protein labeling experiments as a blocking agent to minimize background. This BSA is acetylated to inactivate contaminating nucleases and proteases.

Performance and quality testing
Purity is determined by SDS-PAGE. No detectable contaminating activity is observed in 3´ and 5´ exonuclease, DNA nicking, and restriction endonuclease inhibition assays.

RSPO1 Recombinant Human Protein (aa 1-146, His Tag)

R-Spondin 1 (RSPO1), recombinant human protein is supplied as a lyophilized powder. It is suitable for use in protein studies such as protein structure analysis and protein-protein interactions. In general, recombinant proteins can also be used as an immunogen, as a protein standard, or in cell biology research applications.

This recombinant protein was expressed from a DNA sequence encoding the human RSPO1 (NP_001033722.1) (Met 1-Ala 146) fused to a polyhistidine tag at the C-terminus.

Activity: Measured by its ability to bind recombinant mouse CD36 in a functional ELISA.

Formulation: Lyophilized in 140 mM NaCl, 2.7 mM KCl, 10 mM Na2HPO4, 1.8 mM KH2PO4, pH7.4, 5% Mannitol, 5% Trehalose, 0.02% Tween80.

Reconstitution: Dissolve the protein in sterile double distilled water to a concentration of 0.2 mg/ml or lower. It is recommended that the protein be aliquoted and be used as soon as possible. Store aliquots under sterile conditions at -20°C. Avoid repeated freeze-thaw cycles.

Expiration Date: Expires one year from date of receipt when stored as instructed.

HA Synthetic Peptide (Thermo Scientific™)

Lyophilized peptide with the sequence N-Tyr-Pro-Tyr-Asp-Val-Pro-Asp-Tyr-Ala-C. Molecular weight: 1102.2 g/mol

26184 can be used to competitively elute HA-tagged fusion proteins from immobilized anti-HA affinity resin. Use 4 X 1 column volumes of 1 mg/ml HA peptide solution to elute HA-tagged proteins. Elution yields will increase by incubating the peptide on the column for 10-15 minutes at 30-37ºC for each elution fraction.

Reconstitute to 1 mg/ml using TBS or distilled water.

The HA peptide, NH2-YPYDVPDYA-COOH, is a synthetic peptide used to competitively elute fusion proteins containing the influenza hemagglutinin (HA) tag bound to immobilized anti-HA affinity resins.

BSA, acetylated (20 mg/mL) (Invitrogen™)

Ambion® Acetylated BSA is used as a carrier protein and as an enzyme stabilizer in which the absence of contaminants such as nucleases and proteases is essential. Six tubes of 25 mg are provided, at a concentration of 20 mg/mL. The acetylation process inactivates any trace nuclease activity that might be present. While the acetylation process does change BSA's binding characteristics and makes it inhibitory in PCR reactions, it is routinely added to restriction digests. The molecular weight is ~66 kD. Acetylated BSA is rigorously tested for contaminating nonspecific endonuclease, exonuclease, protease, and RNase activity.

BGG Stock Solution, acetylated

Invitrogen's Acetylated BGG solution is a 0.5% solution in water. The material is tested for level of background fluorescence.

MBP Bovine Protein, Natural (Gibco™)
