Frizzled 1 Recombinant Human Protein (Gibco™)

Recombinant Frizzled 1 is a bioactive protein intended for use in cell culture applications. FZD1 is a member of the Frizzled family, which are 7-transmembrane domain proteins that are receptors for Wnt signaling proteins. Wnt signaling is involved in several developmental processes.

Recombinant Protein G (Invitrogen™)

Recombinant protein G is produced in a strain of E. coli which contains the gene for protein G from Streptococcus sp. It is supplied as a salt-free lyophilized powder.

Recombinant protein G binds to the Fc region of many types of immunoglobulin G including:

• all four subclasses of human IgG
• mouse IgG
• rabbit IgG
• guinea pig IgG
• rat IgG
• goat IgG
• sheep IgG
• cow IgG
• horse IgG

Recombinant protein G does not bind to IgG from chicken or cat, or to human IgA, IgM, or serum albumin. It binds weakly to IgG from dog.

GFP Recombinant Aequorea victoria Protein, N-His Tag

Green Fluorescent Protein (GFP) recombinant Aequorea victoria protein is supplied as a lyophilized powder. In general, recombinant proteins can be used as protein structure analysis and in cell biology research applications.

This recombinant protein is expressed from a DNA sequence encoding the Aequorea victoria GFP (AAB65663) (Ser 2-Lys 238) fused to a polyhistidine tag at the N-terminus.

N-terminal Sequence Analysis: Glu 99.

Activity:
1. Mesured by emission analysis.
2. Measure excitation and emission spectra of GFP at wavelengths of 395 nm and 510 nm respectively.

Formulation: Lyophilized from sterile PBS, pH 7.5.

Reconstitution: Dissolve the protein in sterile double-distilled water to a concentration of 0.2 mg/ml or lower. It is recommended that the protein be aliquoted and used as soon as possible. Store aliquots under sterile conditions at -20°C. Avoid repeated freeze-thaw cycles.

Expiration Date: Expires one year from date of receipt when stored as instructed.

Background
The green fluorescent protein (GFP) is a protein that exhibit bright green fluorescence when exposed to blue light. The protein is in the shape of a cylinder, comprising 11 strands of β-sheet with an α-helix inside and short helical segments on the ends of the cylinder. Its ability to generate a highly visible, efficiently emitting internal fluorophore is both intrinsically fascinating and tremendously valuable. The green-fluorescent protein (GFP) of the jellyfish Aequorea victoria has long been used as a universal reporter in a broad range of heterologous living cells and organisms. GFP has become well established as a marker of gene expression and protein targeting in intact cells and organisms.

The protein sequence of Frizzled-7 corresponds to the extracellular region of mature Human Frizzled 7, aa 33-185, fused to the Fc region of Human IgG1.

The equilibrium binding constant, KD, of Human Frizzled 7 was measured at less than 10nM using mouse Wnt3a as the ligand. Measurements were made using the ForteBio Octet optical biosensor instrument.

Pierce™ 3x DYKDDDDK Peptide (Thermo Scientific™)

Pierce 3x DYKDDDDK Peptide is a lyophilized peptide with the sequence Met-Asp-Tyr-Lys-Asp-His-Asp-Gly-Asp-Tyr-Lys-Asp-His-Asp-Ile-Asp-Tyr-Lys-Asp-Asp-Asp-Asp-Lys (MDYKDHDGDYKDHDIDYKDDDDK). It has a molecular weight of 2861.9 g/mol. For use, reconstitute to 1.5 mg/mL in PBS.

This tricosa-peptide can be used to competitively elute DYKDDDDK-tagged fusion proteins from immobilized anti-DYKDDDDK affinity supports, especially when a DYKDDDDK-tagged protein is sensitive to low pH. Note that Pierce 3x DYKDDDDK Peptide may interfere with protein determination assays and absorbance at 280 nm. However, the peptide can be removed from the eluate by desalting (e.g., with Zeba Spin Desalting Columns, Cat. No. 89891).

Noggin Recombinant Human Protein (Gibco™)

Recombinant Human Noggin is a bioactive protein intended for use in cell culture applications. Noggin binds and inactivates members of the transforming growth factor-beta (TGF-β) superfamily signaling proteins, such as bone morphogenetic protein-4 (BMP4). By diffusing through extracellular matrices more efficiently than members of the TGF-β superfamily, this protein may have a principal role in creating morphogenic gradients. The protein appears to have pleiotropic effect, both early in development as well as in later stages. It was originally isolated from Xenopus based on its ability to restore normal dorsal-ventral body axis in embryos that had been artificially ventralized by UV treatment. The results of the mouse knockout of the ortholog suggest that it is involved in numerous developmental processes, such as neural tube fusion and joint formation.
