Lysing Solution, Whole Blood Lysing Solution (Invitrogen™)

Invitrogen Cal-Lyse™ is a lysing solution formulated to enable the lysis of erythrocytes in samples of anticoagulated human peripheral blood. Thermo Scientific T-PER Tissue Protein Extraction Reagent enables mild extraction of total protein from tissue samples.

Product Details

This tissue cell lysis reagent utilizes a proprietary detergent in 25mM bicine, 150mM sodium chloride (pH 7.6) to maximize the efficiency of protein solubilization from mammalian tissue samples by homogenization. The simple composition is compatible with additives such as protease inhibitors, salts, reducing agents, chelating agents, providing versatility for many different sample types and lysis applications. Cell lysates prepared with T-PER Reagent are directly compatible with reporter assays (e.g., luciferase, beta-galactosidase, chloramphenicol acetyl transferase), protein kinase assays (e.g., PKA, PKC, tyrosine kinase), immunoassays (e.g., Western blots, ELISAs, RIAs) and/or protein purification procedures.

Homogeneous and efficient tissue protein extraction from different samples is challenging due to varying tissue composition and extraction methods. Fibrous and collagenous tissues, like heart and kidney, make protein extraction by freeze-thaw or mortar and pestle methods inefficient without the aid of detergent. Conversely, soft tissues, such as spleen and liver, require a milder extraction protocol for optimal results. T-PER Reagent was designed for use with all tissues, independent of mechanical extraction methods. The mild detergent enhances solubilization of proteins without compromising function, and the neutral pH and salt concentration of this reagent enable more efficient extraction from cellular compartments yielding homogeneous lysates.

T-PER Reagent was validated for protein yield in both fresh and frozen tissue. Frozen tissue and freeze-thaw extraction protocols predictably resulting in higher protein yield. Additionally, T-PER Reagent is shown to efficiently extract target proteins from various cellular compartments such as plasma membrane ( Na-K+ ATPase, pan-cadherin), cytoplasm (HSP90, MAPK), Mitochondria (COXIV), and nucleus (HDAC1) as tissue specific markers from brain (synaptophysin) and heart tissue (Troponin T Cardiac Isoform).

Pierce Universal Nuclease for Cell Lysis is a genetically engineered endonuclease from Serratia marcescens. The enzyme is produced and purified from E. coli and consists of two identical 30-kDa subunits with two critical disulfide bonds. This indiscriminate endonuclease degrades single-stranded, double-stranded, linear and circular DNA and RNA and is effective over a wide range of temperatures and pH. This enzyme has high specific activity (100-fold greater than DNase I) and increased thermal stability compared to other nucleases. Pierce Universal Nuclease is ≥99 pure enzyme, is free of any measurable protease activity and is supplied at 250U/µL. Pierce Universal Nuclease for Cell Lysis is identical in performance to Benzonase™ Nuclease (EMD Merck).

Applications:
• Use with B-PER, Y-PER or other commercial or homebrew cell lysis reagents and/or mechanical disruption to reduce viscosity in protein extracts
• Remove DNA and RNA from recombinant protein preparations prior to downstream processing

Pierce Universal Nuclease for Cell Lysis is commonly used to reduce the viscosity of bacterial and mammalian protein extracts for downstream application by removing the nucleic acids from protein preparations. The enzyme completely digests nucleic acids to oligonucleotides that are less than 5 bases long. Pierce Universal Nuclease for Cell Lysis helps to improve the separation of the lysate pellet from the supernatant, enhances filtration of the treated lysate, improves chromatography processing time and increases the overall protein yield. The endonuclease has also been shown to improve the compatibility of protein extracts for 2D gel electrophoresis. One unit corresponds to the amount of enzyme required to produce a change of 1.0 in the absorbance at 260nm of sonicated Herring DNA over 30 minutes at 37°C, as determined using standard nuclease from the Merck™ Serratia marcescens volumetric activity assay.

PMSF Protease Inhibitor (Thermo Scientific™)

Thermo Scientific PMSF is a protease inhibitor that reacts with serine residues to inhibit trypsin, chymotrypsin, thrombin and papain.

Features of PMSF:

• Targets serine proteases, including trypsin, chymotrypsin, thrombin and papain
• Effective in most protein solutions at 0.1 to 1 mM final concentration
• Because PMSF has limited water solubility, it must be dissolved in a small amount of solvent, such as ethanol, methanol or isopropyl alcohol before addition to a buffer

Halt Phosphatase Inhibitor Cocktail protects phosphoproteins from serine/threonine phosphatases as well as protein tyrosine phosphatases (PTPs) during cell lysis. The cocktail contains a mixture of four inhibitors of broad specificity, including sodium fluoride, sodium orthovanadate, sodium pyrophosphate and beta-glycerophosphate. Conveniently packaged in a 100X format, the cocktail is stable at 4°C and is suitable for use with any cell or tissue extract. The cocktail preserves protein phosphorylation in immunoprecipitation and kinase assays.

Phosphorylation and dephosphorylation comprise a molecular on/off switch that regulates many key biological pathways within the cell, including signal transduction, cell division and apoptosis. Proteases make up a large category of enzymes, including endopeptidases and exopeptidases, which catalyze the hydrolytic breakdown of proteins into peptides or amino acids in a wide variety of cell and tissue types. Proteases are regulated and compartmentalized in living cells, but cell lysis releases and mixes them with other extracted proteins. Their activity can thus lead to the loss of a large number of valuable proteins, adversely affecting downstream applications. For this reason, it is common practice to spike cell lysis buffers or cell extracts with a cocktails of compounds known to inactivate or inhibit proteases. DNA Extract All Reagents Kit (Applied Biosystems™)

The DNA Extract All Reagents Kit provides PCR-ready DNA from a wide variety of sample types, ranging from blood to buccal swabs to plant tissues in 5 minutes.

Simply mix your sample with the DNA lysis solution provided. Incubate for 3 minutes and add the DNA stabilizing solution, also provided.

After extraction, robust PCR amplification can be completed using TaqMan® GTXpress™ Master Mix or TaqMan® OpenArray® Genotyping Master Mix (pre-amplified sample only), depending on your application. PCR can be performed using either Fast or Standard mode thermal cycling conditions. The DNA Extract All Reagents Kit is included in the TaqMan Sample-to-SNP Kit, so workflow details can be found in the TaqMan® Sample-to-SNP user guide below.

This 200 mL size should be sufficient for 4000 extractions.

Note: Thermal cycling conditions should be followed according to the master mix being used. TaqMan®™ GTXpress™ Master Mix is required for PCR amplification of sample prepared using the DNA Extract All Reagents Kit. If sample has undergone preamplification, OpenArray® Genotyping Master Mix may also be used.
