The NE-PER Kit is a nuclear protein extraction method that involves simple, stepwise lysis of cells and centrifugal isolation of nuclear and cytoplasmic protein fractions. A benchtop microcentrifuge, tubes and pipettors are the only tools required. The NE-PER Reagents efficiently solubilize and separate cytoplasmic and nuclear proteins into fractions with minimal cross-contamination or interference from genomic DNA and mRNA. Once desalted or diluted, the isolated proteins can be used to perform immunoassays and protein interaction experiments, such as mobility shift assays (EMSA), co-immunoprecipitation (Co-IP) and pull-down assays.

There are a variety of methods available to isolate nuclei and prepare nuclear protein extracts. Most of the methods for preparing nuclear extracts are lengthy processes requiring mechanical homogenization, freeze/thaw cycles, extensive centrifugation or dialysis steps that may compromise the integrity of nuclear proteins. The NE-PER Nuclear and Cytoplasmic Extraction Reagent Kit is a reagent-based protocol that enables the stepwise lysis of cells, separation of the cytoplasm from the intact nuclei and then extraction of nuclear proteins away from genomic DNA and mRNA. This gentle process takes less than two hours and requires only a standard bench top centrifuge when using cultured cells. Furthermore, both active nuclear proteins and cytoplasmic proteins can be recovered from the same cell culture or tissue sample.

From two million cells, the kit typically yields 200 to 500 µg cytoplasmic protein and 100 to 200 µg nuclear protein (at a concentration of 1 mg/mL). Typical cross-contamination between cytosolic and nuclear fractions is about 10%. The method recovers primarily soluble proteins from the nucleus, not so much proteins bound in chromatin or integral membrane proteins of the nuclear envelope. The protein concentration of the nuclear extracts can be manipulated easily by varying the volume of nuclear extraction reagent (NER) used in the extraction without any significant loss in extraction efficiency. Specific extraction of nuclear proteins from cells is central to the success of many gene regulation studies. While a variety of methods exist to isolate nuclei and prepare nuclear protein extracts, most of these are lengthy processes requiring mechanical homogenization, freeze/thaw cycles, extensive centrifugation or dialysis steps that may compromise the integrity of many fragile nuclear proteins. The NE-PER Kit overcomes these problems, providing high-quality, concentrated nuclear protein extracts for use in EMSA and other analysis techniques.

Proteases make up a large category of enzymes, including endopeptidases and exopeptidases, which catalyze the hydrolytic breakdown of proteins into peptides or amino acids in a wide variety of cell and tissue types. Proteases are regulated and compartmentalized in living cells, but cell lysis releases and mixes them with other extracted proteins. Their activity can thus lead to the loss of a large number of valuable proteins, adversely affecting downstream applications. Fractionation of subcellular proteins enables protein localization assessment and protein enrichment from specific cellular compartments. The Subcellular Protein Fractionation Kit for Cultured Cells contains four extraction buffers, a stabilized nuclease and Thermo Scientific Halt Protease Inhibitor Cocktail. Each kit has enough reagents to fractionate 50 cell pellets of 20 µL each, equivalent to approximately 2g of cell paste. The first kit reagent, when added to a cell pellet, causes selective membrane permeabilization, releasing soluble cytoplasmic contents. The second reagent dissolves plasma, mitochondria and ER-golgi membranes but does not solubilize the nuclear membranes. After recovering intact nuclei by centrifugation, a third reagent yields the soluble nuclear extract. An additional nuclear extraction with micrococcal nuclease is performed to release chromatin-bound nuclear proteins. The recovered insoluble pellet is then extracted with the final reagent to isolate cytoskeletal proteins.

Isolation of exosomes can be a tedious, non-specific, and difficult process. Total Exosome Isolation Reagent (from cell culture media) enables simple and reliable concentration of intact exosomes from cell media samples with a protocol that is scalable depending on your sample size.

By tying up water molecules, the reagent forces less-soluble components such as vesicles out of solution, allowing them to be collected by a short, low-speed centrifugation. The reagent is added to the cell media sample, and the solution is incubated overnight at 2°C to 8°C. The precipitated exosomes are recovered by standard centrifugation at 10,000 x g for 60 min. The pellet is then resuspended in PBS or similar buffer, and the exosomes are ready for downstream analysis or further purification by affinity methods. Total RNA and protein can then be purified using the Total Exosome RNA and Protein Isolation Kit, or the intact exosomes can be used for biological studies of their pathways, functions, and trafficking. Pierce™ IP Lysis Buffer (Thermo Scientific™)

Thermo Scientific Pierce IP Lysis Buffer is optimized for cell lysate yield, purity and compatibility with immunoprecipitation (IP and Co-IP) as the downstream application for the cell lysate.

IP Lysis Buffer is a mammalian whole cell lysis reagent based on a modified RIPA buffer formulation without SDS. This moderate-strength lysis buffer effectively solubilizes cellular proteins but does not liberate genomic DNA or disrupt protein complexes like ordinary RIPA buffer. Pierce IP Lysis Buffer is specially formulated for pull-down and immunoprecipitation assays.

Pierce IP Lysis Buffer is effective for lysing cultured mammalian cells from both plated cells and cells pelleted from suspension cultures. Optimized for pull-down and immunoprecipitation assays, this lysis buffer is also compatible with many other applications, including the Thermo Scientific Pierce BCA and 660 nm Protein Assays, protein purification and immunoassays (e.g., ELISA, Western blot).

Pierce IP Lysis Buffer is composed of 25 mM Tris-HCl pH 7.4, 150 mM NaCl, 1 mM EDTA, 1% NP-40 and 5% glycerol. The buffer does not contain protease or phosphatase inhibitors; however, if desired, inhibitors, such as Thermo Scientific Halt Protease Inhibitor Cocktail or Phosphatase Inhibitor Cocktail can be added just before use to prevent proteolysis and maintain phosphorylation of proteins.

The P-PER Kit includes an organic lysing reagent and two aqueous reagents that effectively lyse plant cells and solubilize protein without harsh mechanical lysis aids, such as a mortar and pestle. Plant protein extracts are prepared in just 10 minutes, and protein yields exceed conventional extraction methods (e.g., freeze/grinding in liquid nitrogen) and other commercially available plant protein extraction reagents.
