Thermo Scientific Y-PER Yeast Protein Extraction Reagent is the first commercially available yeast lysis reagent using a mild detergent lysis procedure to efficiently release functionally active solubilized proteins.

The detergent-based cell lysis buffer eliminates the need to use glass beads or mechanical treatments involving strong reducing agents, chemicals, pH and temperature extremes to disrupt the proteinaceous cell envelope required for protein extractions. The popular glass bead lysis protocol requires special equipment and must be performed at 4°C. The low yields of protein commonly obtained with this technique are the result of denaturation and proteins nonspecifically binding to the glass beads. However, Y-PER Reagent uses a simple room temperature protocol that can be completed in 20 minutes and requires no special equipment.

Y-PER Reagent offers an easy to use standard protocol for protein extractions involving addition of an appropriate volume of Y-PER Reagent to pelleted yeast cells, followed by incubation at room temperature for approximately 20 minutes and then spin down the debris. The resulting supernatant is a concentrated protein solution, surpassing typical yields obtained with traditional glass bead disruption.

The unicellular nature of yeast, combined with its ability to perform post-translational modifications that closely mimic processes in higher eukaryotes, have made them important research tools. The many vectors available, as well as the development of recombinant expression systems, have given yeast an irreplaceable role in the research sector.Y-PER Reagent is effective for S. cerevisiae and other popular species, making it applicable for use in fusion-tagged protein purification and reporter enzyme assays with these model organisms. The lysis reagent also can be used for genomic and plasmid DNA extraction from yeast.

Product Details (Recommendations for Use):
Fresh Cells and Frozen Cells: The Y-PER-Plus Reagent is capable of extracting proteins equally well from both freshly harvested and frozen cells
Cell Density and Strain Variation: Differences in growth rate among organisms, growth temperature and media composition affects the number of cells harvested from a given culture volume. Therefore several suggestions for the amount of Y-PER-Plus Reagent to use for a given cell pellet (wet cell paste) weight are included.
Pichia pastoris: To achieve lysis of cells grown in rich media such as YEPD, cells must be harvested during log-phase growth. To enhance lysis of cells in stationary phase, add DTT or TCEP-HCl (to a final concentration of 0.1M and 20 to 50 mM, respectively) directly to the Y-PER-Plus Reagent before lysis to release much more of the soluble proteins. For best results use 2.5 to 5.0 mL of Y-PER-Plus Reagent per 1 gram of cell paste; scale up or down accordingly. An alternative to adding reducing agents is adding a protease inhibitor and incubating at 45°C.
Saccharomyces cerevisiae: For best results, harvest cells in log-phase. To increase yield for cells in stationary-phase, add DTT or TCEP·HCl (to a final concentration of 0.1M and 20 to 50 mM, respectively) directly to the Y-PER-Plus Reagent before extraction. No differences in efficiency results for either rich- or synthetic-defined media. For best results use 2.5 to 5.0 mL of Y-PER Plus Reagent per 1 gram of cell paste; scale up or down accordingly.
Schizosaccharomyces pombe: For best results, use the original Y-PER Reagent (Part No. 78990) for extracting proteins from S. pombe.
Bacillus subtilis: The Y-PER-Plus Reagent will not extract protein from B. subtilis spores. When using a strain that is able to sporulate, harvest cells during log-phase. For strains unable to sporulate, cells may be grown to saturation before harvesting. Use 2.5 to 5.0 mL of Y-PER-Plus Reagent per 1g of cell paste; scale up or down accordingly.
Escherichia coli: Use 2.5 to 5.0 mL of Y-PER-Plus Reagent per 1g of cell paste; scale up or down accordingly.
Enzyme Activity: Because all proteins differ in structure, solubility and stability, a particular protein might not retain optimum activity in the presence of Y-PER-Plus Reagent. For example, the original Y-PER Reagent works well for assaying beta-galactosidase activity, but Y-PER-Plus Reagent slows down its kinetics. Y-PER-Plus Reagent is compatible with standard affinity-purification protocols for glutathione S-transferase (GST) and His-tagged proteins.

Halt Phosphatase Inhibitor Cocktail protects phosphoproteins from serine/threonine phosphatases as well as protein tyrosine phosphatases (PTPs) during cell lysis. The cocktail contains a mixture of four inhibitors of broad specificity, including sodium fluoride, sodium orthovanadate, sodium pyrophosphate and beta-glycerophosphate. Conveniently packaged in a 100X format, the cocktail is stable at 4°C and is suitable for use with any cell or tissue extract. The cocktail preserves protein phosphorylation in immunoprecipitation and kinase assays.

Phosphorylation and dephosphorylation comprise a molecular on/off switch that regulates many key biological pathways within the cell, including signal transduction, cell division and apoptosis. To effectively study phosphorylation activation states, one must inactivate phosphatases to prevent their uncontrolled activity upon cell lysis. Halt Phosphatase Inhibitor Cocktail accomplishes this task.

Thermo Scientific B-PER Reagent (in Phosphate Buffer) is an easy-to-use cell lysis reagent is a nonionic detergent-based solutions that effectively disrupt cells and solubilize native or recombinant proteins without denaturation. All B-PER Reagents are compatible with downstream applications, such as affinity chromatography (e.g., immobilized metal affinity chromatography, glutathione chromatography), SDS-PAGE, and protein quantification (e.g., Pierce BCA Protein Assay, Pierce 660nm Protein Assay). DNA Extract All Reagents Kit (Applied Biosystems™)

The DNA Extract All Reagents Kit provides PCR-ready DNA from a wide variety of sample types, ranging from blood to buccal swabs to plant tissues in 5 minutes.

Simply mix your sample with the DNA lysis solution provided. Incubate for 3 minutes and add the DNA stabilizing solution, also provided.

After extraction, robust PCR amplification can be completed using TaqMan® GTXpress™ Master Mix or TaqMan® OpenArray® Genotyping Master Mix (pre-amplified sample only), depending on your application. PCR can be performed using either Fast or Standard mode thermal cycling conditions. The DNA Extract All Reagents Kit is included in the TaqMan Sample-to-SNP Kit, so workflow details can be found in the TaqMan® Sample-to-SNP user guide below.

This 200 mL size should be sufficient for 4000 extractions.

Note: Thermal cycling conditions should be followed according to the master mix being used. Add 10 µL of concentrated cocktail per 1 mL of lysis buffer to ensure complete protection of the resulting protein extract. The single-use format is ideal for small-scale cell lysis experiments, where each 100 µL vial provides just what is needed for a 10 mL sample and there is no risk of contaminating or degrading remaining aliquots of cocktail. Each microtube in the strip is heat-sealed with a foil that can be easily punctured by a pipette tip for trouble-free access to a precise amount of Halt Protease Inhibitor Cocktail needed for each experiment.

Proteases make up a large category of enzymes, including endopeptidases and exopeptidases, which catalyze the hydrolytic breakdown of proteins into peptides or amino acids in a wide variety of cell and tissue types. Proteases are regulated and compartmentalized in living cells, but cell lysis releases and mixes them with other extracted proteins. Their activity can thus lead to the loss of a large number of valuable proteins, adversely affecting downstream applications. For this reason, it is common practice to spike cell lysis buffers or cell extracts with a cocktails of compounds known to inactivate or inhibit proteases. Halt Protease Inhibitor Cocktails effectively inhibit serine-proteases, cysteine-proteases, aspartic acid-proteases and aminopeptidases that are typically present in cellular lysate samples. Regular and EDTA-free formulations provide for inhibition of metalloproteases and compatibility with 2D electrophoresis, respectively.

Isolation of exosomes can be a tedious, non-specific, and difficult process. Total Exosome Isolation Reagent (from serum) enables simple and reliable concentration of intact exosomes from blood serum samples with a protocol that is scalable depending on your sample size.

By tying up water molecules, the reagent forces less-soluble components such as vesicles out of solution, allowing them to be collected by a short, low-speed centrifugation. The reagent is added to the serum sample, and the solution is incubated at 2°C to 8°C for 30 min. The precipitated exosomes are recovered by standard centrifugation at 10,000 x g for 10 min. The pellet is then resuspended in PBS or similar buffer, and the exosomes are ready for downstream analysis or further purification by affinity methods. Total RNA and protein can then be purified using theTotal Exosome RNA and Protein Isolation Kit, or the intact exosomes can be used for biological studies of their pathways, functions, and trafficking. For immunoprecipitation of exosomal proteins, use Exosome Immunoprecipitation (Protein A) or Exosome Immunoprecipitation (Protein G).

To isolate intact exosomes from cell culture media, we recommend Total Exosome Isolation Reagent (from cell culture media).

N-PER™ Neuronal Protein Extraction Reagent (Thermo Scientific™)

Thermo Scientific N-PER Neuronal Protein Extraction Reagent is a proprietary cell lysis reagent optimized for efficient extraction of proteins from all cellular compartments of neuronal tissue and primary cultured neurons.

Features of N-PER Neuronal Protein Extraction Reagent:

Typical neuronal protein yields are 70-90 µg of protein per mg of brain tissue or 300 µg of total protein from 106 primary neuronal cells.

Typical neuron structures include the cell body (soma), dendrites and an axon. The unique morphology and rich sphingolipid and cholesterol composition of neuron cell membranes creates challenges in preparing neuronal protein extracts. N-PER Neuronal Protein Extraction Reagent is specifically formulated to overcome the challenges of extracting functional native proteins from brain or other neuronal tissue and primary cultured neurons while increasing protein yield compared to other extraction methods.

The mild formulation of the N-PER Cell Lysis Reagent is ideal for downstream applications requiring active neuronal proteins. In addition, the N-PER Reagent is compatible with many applications including enzyme assays (e.g., phosphatase, kinase), immunoassays, various chromatography procedures and electrophoresis. Thermo Scientific Halt Protease Inhibitor Cocktail or Phosphatase Inhibitor Cocktail can be added just before use to prevent proteolysis or to offer additional protection from the high phosphatase activity normally present in brain tissue.

For each extraction, a ratio of 1g of tissue to 10 mL of N-PER Reagent or 0.5-1 mL N-PER Reagent per 10 cm petri dish of primary cultured cells is recommended.
