LanthaScreen™ Tb-anti-Histone H3K9me2 Antibody (Invitrogen™)

The LanthaScreen® Tb-anti-Histone H3K9me2 Antibody is a key component of the BacMam Histone H3K9me2 Cellullar Assay (A14161). When this terbium (Tb)-labeled antibody is paired with baculovirus-mediated (BacMam) expression of a Histone H3-GFP fusion protein, the TR-FRET readout enables sensitive detection of H3 lysine 9 dimethylation. The resulting high-throughput screening (HTS) compatible assay can be used to identify inhibitors of methyltransferases and demethylases acting on lysine 9 of histone H3 in the cell type of your choice.

The LanthaScreen® Histone H3K9me2 antibody allows for the investigation of dimethylation at lysine 9 on histone H3. This enables screening for potential inhibitors of methyl transferases and demethylases associated with Lys9 dimethylation in their natural complexes in a physiologically relevant cell type.

BacMam and LanthaScreen® Convenience Saves Sample and Time
Assays are run in a fully homogenous, addition-only format without any washing, lysate transfer, or separation procedures required for traditional methods such as western blotting and ELISA, making it ideal for HTS applications. By using the same BacMam reagent for GFP-Histone H3, other histone H3 modifications (Ph, Me, Ac) can be monitored by simply exchanging the Terbium (Tb)-labeled antibody. In addition, the application of the LanthaScreen® technology includes all of the advantages of TR-FRET detection including reduced data noise, less interference from fluorescent compounds, and high sensitivity, which results in the need to use fewer cells than traditional western or ELISA methods.

LanthaScreen™ Tb-pMAP2K1 (pSer217/221) Antibody

A member of the MAP Kinase family, MAP2K1 (mitogen-activated protein kinase kinase 1) is phosphorylated at Ser217⁄221 by a range of MAPK family members including Raf and B-Raf.
For use in combination with the Fluorescein-MAP2K1 physiological protein substrate (PV4812) to evaluate inhibitiors in medium to high throughput screening applications.

LanthaScreen™ Tb-anti-Biotin Antibody Kit

The LanthaScreen® terbium (Tb)-labeled anti-biotin antibody is a generic reagent that can be used to easily label biotinylated biomolecules for use in TR-FRET assays. Although the terbium chelate donor can be directly attached to a biomolecule such as a protein or peptide, it can also be indirectly attached to a target molecule through a biotin-mediated association, in the form of a Tb-labeled streptavidin or anti-biotin antibody. Invitrogen’s Tb-labeled biotin-binding reagents are universal reagents that easily label biotinylated biomolecules with terbium for use in TR-FRET assays for proteases, kinases, etc.



Contents and Storage:
Each LanthaScreen® Tb-labeled anti-biotin antibody is supplied in HEPES-buffered saline. The kit also includes 400 µl of a fluorescein-labeled positive control and 100 ml of TR-FRET dilution buffer. Store components as indicated in the product Certificate of Analysis (-20°C, or +20-30°C).

LanthaScreen™ Tb-anti-Histone H3K27me3 Antibody

The LanthaScreen® Tb-anti-Histone H3K27me3 antibody is a key component of the BacMam Histone H3K27me3 Cellullar Assay (A14159). When this terbium (Tb)- labeled antibody is paired with baculovirus-mediated (BacMam) expression of a Histone H3-GFP fusion protein, the TR-FRET readout enables sensitive detection of H3 lysine 27 trimethylation. The resulting high-throughput screening (HTS) compatible assay can be used to identify inhibitors of methyltransferases and demethylases acting on lysine 27 of histone H3 in the cell type of your choice.

The LanthaScreen® Histone H3K27me3 antibody allows for the investigation of trimethylation at lysine 27 on histone H3. This enables screening for potential inhibitors of methyl transferases and demethylases associated with Lys27 trimethylation in their natural complexes in a physiologically relevant cell type. Please note that the LanthaScreen® Tb-anti-Histone H3K4me3 antibody has been shown to have higher affinity for trimethylation compared to dimethylation at lysine 4 using a histone H3 peptide; however, when interpreting your results it is important to consider that the antibody does show some affinity for the dimethylated peptide. Further discrimination between di- and trimethylation can be accomplished by using our complimentary LanthaScreen® Tb-anti-Histone H3K4me2 Antibody (Cat # A14158 and A14153), which is specific for dimethylation.
