About Dynabeads® CD19 Pan B

Because of the bead size, Dynabeads® CD19 pan B can easily and efficiently isolate or deplete cells from viscous samples such as whole blood and bone marrow in about 30 minutes. Dynabeads® CD19 pan B will also afford excellent recovery of high-purity, viable cells for use in downstream molecular studies; for example, those in which cells are to be lysed while still attached to beads, and nucleic acids or proteins further purified. Note that intact cells will not be released from these beads, so if you want to isolate CD19+ cells for cell-based applications, or you need to check your samples with flow cytometry, you can combine with DETACHaBEAD® CD19 (purchased separately to get bead- and antibody-free cells) or use Dynabeads® Untouched™ Human B Cells to obtain untouched human B cells.

Magnetic bead-based separation offers easy handling
Dynabeads® CD19 Pan B is added to the sample under continuous mixing to optimize the binding of the Dynabeads® to the target cells. By placing the sample on a magnet, it separates the bead-bound target cells from the rest of the sample in just 1–2 minutes. For depletion, remove the supernatant to a new tube for further studies and discard the bead-bound cells. For positive isolation for molecular studies, remove the supernatant and wash the bead-bound cells 2–3 times in buffer to get optimal purity. The cells can be lysed while still attached to the beads and the supernatant transferred to a new tube for downstream molecular analysis. Starting samples can be whole blood, buffy coat, bone marrow, PBMC, or tissue digests.

Dynabeads™ Regulatory CD4+/CD25+ T Cell Kit (Invitrogen™)

Use this kit to isolate highly pure CD4+ CD25+ regulatory T-cells (Treg) that express the intracellular transcription factor Foxp3. The CD4+ CD25- cell fraction can be used as effector cells in downstream inhibitory assays. You can expect >95% purity (CD4+ CD25+ expression) and >80% Foxp3 expression.

Isolate bead- and antibody-free human CD4+ CD25+ Foxp3+ Treg cells

Methodology

The isolation procedure includes the following steps:

• Negative isolation of untouched human CD4+ T-cells from PBMC.

Use this kit to isolate pure and viable untouched B cells from PBMC by negative isolation. The kit depletes T cells, NK cells, monocytes, platelets, dendritic cells, granulocytes and erythrocytes. The negatively isolated human B cells are left in the sample and have not been in contact with the Dynabeads. The bead and antibody-free B cells are truly untouched, and in perfect shape for any functional assay and application.

This gentle and easy-to-use kit allows you to isolate flow-compatible human CD8+ T-cells from PBMC or directly from whole blood. Immediately after positive isolation, the beads are released and removed from your cell sample. In this way, you avoid exposing your cells to potentially cytotoxic or immunogenic foreign substances like iron oxides or dextrans, a potential problem when working with biodegradable particles. Use this kit to isolate pure and viable untouched CD4+ T-cells from PBMC by negative isolation. Methodology: An antibody mix towards the non-CD4 cells is added to the sample and allowed to bind to the cells. The remaining negatively isolated and untouched human CD4+ T-cells can be directly analyzed in a flow cytometer and used in any application.

About Dynabeads® CD45
Dynabeads® CD45 are uniform, superparamagnetic beads (4.5 µm diameter) coated with a primary monoclonal mouse IgG2a antibody specific for a CD45 membrane antigen common to all known isoforms of CD45. CD45 is expressed on all human leucocytes. Because of the bead size, Dynabeads® CD45 can easily and efficiently isolate or deplete cells from viscous samples such as whole blood and bone marrow in about 30 minutes. Dynabeads® CD45 is often used for enrichment of circulating non-hematopoietic tumor cells, by depletion of all CD45+ leucecytes from a MNC sample. Positively isolated cells can be used for downstream molecular studies; for example, those in which cells are to be lysed while still attached to beads, and nucleic acids or proteins further purified. Note that intact cells will not be released from these beads, and we do not recommend to analyze the bead-bound cells with a flow cytometer.

Dynabeads® CD45 is added to the sample under continuous mixing to optimize the binding of the Dynabeads® to the target cells. By placing the sample on a magnet it separates the bead-bound target cells from the rest of the sample in just 1–2 minutes. For depletion, remove the supernatant to a new tube for further studies and discard the bead-bound cells. For positive isolation for molecular studies, remove the supernatant and wash the bead-bound cells 2–3 times in buffer to get optimal purity. The cells can be lysed while still attached to the beads and the supernatant transferred to a new tube for downstream molecular analysis. The MagniSort™ Human CD4 2-Step Enrichment Kit is designed for the magnetic separation of CD4+ T cells by positive selection after first depleting monocytes. It has been optimized for the isolation of CD4+ T cells from human peripheral blood monocuclear cells, utilizing biotinylated Anti-Human CD14 and Anti-Human CD4 antibodies, as well as streptavidin-coated magnetic beads. In the first step, monocytes are bound by Anti-Human CD14 antibody and then magnetic beads. When placed in a magnetic field, the monocytes are held in the magnet while the remaining cells are decanted and used for the second step. In the second step, the monocyte-depleted cells are bound by Anti-Human CD4 antibody and then magnetic beads. Now, when placed in the magnetic field, the CD4+ T cells are held in the magnet while undesired cells can be decanted.

The MagniSort™ Human CD3 Positive Selection Kit is designed for the magnetic separation of CD3+ cells by positive selection. It has been optimized for the isolation of CD3+ cells from human peripheral blood monocuclear cells, utilizing a biotinylated Anti-Human CD3 antibody and streptavidin-coated magnetic beads. CD3+ cells are bound by antibody and then magnetic beads. When placed in a magnetic field, the undesired cells can be separated from CD3+ cells by decanting.

After positive selection, the purity of selected cells can be verified by staining with Anti-Human CD3, clone UCHT1 or SK7.

MagniSort™ Human B Cell Enrichment Kit (2nd Generation) (Invitrogen™)

The MagniSort™ Human B cell Enrichment Kit (2nd Generation) is designed for the magnetic separation of B cells by negative selection with improved purity. It has been optimized for the isolation of human B cells from normal human peripheral blood mononuclear cells utilizing a biotinylated antibody cocktail and streptavidin-coated magnetic beads. Undesired cells are bound by antibody and then magnetic beads that, when placed in a magnetic field, leave B cells untouched and free in solution.

CELLection™ Epithelial Enrich Dynabeads™ (Invitrogen™)

Use this kit to obtain up to 5-log enrichment of human epithelial tumor cells, directly from whole blood, bone marrow or PBMC. Tumor cell clumps can be enriched from bone marrow. Enriched cells are bead-free, viable and suitable for immunocytochemical staining or any other downstream application.

Methodology: Add CELLection™ Dynabeads® to the sample. The beads will bind the tumor cells after a short incubation. The bead-bound tumor cells are quickly separated on a magnet and subsequently released from the beads with the supplied Release Buffer. (CELLection™ Dynabeads® contains a DNA linker that is cleaved by the DNase I in the Release Buffer.)

Note: If you want to deplete or use the isolated tumor cells for molecular applications, please use the equivalent product Dynabeads® Epithelial Enrich (Cat. No. 161-02). This product contains the same antibody, but with no release of the cells from the beads after positive isolation.

The MagniSort™ Human CD117 (c-Kit) Positive Selection Kit is designed for the magnetic separation of CD117+ cells by positive selection. It has been optimized for the isolation of CD117+ cells from normal human peripheral blood cells. CD117+ cells are bound by antibody and then magnetic beads. When placed in a magnetic field, the undesired cells can be separated from CD117+ cells by decanting.

After positive selection, the purity of selected cells can be verified by staining with Anti-Human CD117, clone YB5.B8, and Anti-Human CD34, clone 4H11.

Dynabeads® CD14 are superparamagnetic beads coupled with an anti-human CD14 antibody that enable easy isolation or depletion of human CD14+ cells directly from any sample, including whole blood, bone marrow, buffy coat, mononuclear cells (MNC), and tissue digests. Advantages of Dynabeads® CD14:

About Dynabeads® CD14
Dynabeads® CD14 are uniform, superparamagnetic beads (4.5 µm diameter) coated with a primary monoclonal antibody specific for the CD14 membrane antigen mainly expressed mainly on human monocytes. Because of the bead size, Dynabeads® CD14 can easily and efficiently isolate or deplete cells from viscous samples such as whole blood and bone marrow in about 30 minutes. Dynabeads® CD14 will also afford excellent recovery of high-purity, viable cells for use in downstream molecular studies; for example, those in which cells are to be lysed while still attached to beads, and nucleic acids or proteins further purified. Note that intact cells will not be released from these beads, so if you want to isolate CD14+ cells for cell-based applications, or you need to check your samples with flow cytometry, check out our total human monocyte product offerings.

Magnetic bead–based separation offers easy handling
Dynabeads® CD14 is added to the sample under continuous mixing to optimize the binding of the Dynabeads® to the target cells. By placing the sample on a magnet, it separates the bead-bound target cells from the rest of the sample in just 1–2 minutes. For depletion, remove the supernatant to a new tube for further studies and discard the bead-bound cells. For positive isolation for molecular studies, remove the supernatant and wash the bead-bound cells 2–3 times in buffer to get optimal purity. The cells can be lysed while still attached to the beads and the supernatant transferred to a new tube for downstream molecular analysis. Starting samples can be whole blood, buffy coat, bone marrow, PBMC, or tissue digests.
