MagniSort™ Mouse CD11b Positive Selection Kit (Invitrogen™)

The MagniSort™ Mouse CD11b Positive Selection Kit is designed for the magnetic separation of CD11b+ cells by positive selection. It has been optimized for the isolation of CD11b+ cells from mouse bone marrow cells or splenocytes utilizing a biotinylated Anti-Mouse CD11b antibody and streptavidin-coated magnetic beads. The MagniSort™ Mouse CD49b Positive Selection Kit is designed for the magnetic separation of CD49b+ cells by positive selection. It has been optimized for the isolation of CD49b+ cells from mouse spleens or lymph nodes utilizing a biotinylated Anti-Mouse CD49b antibody and streptavidin-coated magnetic beads. CD49b+ cells are bound by antibody and then magnetic beads. When placed in a magnetic field, the undesired cells can be separated from CD49b+ cells by decanting.

The MagniSort™ Mouse CD45 Depletion Kit is designed for the magnetic depletion of mouse CD45+ cells from mouse spleens, bone marrow or digested lung tissue. It utilizes a biotinylated Anti-Mouse CD45 antibody and streptavidin-coated magnetic beads. CD45+ cells are bound by antibody and then magnetic beads. When placed in a magnetic field, the CD45- cells can be separated from CD45+ cells by decanting.

After depletion, the purity of enriched CD45- cells can be verified by staining with Anti-Mouse CD45, clone 30-F11.

Dynabeads™ Untouched™ Mouse CD8 Cells Kit (Invitrogen™)

Dynabeads® Untouched™ Mouse CD8 Cells supplies both 4.5 µm superparamagnetic Dynabeads® (with a polyclonal sheep anti-rat IgG antibody covalently bound to the bead surface) and a monoclonal Antibody Mix, together designed for the depletion of CD8-negative leucocytes from mouse spleen or lymph node single-cell suspensions. Following the depletion procedure, the negatively isolated, untouched target mouse CD8+ cells are pure, viable, and ready to be used in any downstream application.

The Antibody Mix contains a cocktail of rat IgGs that binds mouse CD4+ T cells, B cells, NK cells, monocytes/macrophages, dendritic cells, erythrocytes, and granulocytes. The Antibody Mix is first added to the sample in a tube to bind the unwanted CD8-negative cells. The cells are then washed to remove excess antibodies. Mouse Depletion Dynabeads® (coated with a rat IgG antibody) are added to bind the antibody-labeled, CD8-negative cells. After a short incubation, the bead-bound cells are then separated from unbound cells in 1–2 minutes using a magnet. The purified target mouse CD8+ cells in the supernatant can be transferred to a new tube for use in any downstream application.

This fast and gentle isolation method does not require the use of columns, and helps ensure high purity, recovery and viability of the untouched CD8+ cells. The protocol can be scaled up according to your sample size.

Dynabeads™ FlowComp™ Mouse CD4+CD25+ Treg Cells Kit (Invitrogen™)

This gentle and easy-to-use kit allows you to isolate highly pure and functional mouse Treg cells from mouse lymph nodes or spleen samples. Your cells are not exposed to the stress of being passed through a column. The isolated CD4+CD25+Foxp3+ Treg cells are bead-free and flow-compatible. The cells can be readily expanded and retain their functional suppressive capacity. You can expect a purity level of >90% Treg cells. Over 88% of the isolated CD25+ cells express Foxp3. You will save time and money, and get increased confidence in your results.

Methodology:
Negative isolation of CD4+ T cells, followed by positive isolation of CD25+ regulatory T cells. First, an antibody mix is added to the sample and the Ab will bind the non-CD4+ T cells. Mouse Depletion Dynabeads® are then added and will bind to the Ab-cell complex during a short incubation. Bead-bound cells are separated on a magnet and discarded. The remaining untouched mouse CD4+ T-cells are then labeled with FlowComp™ CD25 Antibody and positively isolated using FlowComp™ Dynabeads®. A short incubation with FlowComp™ Release Buffer then releases the isolated mouse regulatory T-cells from the Dynabeads®. The isolation method is easy to use and to scale up.

This gentle and easy-to-use kit allows you to isolate flow-compatible mouse CD4+ T cells. Immediately after positive isolation, the beads are released and removed from your cell sample. In this way, you avoid exposing your cells to potentially cytotoxic or immunogenic foreign substances like iron oxides or dextrans, a potential problem when working with biodegradable particles. In addition, your cells will not be exposed to the stress of being passed through a column.

You can expect a purity level of >85% and viability of >85% of mouse CD4+ cells.

Dynabeads® Mouse pan B (B220) are uniform, superparamagnetic beads (4.5 µm diameter) that enable easy isolation of mouse B cells from spleen or lymph nodes, although any single cell suspension may be used. Dynabeads® Mouse pan B beads are coated with a primary monoclonal antibody specific for the B220 membrane antigen. B220 (also known as CD45R) is expressed on all B cells throughout development, from the early pro-B stage of B cell differentiation. Dynabeads® Mouse pan B binds to B cells during a short incubation period in a tube, after which the bead-bound cells are separated from unbound cells using a magnet. This fast and gentle isolation method does not require the use of columns, and helps ensure high purity, recovery and viability of the isolated B cells.

Ideal for Both Depletion and Positive Isolation for Downstream Molecular Assays
Because of the powerful magnets used for separation, Dynabeads® Mouse pan B (B220) can be used in viscous samples such as single cell suspensions from tissue, whole blood and bone marrow, and enable the efficient depletion of mouse B cells in about 30 minutes. Dynabeads® Mouse pan B will also afford excellent recovery of high-purity, viable cells when used for positive isolation for downstream studies; for example, those in which cells are to be lysed while still attached to beads, and nucleic acids or proteins further purified. Note that viable cells will not be released from these beads, so if you want to isolate mouse B cells for cell-based applications, or you need to check your samples with flow cytometry, you should use Dynabeads® Mouse CD43 (Untouched™ B cells).
