MagniSort™ Mouse CD11b Positive Selection Kit (Invitrogen™)

The MagniSort™ Mouse CD11b Positive Selection Kit is designed for the magnetic separation of CD11b+ cells by positive selection. It has been optimized for the isolation of CD11b+ cells from mouse bone marrow cells or splenocytes utilizing a biotinylated Anti-Mouse CD11b antibody and streptavidin-coated magnetic beads. Please refer to the MagniSort™ Mouse CD11b Positive Selection Protocol for specific optimal conditions for isolating CD11b+ cells from either of these two tissues. Dynabeads™ Mouse CD43 (Untouched™ B Cells) (Invitrogen™)

Use this kit to isolate pure and viable untouched B cells from mouse lymphoid organs by negative isolation. The kit depletes T cells, monocytes/macrophages, NK cells, plasma cells, CD5+ B1 cells, platelets, dendritic cells, granulocytes and erythrocytes. The negatively isolated mouse B cells are left in the sample and have not been in contact with the Dynabeads. The bead and antibody-free B cells are truly untouched, and in perfect shape for any functional assay and application.

Methodology:
Dynabeads Mouse CD43 (Untouched B Cells) is added directly to the sample and binds the unwanted cells. The bead-bound cells are quickly separated on a magnet and discarded. Dynabeads® Mouse pan T (Thy 1.2) are magnetic beads that provide a robust and versatile tool to isolate or deplete mouse T-cells directly from any sample, including tissue digests such as spleen or lymph nodes, whole blood, bone marrow, and mononuclear cells (MNC).

Excellent Yield, Purity, and Viability of Recovered Cells
Dynabeads® Mouse pan T (Thy 1.2) are uniform, superparamagnetic beads (4.5 µm diameter) that enable easy isolation of mouse T-cells from spleen or lymph nodes, although any single-cell suspension may be used. Dynabeads® Mouse pan T beads are coated with a primary monoclonal antibody specific for the Thy 1.2 membrane antigen expressed on peripheral T cells, thymocytes and intraepithelial T lymphocytes of all common mouse strains. Dynabeads® Mouse pan T binds to T-cells during a short incubation period in a tube, after which the bead-bound cells are separated from unbound cells using a magnet. This fast and gentle isolation method does not require the use of columns, and helps ensure high purity, recovery and viability of the isolated T cells.

Ideal for Both Depletion and Positive Isolation for Downstream Molecular Assays
Because of the powerful magnets used for separation, Dynabeads® Mouse pan T (Thy 1.2) can be used in viscous samples such as single cell suspensions from tissue, whole blood and bone marrow, and enable efficient depletion of mouse T-cells in about 30 minutes. Dynabeads® Mouse pan T will also afford excellent recovery of high-purity, viable cells when used for positive isolation for downstream studies; for example, those in which cells are to be lysed while still attached to beads, and nucleic acids or proteins further purified. Note intact cells will not be released from these beads, so if you want to isolate mouse T-cells for cell-based applications, or you need to check your samples with flow cytometry, you should use Dynabeads® FlowComp Mouse Pan T (CD90.2) (for bead-free cells).

Dynabeads™ FlowComp™ Mouse CD4 Kit (Invitrogen™)

This gentle and easy-to-use kit allows you to isolate flow-compatible mouse CD4+ T cells. Immediately after positive isolation, the beads are released and removed from your cell sample. In this way, you avoid exposing your cells to potentially cytotoxic or immunogenic foreign substances like iron oxides or dextrans, a potential problem when working with biodegradable particles. In addition, your cells will not be exposed to the stress of being passed through a column.

You can expect a purity level of >85% and viability of >85% of mouse CD4+ cells.

After positive selection, the purity of selected cells can be verified by staining with Anti-Mouse CD49b (integrin alpha 2), clone DX5, and Anti-Mouse NK1.1, clone PK136. The Mouse Depletion Dynabeads® suspension contains uniform, superparamagnetic beads (4.5 µm diameter) that enable easy isolation of cells from any single cell suspension. This reagent is therefore especially well-suited for enriching tissue digests such as mouse spleen and lymph node cells. The Mouse Depletion Dynabeads® are coated with a secondary polyclonal antibody that binds rat IgGs. The Antibody Mix contains a cocktail of rat IgGs that binds mouse CD8+ T cells, B cells, NK cells, monocytes/macrophages, dendritic cells, erythrocytes and granulocytes. The Antibody Mix is first added to the sample in a tube to bind the unwanted non-CD4+ cells. The cells are then washed to remove excess antibodies, and Mouse Depletion Dynabeads® are added. After a short incubation, the bead-bound cells are then separated from unbound cells in 1 – 2 minutes using a magnet. The purified target mouse CD4+ cells in the supernatant can be transferred to a new tube for use in any downstream application. This fast and gentle isolation method does not require the use of columns, and helps ensure high purity, recovery and viability of the untouched CD4+ cells. The protocol can easily be scaled up according to your sample size.
