Dynabeads™ Pan Mouse IgG (Invitrogen™)

Dynabeads® Pan Mouse IgG binds monoclonal mouse IgGs of your choice for specific cell isolation or depletion. It will bind all subclasses of mouse IgG and is Fc-specific. Low amount of IgG is required for binding. All cells from all species except from mouse can be isolated with this product, depending only on the specific antibody of choice.

• Depletion: cells can be depleted from a sample
• Positive isolation: cells can be positively isolated from a sample for molecular applications
• Negative isolation: cells can be negatively isolated from a PBMC sample by adding an antibody cocktail of mouse IgGs towards the unwanted cells. Compatible with flow cytometry.

Note: For cellular assays we recommend positive isolation followed by release of beads from the cells. Recommended product for bead-free cells: Dynal® CELLection™ Pan Mouse IgG (Cat. no 115-31D) in combination with your mouse IgG.

Dynabeads™ M-450 Epoxy (Invitrogen™)

Dynabeads® M-450 Epoxy beads are ideal for custom cell-isolation protocols upstream of molecular applications. Purified antibodies can be used in conjunction with these beads to create a magnetic cell separation media with the good antibody orientation.

Ideal for Cell Separation when you have a custom antibody:
Low background and covalent linking of antibodies to the bead surface make these beads an excellent choice for cell separation applications when a downstream release from the beads is not required.

Ultra rapid protocols:

Quickly isolate targeted cell populations upstream of molecular analyses

• Excellent for cell isolation procedures that require custom antibodies
• Couple different custom antibodies to different beads to select specific subpopulations of cells

Not recommended for Immunoprecipitation (IP):
These 4.5 micron beads are not recommended for IP as they have a lower surface capacity per unit mass when compared to 1 µm and 2.8 µm Tosylactivated and Epoxy coated Dynabeads®. Use rather Dynabeads Protein A (10001D, 10002D), Dynabeads Protein G (10003D, 10004D) or one of the Dynabeads Immunoprecipitation Kits (10006D, 10007D)

Coupling procedure outline:
Any purified antibody can be covalently coupled to the bead surface in a easy-to-perform overnight reaction. Simply incubate the antibody you wish to use for your cell isolation procedure with these 4.5 µm Dynabeads®. Optimal coupling occurs at high pH (8.5-9.5) and at 37°C. For pH labile antibodies, coupling can be performed in an alternative buffer at pH 7.4.

About Dynabeads®:
Dynabeads® are non-porous monodisperse superparamagnetic beads. They are highly mobile in solution enabling antibodies coupled to the beads to continuously interact with the cell suspension. These 4.5 µm superparamagnetic beads experience a strong pull when placed in a magnetic field and gently pull targeted cells to the tube walls when the tube is transferred to a rack containing a strong magnetic field. With the beads and attached cells concentrated at the tube wall the supernatant liquid containing non-targeted cells is easily and quickly decanted or pipetted away. Washing steps are performed similarly.

Dynabeads™ Rat Anti-Mouse IgM (Invitrogen™)

Dynabeads® Rat anti-Mouse IgM binds mouse IgM of your choice for specific cell isolation or depletion. All cells from all species except from mouse can be isolated with this product, depending only on the specific antibody of choice.

• Depletion: cells can be depleted from a sample
• Positive isolation: cells can be positively isolated from a sample for molecular applications
• Negative isolation: cells can be negatively isolated from a PBMC sample by adding an antibody cocktail of mouse IgMs towards the unwanted cells. Compatible with flow cytometry.

Dynabeads™ Goat Anti-Mouse IgG (Invitrogen™)

Use with any mouse IgG for depletion, or for positive isolation of cells for molecular purposes. Any cell can be isolated from any samples from all species, except mouse and used in further molecular analysis.

• Deplete any target cell from all species, except mouse
• Isolate cells directly from whole blood for molecular applications
• Use a cocktail of mouse IgG's to deplete unwanted labeled cells and obtain untouched, bead and antibody-free cells.

This Product Contains: 5 ml Dynabeads® coated with goat polyclonal against all mouse IgG subclasses.

CELLection™ Biotin Binder Kit (Invitrogen™)

Use this kit with any biotinylated antibody for the positive isolation of a cell population including mouse cells. Biotinylated antibodies are attached to CELLection™ Dynabeads® via a DNA linker; this linker provides a cleavable site to remove the beads from the cells after isolation. Detached cells are viable and can be used in any downstream application.

This Product Contains: CELLection™ Dynabeads® coated with recombinant streptavidin and DNase Releasing Buffer

Applications:

• Positively isolate and detach any cell from any species using your own biotinylated antibody
• Compatible with flow cytometry

Dynabeads™ FlowComp™ Flexi Kit (Invitrogen™)

This gentle, easy-to-use and highly flexible kit allows you to positively isolate bead-free and flow-compatible cells in combination with your own antibody of choice. Now you can isolate practically any cell type from any species or starting sample using your own antibody. Immediately after positive isolation, the beads are released and removed from your cell sample. In this way, you avoid exposing your cells to potentially cytotoxic or immunogenic foreign substances like iron oxides or dextrans, a potential problem when working with biodegradable particles. In addition, your cells will not be exposed to the stress of being passed through a column.

The kit supplies you with everything you need for DSB-X™ biotinylation of your antibodies and subsequent isolation of your cell type of interest. This technology has been successfully used to isolate a number of cell subsets including human and mouse T cells, B cells, NK cells, monocytes, and endothelial cells.

Capacity:
The kit will process 2 x 109 mononuclear cells (MNC). Please note that the choice of antibody clone is of key importance for successful isolation (see package insert for technical recommendations).

Concentration:
Supplied at 15 mg beads per ml in phosphate buffered saline (PBS), pH 7.4, containing 0.1% bovine serum albumin (BSA) and 0.02% sodium azide.

Dynabeads™ Sheep-Anti Mouse IgG (Invitrogen™)

Dynabeads® Sheep anti-Mouse IgG binds monoclonal mouse IgG of your choice for specific cell isolation or depletion. All cells from all species except from mouse can be isolated with this product, depending only on the specific antibody of choice.

• Depletion: cells can be depleted from a sample
• Positive isolation: cells can be positively isolated from a sample for molecular applications
• Negative isolation: cells can be negatively isolated from a PBMC sample by adding an antibody cocktail of mouse IgGs towards the unwanted cells. Compatible with flow cytometry.

Note: For a more universal binding of your mouse IgG of any subclass, please use Dynabeads® Pan Mouse Ig (Cat.no. 110-41D or 110-42D).

Dynabeads™ Biotin Binder (Invitrogen™)

Dynabeads® Biotin Binder are 2.8 µm superparamagnetic beads that bind biotinylated antibodies (or other biotinylated ligands), allowing you to isolate cells from any species (e.g., human, mouse, rat, non-human primates, and cell lines).

• Works with any cell type, from any sample and from any species
• Fast and efficient depletion or positive isolation of any target cells for molecular downstream assays using a biotinylated primary antibody
• Obtain pure and untouched cells using a mixure of biotinylated antibodies to deplete unwanted cells

Adaptable for various isolation strategies
The biotinylated primary antibody may be added to the cell sample (indirect technique) or pre-coated onto the Dynabeads® Biotin Binder (direct technique). Regardless of which of these techniques is used, when the cell sample and the Dynabeads® Biotin Binder are mixed, the beads bind to the target cells. Placing the sample on a magnet separates the bead-bound cells from the rest of the sample. One or more biotinylated antibodies may be used to either deplete the sample of unwanted cells (negative isolation) or to capture specific cells for downstream experiments (positive isolation). For isolation of bead-free cells (e.g., for flow cytometry), we recommend using your biotinylated antibody in combination with the CELLection™ Biotin Binder Kit.

Useful for many sample types
Using Dynabeads® Biotin Binder, you can isolate cells basically from any species, and from many starting samples such as whole blood, cord blood, peripheral blood mononuclear cells (PBMC), buffy coat, mouse spleen/lymph nodes, and tissue digests.

Dynabeads™ M-450 Tosylactivated (Invitrogen™)

Dynabeads® M-450 Tosylactivated are 4.5 µm superparamagnetic beads containing surface tosyl groups. The beads covalently bind primary amino and sulfhydryl groups in antibodies to position them in the optimal orientation for cell-surface protein binding. Low background and covalent binding of the antibodies make these beads an excellent choice for cell separation applications when a downstream release from the beads is not required.

• Quickly deplete cells or positively isolate target cells for downstream molecular analysis
• Excellent for cell isolation procedures using your own unique antibody
• Several antibodies can be coupled on the same beads for non-cell-based applications, such as cell activation

Coupling procedure
Covalent coupling is performed overnight by incubating the antibody of choice with the Dynabeads® M-450 Tosylactivated beads. Optimal coupling occurs at high pH (8.5–9.5) and at 37°C. For pH-labile antibodies, coupling can be performed in an alternative buffer at pH 7.4.

Use this kit with any mouse IgG for the positive isolation of a cell population from all species except mouse. Antibodies are attached to the surface of the CELLection™ Dynabeads® via a DNA linker; this linker provides a cleavable site to release and remove the beads from the cells after isolation. Bead-free cells are viable and can be used in any downstream application.

This Kit Contains: CELLection™ Dynabeads® coated with a human anti-mouse IgG monoclonal, and DNase Releasing Buffer

Applications:

• Positively isolate and detach any cell from any species (except mouse) using your own mouse IgG

Dynabeads™ Sheep Anti-Rat IgG (Invitrogen™)

Dynabeads® Sheep Anti-Rat IgG are 4.5 µm superparamagnetic beads with affinity purified polyclonal sheep anti-rat IgG covalently bound to the bead surface. These beads can be used to isolate any cell from any species (except rat), depending on the specificity of the monoclonal rat IgG employed. Dynabeads® Sheep Anti-Rat IgG are especially well suited for cell isolation from mouse samples (cross-reactivity to mouse antibodies is high and cross-reactivity to human antibodies is minimal).

• Fast and efficient depletion or positive isolation of any target cell for molecular downstream assays using rat primary antibody
• Obtain pure and untouched cells using a mixture of rat IgGs to deplete unwanted cells

Magnetic bead-based separation offers easy handling
The rat primary antibody may be added to the cell sample (indirect technique) or pre-coated onto the Dynabeads® Sheep Anti-Rat IgG (direct technique). Regardless of which of these techniques is used, when the cell sample and the Dynabeads® Sheep Anti-Rat IgG are mixed, the beads bind to the target cells. Placing the sample on a magnet separates the bead-bound cells from the rest of the sample. One or more rat primary antibodies may be used to either deplete the sample of unwanted cells (negative isolation) or to capture specific cells for molecular downstream experiments (positive isolation).
