NorthernMax™ High Stringency Wash Buffer (Invitrogen™)

The equivalent of 0.1X SSC or SSPE, this Ambion® wash buffer is functionally tested by northern analysis and is certified RNase-free. Supplied in one bottle containing 1 L. NorthernMax® reagents are the same components included in the NorthernMax® Ultrasensitive Northern Blotting Kits but in larger sizes. ULTRAhyb™ Ultrasensitive Hybridization Buffer (Invitrogen™)

Ambion® ULTRAhyb® contains a unique blend of hybridization accelerators and blocking agents that greatly enhance the levels of hybridization so that signals that once took days to visualize are now apparent in hours. It is supplied in four bottles containing 125 mL each.

• Increase the sensitivity of any blot hybridization experiment
• Increase random-primed DNA probe sensitivity up to 100x; RNA probe sensitivity 20x
• Use less probe per blot
• Decrease hybridization time to just 2 hours

Using standard hybridization buffers, only 1–5% of target molecules on a blot hybridizes to probe, making blots a relatively insensitive method for nucleic acid analysis (Vernier et al. (1996) Anal Biochem 235: 11-19). With ULTRAhyb® Ultrasensitive Hybridization Buffer, the hybridization reaction approaches completion, so that as few as 10,000 molecules can be detected.

For Any Hybridization Protocol
ULTRAhyb® is compatible with RNA and DNA probes labeled isotopically and nonisotopically in northerns, Southerns, and dot/slot blots. It contains 50% formamide and is compatible with positively charged membranes (see Accessory Products). ULTRAhyb® requires 10-100x less nonisotopic DNA probe than standard hybridization buffers, making it possible to perform many more hybridizations with a single labeling reaction.

The BrightStar® -Plus Positively Charged Nylon Membranes (SKU# AM10100, AM10102, or AM10104) are recommended for use with ULTRAhyb®. Denhardt's Solution (50X) (Invitrogen™)

Denhardt's Solution is a mixture of blocking agents used in membrane-based hybridization protocols. The solution contains 1% Ficoll (type 400), 1% polyvinylpyrrolidone, and 1% bovine serum albumin. Sterile filtered.

ULTRAhyb™-Oligo (Invitrogen™)

Ambion® ULTRAhyb®-Oligo is specifically formulated for use with oligonucleotide probes in northern analysis. It contains a unique blend of hybridization accelerators and blocking agents that greatly enhance the levels of hybridization so that signals that once took days to visualize are now apparent in hours. It is supplied in one bottle containing 125 mL.

Oligonucleotides ranging in length from 26 to 45 are used with ULTRAhyb®-Oligo. It works well with oligonucleotides having Tms from 78°–90°C and 47–62% GC contents. However, the optimal hybridization temperature for oligonucleotide probes with different GC content and Tm values should be determined empirically.

Nucleic Acid Detection Blocking Buffer is intended for use with the Thermo Scientific Chemiluminescent Nucleic Acid Detection Module Kit, of which it is also a component. Non-specific binding of probe to membrane can be minimized by incubating the membrane in Blocking Buffer for 15 minutes.
