NorthernMax™ One-Hour Transfer Buffer (Invitrogen™)

The Ambion® NorthernMax® Transfer Buffer is faster and more efficient than standard, overnight transfer buffer. Transfer is complete in 1.5–2 hr. One bottle containing 1 L is provided. Transfer buffer is not compatible with nitrocellulose membrane. NorthernMax® reagents are the same components included in the NorthernMax® Ultrasensitive Northern Blotting Kits but in larger sizes. These reagents are supplied ready to use and are certified RNase-free. Comprehensive instructions are included.

NorthernMax™ Kit (Invitrogen™)

The Ambion® NorthernMax® Kit contains a complete set of RNase-free reagents for running formaldehyde-based northern analysis. This kit is ideal for the investigator who wants to use familiar reagents while taking advantage of NorthernMax® Kit's high sensitivity. Sufficient reagents are provided to process 1000 cm2 membrane.

The included ULTRAhyb® Ultrasensitive Hybridization Solution increases the sensitivity of RNA probes up to 20x and DNA probes up to 100x. Hybridization can be completed in just 2 hours for many messages. The NorthernMax® Kit is compatible with DNA, RNA, or oligo probes labeled isotopically or nonisotopically. The NorthernMax® Kit contains everything you need to conduct northerns. Positively charged nylon membranes must be purchased separately (see Accessory Products). An RNA sample and template for probe synthesis are also provided as controls for each kit.

NorthernMax™ Denaturing Gel Buffer (10X) (Invitrogen™)

An improved formula for preparing denaturing formaldehyde/MOPS agarose gels. Formaldehyde is included. Gels made with this buffer are considerably less brittle than conventional formaldehyde gels, making handling of the gels much easier. One bottle containing 250 mL is provided. NorthernMax® reagents are the same components included in the NorthernMax® Ultrasensitive Northern Blotting Kits but in larger sizes. These reagents are supplied ready to use and are certified RNase-free. Comprehensive instructions are included.

NorthernMax™ Formaldehyde Load Dye (Invitrogen™)

Ambion® NorthernMax® Formaldehyde Load Dye as ready-to-use solution is added to RNA sample (3 parts solution: 1 part sample) and heated briefly. The samples are then ready to be loaded. Ethidium bromide can be added to the samples if desired. Supplied in six tubes containing 1 mL each.

All Ambion® Gel Loading Solutions are rigorously tested for non-specific endonuclease activity, exonuclease activity, RNase activity, and for functionality. These are the same solutions we use in-house and in our kits.

NorthernMax™-Gly Kit (Invitrogen™)

The Ambion® NorthernMax®-Gly Kit is an alternative to formaldehyde-based systems for northern analysis and uses a glyoxal/DMSO Loading Solution for sample denaturation. Sufficient reagents are provided for processing 1000 cm2 membrane.

Use of glyoxal/DMSO instead of formaldehyde for sample preparation avoids the need to pour and run gels in a fume hood, and eliminates the safety issues associated with use of formaldehyde. Also, RNA samples denatured with glyoxal may show sharper bands on northern blots compared to samples denatured and run in the presence of formaldehyde. The streamlined NorthernMax®-Gly procedure is more convenient and faster than the standard glyoxal gel protocol. A single reagent is used for sample denaturation and loading, the incubation time for sample denaturation has been reduced, and no recirculation of buffer is required during electrophoresis. The volume ratio of glyoxal denaturation solution to sample RNA is lower than in other published protocols, so that sample precipitation prior to gel loading is usually not required. The NorthernMax®-Gly Kit can be used with either radiolabeled or nonisotopically labeled RNA or DNA probes. Each NorthernMax®-Gly Kit provides a complete system. Positively charged nylon membranes must be purchased separately (see Accessory Products). An RNA sample and template for probe synthesis are also provided as controls for each kit.

NorthernMax™ Prehybridization/Hybridization Buffer (Invitrogen™)

This buffer is functionally tested by northern blotting and is certified RNase-free. No BSA, Denhardt's, or other potentially nuclease-contaminated biological materials are used in preparing this buffer, ensuring maximum sensitivity and reproducibility. One bottle containing 500 mL is provided. The Ambion® NorthernMax® Prehybridization/Hybridization Buffer contains 50% formamide with proprietary blocking and background reduction agents. Blocking agents minimize cross hybridization to ribosomal sequences. Signal-to-noise ratios are increased by the background reducing agents' suppression of hybridization to the membrane, which is particularly important with nonisotopic detection protocols. NorthernMax® reagents are the same components included in the NorthernMax® Ultrasensitive Northern Blotting Kits but in larger sizes. North2South™ Complete Biotin Random Prime Labeling and Detection Kit (Thermo Scientific™)

The Thermo Scientific™ North2South™ Biotin Random Prime Labeling Kit provides the components and procedure to successfully prepare biotin-labeled DNA probes for use in Southern blotting and other DNA hybridization methods involving streptavidin detection systems.

This kit includes the North2South Chemiluminescent Detection Kit, which combines a novel ECL substrate for horseradish peroxidase (HRP) and optimized hybridization and blocking buffers to ensure consistent, high-sensitivity Southern blot results.

After a brief membrane blocking step, the biotin label is detected with streptavidin- HRP and a 5-minute incubation with the chemiluminescent substrate. Results are easily recorded by brief exposure to X-ray film (1-15 minutes) or CCD camera.

Chemiluminescent Nucleic Acid Detection Module Kit (Thermo Scientific™)

The Thermo Scientific Pierce Chemiluminescent Nucleic Acid Detection Module is a complete system for detection of biotin-labeled nucleic acid probes in various membrane blotting applications.

Nearly any DNA or RNA method based on nitrocellulose or nylon membranes and biotinylated probes can be detected with sensitivity equivalent to traditional radioactive (32P) systems. The module consists of the essential detection components used in the Thermo Scientific North2South Kit and Thermo Scientific LightShift EMSA Kit. Biotin Chromogenic Detection Kit (Thermo Scientific™)

Thermo Scientific Biotin Chromogenic Detection Kit is a convenient tool for the chromogenic detection of biotinylated nucleic acid probes. The kit is optimized to reproducibly provide high sensitivity with a low background in applications, such as Southern, Northern, dot and slot blotting, and screening of viral plaques and bacterial colonies. Biotinylation of DNA/RNA probes is widely used as a safe and convenient alternative to radioactive labeling. Biotin can be incorporated into nucleic acids using various enzymatic or non-enzymatic methods, including the Biotin DecaLabel DNA Labeling Kit.

Features

Biotinylated probes are detected with streptavidin coupled to alkaline phosphatase (AP). Streptavidin-AP conjugates bind specifically and irreversibly to the biotin-labeled probes. The probes are then visualized using a chromogenic substrate for alkaline phosphatase BCIP/NBT, which produces a blue-purple precipitate. Visualization does not require X-ray film or other specific equipment (see Figure 1).

North2South™ Chemiluminescent Substrate for HRP (Thermo Scientific™)

The Thermo Scientific North2South Chemiluminescent Substrate is used for detecting nucleic acids in northern and Southern blot applications. This robust system uses an enhanced luminol substrate for horseradish peroxidase (HRP) that greatly reduces processing and film exposure time. Post-hybridization processing has been reduced from the standard 2.5 hours to 1 hour. Film exposure times range from 30 seconds to 10 minutes with the substrate emitting light with relatively constant intensity for up to 6 hours, allowing for multiple exposures.
