MagMAX™ Saliva gDNA Wash I Solution Applied Biosystems™

PARIS™ Kit Invitrogen™

The Ambion® PARIS™ system is for the isolation of both RNA and native protein from the same experimental sample. The kit also permits separation of nuclear and cytoplasmic fractions prior to RNA and/or protein isolation. The kit contains sufficient reagents for 50 purifications from1 to 75 mg of tissue or 100 to 107 cells each.

Isolation of high-quality RNA is the first step for a variety of gene expression analyses. Very often, complementary studies at the protein level are also required. Usually these analyses are performed using different aliquots of the same experimental sample. However, when working with rare, difficult to obtain, or very small samples, it is sometimes impractical to isolate RNA and native proteins independently. In studies involving large numbers of samples, expensive reagents, or inherent variability (e.g., cell transfection), the addition of independent experimental samples is not only costly and time consuming, but may also lead to inconsistent results. To solve these issues, the Protein And RNA Isolation System (PARIS™ system) has been developed. It allows for the isolation of both RNA and protein from the same experimental sample (see Figure). Using the PARIS™ system, RNA and protein can be isolated simultaneously from whole cell lysates. Alternatively, RNA and protein can be isolated from separate nuclear and cytoplasmic fractions. Tissue or cultured cells are first homogenized in ice-cold Cell Disruption Buffer to prepare a total cell lysate. Since the homogenization is performed quickly on ice and in the presence of detergent, both protein and RNA can be purified directly from this lysate. For RNA isolation, a part of the total cell lysate is immediately mixed with an equal volume of Lysis/Binding Solution. Total RNA is then purified from the mixture using an RNA binding glass fiber filter. After three rapid washing steps, high-quality RNA is eluted in a concentrated form. The entire procedure can be completed in less than 20 minutes. Note: This kit is not recommended for tissues with high levels of ribonucleases, such as pancreas.

Compatible with Most Downstream Applications
The RNA isolated from total, nuclear, or cytoplasmic fractions with the PARIS™ procedure can be used in a variety of downstream applications, including blot hybridization, in vitro translation, cDNA synthesis, and RT-PCR. A DNase I treatment is recommended for RNA that will be used for RT-PCR experiments (see Accessory Products). Protein fractions can be used directly for most common applications, including functional assays, immunoprecipitation, western blotting, or two-dimensional gel electrophoresis (see Figures).

Accessory Products:
TURBO™ DNA-free™ or DNA-free™ DNase Treatment and Removal Reagents (SKU #AM1907 and AM1906, respectively) are ideal to quickly remove trace amounts of DNA from the total and nuclear RNA samples without phenol extraction or alcohol precipitation. The cytoplasmic RNA fraction is virtually free of DNA contamination.

Total Exosome RNA & Protein Isolation Kit Invitrogen™

The Total Exosome RNA and Protein Isolation Kit enables isolation of total RNA and proteins from exosome samples extracted from biological fluids (e.g., by using Total Exosome Isolation (from cell culture media) or Total Exosome Isolation (from serum)).

The Total Exosome RNA and Protein Isolation Kit is designed for the isolation of both RNA and protein from pre-isolated or enriched exosomes (not supplied). It is suitable for studies of RNA expression (specifically miRNA), processing, or function. The kit allows recovery of both RNA and proteins from the same purified exosome preparation. One portion of the sample is subjected to organic extraction, followed by immobilization of RNA on glass-fiber filters to purify total RNA or enrich for small RNA species. The other portion is then used directly for common protein applications such as Western blotting.

High yields of ultra-pure RNA can be prepared in about 30–60 minutes. The RNA can then be used in a variety of downstream applications, including qRT-PCR, high-throughput sequencing (e.g., using the Ion Torent™ PGM™, Ion Torrent™ Proton™, or SOLiD® sequencing instruments), microarray analysis, solution hybridization assays, and blot hybridization.

The MagMAX Saliva gDNA Isolation Kit is a scalable and automatable magnetic bead-based sample preparation technology for the isolation of genomic DNA (gDNA) from fresh and stabilized saliva. Isolate DNA from stabilized and freshly collected saliva
The MagMAX Saliva gDNA Isolation Kit isolates gDNA from 200 µL–2 mL of freshly collected saliva or stabilized saliva collected using a commercially available saliva collection device (see figure below). MagMAX Saliva gDNA isolation chemistry is compatible with the Oragene® saliva collection devices from DNA Genotek, the SalivaGard® saliva collection device from Biomatrica, and the GeneFiX™ Saliva DNA Collection Device from IsoHelix.

The MagMAX Saliva gDNA Isolation Kit maximizes gDNA yields by treating samples with an innovative lysis/binding solution to simultaneously lyse cells, denature proteins, and capture gDNA on high-capacity magnetic binding beads. The Saliva gDNA Wash I Solution removes proteins, detergents, and other impurities from the bound nucleic acid. The gDNA is then released from the beads using a low-TE elution solution.

Fast and hassle-free magnetic bead-based workflows
The MagMAX Saliva gDNA Isolation Kit combines multiple laborious sample processing steps together into a single step to minimize the time and effort typically required by other methods, without compromising yield and purity. Process a broad range of sample volume inputs simultaneously without sample normalization or reagent volume adjustments for hassle-free sample prep. Simply match your saliva input volumes to one of three standard input workflows (50 µL to 500 µL, up to 1 mL, up to 2 mL) and follow the instructions for semi-automated sample preparation using the KingFisher Duo Prime or KingFisher Flex purification systems.

Optimized for KingFisher purification systems
The MagMAX Saliva gDNA Isolation Kit is optimized for use on the moderate-throughput KingFisher Duo Prime (Cat. No. 5400110) and the higher-throughput KingFisher Flex (Cat. No. 5400630) purification systems. Users can process up to 500 µL of saliva in high-throughput capacity SBS 96-well deep-well microtiter plates using the KingFisher Flex system. For smaller sample batches, users can process 12 samples using the Duo Prime system. Larger sample input volumes (>500 µL, and 1–2 mL), which are more challenging to process, are purified in the larger sample volume capacity 24-well deep-well SBS plates for 24 samples on the Flex or 6 samples on the Duo Prime system. KingFisher instrument BindIt software scripts for all sample types and both Flex and Duo Prime systems can be downloaded below and installed onto the KingFisher instruments.

Yeast DNA Extraction Kit Thermo Scientific™

Thermo Scientific Pierce Yeast DNA Extraction Reagent Kit utilizes Thermo Scientific Y-PER Reagent to quickly and easily lyse yeast cells to release DNA for study.

Features of Yeast DNA Extraction Kit:

This kit provides greater yields than traditional methods for DNA isolation from yeast. Typically, extraction and purification of DNA from yeast are time-consuming and labor-intensive. The yeast proteinaceous cell wall is notoriously difficult to lyse and requires harsh treatments that are time-consuming and can damage the extracted DNA. Yeast DNA Extraction Kit protocol requires less than one hour to complete, is effective without enzymatic treatment or glass beads, and yields little to no RNA contamination regardless of RNase presence. In studies with Saccharomyces cerevisiae, high yields of genomic and plasmid DNA are consistently obtained. DNA purified using this kit is suitable for PCR amplification, bacterial transformations, restriction digestions and hybridization applications.

mirVana™ PARIS™ RNA and Native Protein Purification Kit Invitrogen™

The mirVana™ PARIS™ Kit is for the purification of both native protein and RNA (including small RNA) from the same sample. Each kit contains sufficient reagents and consumables for either 40 isolations of protein and total RNA (including small RNAs), or 20 isolations of protein and separate large and small RNA fractions. Features of this kit:

• Isolate small RNA-containing total RNA and native protein from the same sample
• Start with 100–107 cultured cells or up to 100 mg of many mammalian tissues
• Enrich for small RNA (<200 nt) to increase sensitivity in downstream analyses
• Follow a simple, 30-minute procedure

How the kit works
The mirVana™ PARIS™ Kit procedure begins with homogenization of samples with a special cell disruption buffer that includes nonionic detergent. Protein remains intact, so a portion of the lysate can be used directly for common protein analysis applications. The remainder of the lysate is used for RNA isolation, using a procedure that combines the advantages of organic and solid-phase extraction, while avoiding the disadvantages of both.

Accessory products
For tissues that have very high levels of ribonucleases, such as pancreas or spleen, the mirVana™ miRNA Isolation Kit is recommended, although it is not designed to recover protein and small RNA from the same sample.
