AccuCheck Counting Beads (Invitrogen™)

AccuCheck Counting Beads are an efficient single-platform method for absolute cell counts that combines the advantages of direct flow cytometric immunophenotyping with the use of two different fluorescent beads (A and B beads). These two fluorospheres are used as a double internal standard for blood volume calculation. A known volume of AccuCheck Counting Beads is added to the same known volume of stained blood in a lyse-no-wash technique. The beads are counted along with cells. Since the concentration of beads is known, the number of cells per microliter (the absolute count) is obtained by relating the number of cells counted to the total number of fluorescent bead events. The cell number is then multiplied by the number of total fluorospheres per unit of volume. As the AccuCheck Counting Beads system contains two different fluorospheres in a known proportion, we can first assure the accuracy of the assay by verifying the proportion of both types of beads.

How it works

The emission profiles for these standards are intentionally narrow in comparison to cells labeled with a corresponding fluorescent dye. Due to their narrow emission profiles, beads of two different fluorescent colors exhibit minimal spectral overlap, and little or no color compensation is needed when setting up for multicolor experiments.

Ready-to-use control
Unlike other methods that require sample preparation to make compensation controls using cellular material, the ArC™ Amine Reactive Compensation Bead Kit is ready to use out of the dropper bottle. The kit includes two types of specially modified polystyrene microspheres that have been optimized to react with the amine-reactive dye in the LIVE/DEAD™ Fixable dead cell stain kits. The ArC™ reactive beads (Component A) bind any of the amine-reactive dyes and provide a positive signal; the ArC™ negative beads (Component B) have no reactivity and provide a negative compensation control. This ready-to-use product preserves the original sample material for experiments; precious sample is not used for the creation of compensation controls.

Accurate and consistent results
Unlike biological controls, which can have variable expression levels depending upon disease or development state, the Arc™ beads provide a bright, consistent signal (see figure below). Consistency in the staining pattern of compensation controls is important for obtaining the right compensation value. If the signal varies with the control material used, the compensation value can be affected, leading to erroneous results.

How it works
One drop of beads is added to a test tube containing LIVE/DEAD™ Fixable dead cell stain. After incubation with amine-reactive dye, the beads are washed in staining buffer, a drop of negative control beads is added (if required), and the beads are resuspended and analyzed by flow cytometry. The two components provide a distinct positive bead that is at least 50-fold brighter than the negative beads, which can be used to set compensation.

CountBright™ Absolute Counting Beads, for flow cytometry (Invitrogen™)

CountBright™ absolute counting beads are a calibrated suspension of microspheres that are brightly fluorescent across a wide range of excitation and emission wavelengths (UV to 635 nm excitation and 385-800 nm emission). CountBright™ absolute counting beads are mixed with the cell sample and assayed via flow cytometry. By comparing the ratio of bead events to cell events, absolute numbers of cells in the sample can be calculated. Because CountBright™ beads are mixed in the test sample, absolute cell counts using this single-platform method are more accurate and less complicated than cell concentration determined using multiple-platform testing. CountBright™ absolute counting beads can be used with any sample type, including no-wash/lysed whole blood.

Flow Cytometry Size Calibration Kit (nonfluorescent microspheres) (Invitrogen™)

Our Flow Cytometry Size Calibration Kit has nonfluorescent particle-size calibration standards that provide a simple, accurate way to determine cell sizes by flow cytometry. The kit contains six suspensions of highly uniform polystyrene microspheres with the following diameters: 1.0 µm, 2.0 µm, 4.0 µm, 6.0 µm, 10.0 µm and 15.0 µm. All of the microsphere suspensions are provided in convenient dropper vials.
