The InSpeck™ Red Microscopy Image Intensity Calibration Kit provides microsphere standards that generate a series of well-defined fluorescent intensity levels for constructing calibration curves and evaluating sample brightness. Each kit includes six separate suspensions of 2.5 µm fluorescent microspheres with relative fluorescence intensities of 100%, 30%, 10%, 3%, 1% and 0.3% covering the range of intensities commonly encountered in microscopy applications. Unstained control beads and mounting medium are also supplied. The aqueous suspensions of microspheres may be applied directly to the sample for calibrating fluorescence intensities or mounted separately in an adjacent well or on another slide.

MeltDoctor™ HRM Calibration Plate, Fast 96-well (Applied Biosystems™)

Applied Biosystem® ready-to-use MeltDoctor™ HRM Calibration Plates contain all the components required for pure dye and HRM calibration, reducing the complexity of getting started with HRM experimentation. Instrument Control Terbium TR-FRET Kit

The Instrument Control Terbium TR-FRET Kit allows for the rapid assessment of instrument setup for terbium (Tb)-based TR-FRET assays. LanthaScreen® and Premo™ assays require specific instrument settings. Proper instrument setup is critical to experimental success. Determine if Your Instrument is Set Up to Read Tb TR-FRET Assays
Setting up your instrument to read Tb TR-FRET is the first step in running a successful Tb TR-FRET assay. This kit allows you to quickly determine if your instrument is set up properly to read Tb TR-FRET assays. Simply add a small volume into separate wells, place the plate into the instrument and read. If the ratio of high to low control signal is below 2 the instrument may not be setup properly. If the ratio of high to low control signal is between 2 and 4 you can proceed with a Tb TR-FRET.

Use the Kit as a Positive Control in Tb TR-FRET Assays
When running Tb TR-FRET assays, add a small volume of the high and low controls to separate wells, on your assay plate. Place the plate into the instrument and read, according to instructions. A high to low control signal ratio of between 2 and 4 offers you the confidence that the instrument is performing well.

Cell Sorting Set-up Beads (for green-yellow lasers) (Invitrogen™)

Molecular Probes® Cell Sorting Set-up Beads are reliable standards for flow cytometry sorter instrument setup and calibration. The Cell Sorting Set-up Beads approximate the size, emission wavelength, and intensity of many biological samples, and so can be used to calibrate a flow cytometer’s cell sorting system, including laser source, optics, and stream flow. The beads are optimized for checking cell sorter settings such as drop delay and efficiency (cell loss during sorting).

Cell Sorting Set-up Beads are available for use with UV, blue, green/yellow, and red lasers.These 6.0 µm beads for green/yellow lasers are fluorescent-dye infused microspheres that are ideally excited at 570 nm with emission of 575 nm, but may also be exited at 488 nm.

Bead Labeling
As with all of our flow cytometry standard microspheres, the Cell Sorting Set-up Beads are stained internally rather than on the surface. The dyes are therefore insulated from environmental interactions that could cause variable fluorescence output, resulting in excellent signal stability. These 6.0 µm fluorescent polystyrene microspheres are supplied as suspensions packaged in dropper vials for convenient dispensation and are available in a choice of four colors (UV, blue, green/yellow, red) to match your laser.

The dyes used in the manufacture of the Cell Sorting Set-up Beads have been carefully selected to provide emission peaks coincident with cells labeled with commonly used fluorescent dyes. The emission profiles for these standards are intentionally narrow in comparison to cells labeled with a corresponding fluorescent dye. Due to their narrow emission profiles, beads of two different fluorescent colors exhibit minimal spectral overlap, and little or no color compensation is needed when setting up for multicolor experiments.

Accurate compensation is crucial for effective multicolor analysis in flow cytometry. Often, however, cellular-based compensation controls are not completely effective, as many antigens are not highly expressed, and dimly stained cells can lead to inaccurate compensation settings. The AbC™ Anti-Rat/Hamster Bead Kit was developed to address this issue.

How it works
Each kit provides polystyrene microspheres that have fluorescent antibody conjugate capture capacity (positive compensation beads) or are inert (negative compensation beads). After mixing with a fluorophore-conjugated rat or hamster antibody, the two components provide a distinct high-signal positive control with an appropriate negative population that can then be used to set compensation properly regardless of the intensity of the cells in the actual experiment.

The AbC™ Anti-Rat/Hamster Bead Kit is ideal for compensation with all isotypes of rat (IgG1, IgG2a, IgG2b, IgG2c, IgM) and hamster (Armenian IgG and Syrian IgG) immunoglobulins.

Maximum laser compatibility
All AbC™ Compensation Bead Kits are ideal for use with any excitation source, including the 405 nm laser which has historically provided higher background signals due to auto-fluorescence with other kits on the market.

Ion S5™ ExT Calibration Standard (Ion Torrent™)

The Ion S5™ ExT Calibration Standard is a small panel consisting of known sequence content with comprehensive and uniform representation of long homopolymers up to 10-mers. The Ion S5 ExT Calibration Standard can be used in conjunction with the Ion 520™ & Ion 530™ ExT workflow for de novo research sequencing applications to allow for greater accuracy of base-calling for templates between 400 and 600 bp.

GeneScan™ 400HD ROX™ dye Size Standard (Applied Biosystems™)

The GeneScan™ 400HD ROX™ Size Standard is a ROX™ dye-labeled size standard for the reproducible sizing of fragment analysis data. The standard contains 21 ROX™ dye-labeled DNA fragments.

GeneScan™ 400 HD ROX™ Size Standard is designed for sizing DNA fragments in the 50-400 nucleotides range and provides 21 labeled fragments of: 50, 50, 90, 100, 120, 150, 160, 180, 190, 200, 220, 240, 260, 280, 290, 300, 320, 340, 360, 380 and 400 nucleotides. The sizing curve generated from these fragments make the GeneScan™ 400 HD ROX™ Size Standard ideal for a variety of fragment analysis applications such as Microsatellites, Fragment Length Polymorphisms and Relative Fluorescent Quantitation. Each of the DNA fragments is labeled with the ROX™ fluorophore.

Matrix Standard Set DS-02 for the 310 Genetic Analyzer (Applied Biosystems™)

The dye-labeled oligonucleotides included in the Matrix Standard Set DS-02 (dR110, dR6G, dTAMRA™, dROX™, and LIZ® dyes) for the 310 system are used to generate the multicomponent matrix required for the SNaPshot® Multiplex System on the 310 Genetic Analyzer.

The 310 Data Collection software then utilizes the multicomponent matrix to automatically correct for the spectral overlap in samples labeled with DS-02 dyes. Matrix standards do not need to be run with every set of sample injections. This set of five matrix standards only needs to be run once in order to generate a matrix file which is then applied to samples run under similar conditions. For more information on the use of matrix standards, please refer to the instrument User's Manual.

This matrix standard is to be used on 310 Genetic Analyzer only.

AccuCheck Counting Beads (Invitrogen™)

AccuCheck Counting Beads are an efficient single-platform method for absolute cell counts that combines the advantages of direct flow cytometric immunophenotyping with the use of two different fluorescent beads (A and B beads). These two fluorospheres are used as a double internal standard for blood volume calculation. A known volume of AccuCheck Counting Beads is added to the same known volume of stained blood in a lyse-no-wash technique. The beads are counted along with cells. Since the concentration of beads is known, the number of cells per microliter (the absolute count) is obtained by relating the number of cells counted to the total number of fluorescent bead events. The cell number is then multiplied by the number of total fluorospheres per unit of volume. As the AccuCheck Counting Beads system contains two different fluorospheres in a known proportion, we can first assure the accuracy of the assay by verifying the proportion of both types of beads.

PS-Speck™ Microscope Point Source Kit (blue, green, orange & deep-red fluorescent beads) (Invitrogen™)

The PS-Speck™ Microscope Point Source Kit contains four different colors of fluorescence microspheres; each microsphere has a diameter of 0.175 +/- 0.005 µm. This exceptionall small diameter coefficient of variation makes the PS-Speck™ microspheres ideal as uniform, subresolution fluorescent point sources for calibrating instrument optics, especially in three-dimensional imaging applications. This kit's four ready-to-use 1 mL suspensions contain bright, monodisperse microspheres with excitation/emission wavelengths of 360/440 nm (blue), 505/515 nm (green), 540/560 nm (orange) and 633/660 nm (deep red). The Thermo Scientific™ Pierce™ BSA Protein Digest Standard, LC-MS Grade, is a validated protein digest optimized for use as a quality control sample for liquid chromatography (LC) and mass spectrometry (MS) analysis of proteomic samples.

The peptide digest standard provides excellent sequence coverage with minimal over-alkylation and missed cleavages. The TetraSpeck™ Fluorescent Microspheres Sampler Kit contains samples of the 0.1 µm, 0.5 µm, and 4.0 µm TetraSpeck™ beads. Quantitative proteomic strategies using TMT reagents enable sample multiplexing and precise measurement of protein abundance. However, successful execution of this workflow includes multiple steps that may require optimization, including chromatography, mass spectrometry (MS), and data analysis. A critical parameter for success in this workflow is the ability to consistently detect the TMT fragment ions across the 11 channels of multiplexing. This standard has been specifically designed as a tool to monitor LC-MS/MS system performance for TMT quantitation and MS method optimization and validation.

The Pierce TMT11plex Yeast Digest Standard consists of four congenic strains of Saccharomyces cerevisiae (a parental line, BY4741, and three lines lacking the non-essential proteins Met6, His4, or Ura2) labeled in triplicate (knock-out strains) or duplicate (parental strain) with TMT11-plex label reagents (see figure). The knock-out strains are used in combination with the parental strain to enable comparison of peptide and protein abundance in the deleted and parental strains to monitor system performance for quantitation.

In addition to routine quality control assessment, the standard can also be used as a method development tool to measure the accuracy, precision, and dynamic range of different MS approaches, including optimization of offline fractionation, chromatography, MS acquisition, or data analysis settings.

The multifunctional applications for this standard will allow more reproducible TMT quantitation from day-to-day and experiment-to-experiment.

How it works
Each kit provides polystyrene microspheres that have fluorescent antibody conjugate capture capacity (positive compensation beads) or are inert (negative compensation beads). After mixing with a fluorophore-conjugated mouse, rat, hamster, or rabbit antibody, the two components provide a distinct high-signal positive control with an appropriate negative population that can then be used to set compensation properly regardless of the intensity of the cells in the actual experiment.

Attune™ Performance Tracking Beads

The Attune Performance Tracking Beads are designed for use with Attune Cytometric Software and Attune NxT Software to automatically characterize, track, and report the performance of the Attune Acoustic Focusing Cytometer and Attune NxT Acoustic Focusing Cytometer daily and over time. Regular calibration of your instrument with these beads will ensure the accuracy and sensitivity of the instrument and allow easy identification of potential shifts or trends in instrument performance. The tracking beads contain distinct sub-populations of beads labeled at different intensities, and are used to establish optimal dynamic range settings for the instrument. The beads are pre-calibrated at the factory and the lot-specific bead performance data captured as a downloadable file for use by the Attune Cytometric/Attune NxT software. Each vial contains enough beads for 50 measurements (Attune) or 25 measurements (Attune NxT). The beads are provided at a concentration of 5 × 106 beads/mL.

TaqMan™ RNase P Instrument Verification Plate, 96-well (Applied Biosystems™)

The TaqMan® RNase P Instrument Verification Plate is used to verify the performance of the Applied Biosystems® 7900HT 96-Well Real-Time PCR System and the ABI PRISM® 7000 and 7700 Sequence Detection Systems. The TaqMan® RNase P Instrument Verification Plate is a 96-well PCR plate preloaded with the necessary reagents to detect and quantitate genomic copies of the human RNase P gene, a single-copy gene encoding the RNA moiety of the RNase P enzyme. Each well contains preloaded reaction mix (1X TaqMan® Universal PCR Master Mix, RNase P primers, and FAM™ dye–labeled probe) and template. The TaqMan® RNase P Instrument Verification Plate satisfies the same requirements as the Applied Biosystems® 7900HT 96-Well Real-Time PCR System, and the ABI PRISM® 7000 and 7700 Sequence Detection Systems installations. To pass installation, the instruments must demonstrate the ability to distinguish between 5,000 and 10,000 genomic equivalents with a 99.7% confidence level for a subsequent sample run in a single well. To ensure accurate results, Applied Biosystems® recommends that the performance of your instrument be verified every six months.

31xx, 3500, and SeqStudio™ Matrix Standards Kit, BigDye™ Terminator v3.1 (Applied Biosystems™)

This kit contains two tubes of standards used to generate a spectrum on the 3100, 3100-Avant, 3130, 3130xl, 3500, 3500xl, and SeqStudio genetic analyzers. Each tube contains a blend of four different colors of fluorescent dye-labeled DNA fragments.

Calibration Install Kit, 384-well (Applied Biosystems™)

This kit is used to maintain your ViiA™ 7, QuantStudio™ 6 Flex, QuantStudio™ 7 Flex, or QuantStudio™ 12K Flex system with 384-well block. It includes calibration plates to perform a spectral calibration with FAM™, VIC®, ROX™, SYBR® Green, TAMRA™, and NED™ dyes, as well as plates to perform region-of-interest (ROI) calibration, normalization calibration, and RNase P verification.
