This easy-to-use dye calibration kit establishes the pure dye spectra and multicomponent values for FAM™, VIC™, ROX™, SYBR™ , ABY™, JUN™, MUSTANG PURPLE™, TAMRA™, NED™, and Cy®5 dyes on the Applied Biosystems™ QuantStudio™ 3 and 5 96-well 0.1-mL real-time PCR systems.

The QuantStudio™ 3/5 10-Dye Spectral Calibration Kit contains the following:
• 96-well 0.1-mL Spectral Calibration Plate 1 (containing FAM, VIC, ROX, and SYBR dyes)
• 96-well 0.1-mL Spectral Calibration Plate 2 (containing ABY, JUN, and MUSTANG PURPLE dyes)

Ready-to-use control
Unlike other methods that require sample preparation to make compensation controls using cellular material, the ArC Amine Reactive Compensation Bead Kit is ready to use out of the dropper bottle. The kit includes two types of specially modified polystyrene microspheres that have been optimized to react with the amine-reactive dye in the LIVE/DEAD Fixable dead cell stain kits. The ArC reactive beads (Component A) bind any of the amine-reactive dyes and provide a positive signal; the ArC negative beads (Component B) have no reactivity and provide a negative compensation control. This ready-to-use product preserves the original sample material for experiments; precious sample is not used for the creation of compensation controls.

Accurate and consistent results
Unlike biological controls, which can have variable expression levels depending upon disease or development state, the Arc beads provide a bright, consistent signal (see figure below). Consistency in the staining pattern of compensation controls is important for obtaining the right compensation value. If the signal varies with the control material used, the compensation value can be affected, leading to erroneous results.

How it works
One drop of beads is added to a test tube containing LIVE/DEAD Fixable dead cell stain. After incubation with amine-reactive dye, the beads are washed in staining buffer, a drop of negative control beads is added (if required), and the beads are resuspended and analyzed by flow cytometry. The two components provide a distinct positive bead that is at least 50-fold brighter than the negative beads, which can be used to set compensation.

Besides the ArC Amine Reactive Compensation Beads, AbC™ Anti-Mouse Bead Kit is also available for capture of mouse anti-human antibodies, as well as the AbC™ Anti-Rat/Hamster Bead Kit for capture of rat anti-mouse or hamster anti-mouse antibodies.

TaqMan™ RNase P Instrument Verification Plate, Fast 48-well (Applied Biosystems™)

Applied Biosystems® TaqMan® RNase P Fast 48-Well Instrument Verification Plate is used to verify the performance of the Applied Biosystems® StepOne™ Real-Time PCR System. The TaqMan® RNase P Fast 48-Well Instrument Verification Plate is a 48-well PCR plate preloaded with the necessary reagents to detect and quantitate genomic copies of the human RNase P gene, a single-copy gene encoding the RNA moiety of the RNase P enzyme. Each well contains preloaded reaction mix (1X TaqMan® Universal PCR Master Mix, RNase P primers, and FAM™ dye–labeled probe) and template. The TaqMan® RNase P Fast 48-Well Instrument Verification Plate satisfies the same requirements as the Applied Biosystems® StepOne™ Real-Time PCR Systems installation. To pass installation, the instrument must demonstrate the ability to distinguish between 5,000 and 10,000 genomic equivalents with a 99.7% confidence level for a subsequent sample run in a single well. To ensure accurate results, Applied Biosystems® recommends that the performance of your instrument be verified every six months.

Pierce™ 6 Protein Digest, equimolar, LC-MS grade (Thermo Scientific™)

The Thermo Scientific™ Pierce™ 6 Protein Digest Standard, Equimolar, LC-MS Grade, is a highly validated equimolar protein digest mixture for use as a quality control sample for mass spectrometry (MS) analysis of proteomic samples.

Positive control sample—optimized and validated as a quality control standard for mass spectrometry applications
Excellent sequence coverage—on average, greater than 70% sequence coverage of the 6-protein mixture
Validated peptide quality—digestion procedure optimized for minimal missed cleavages and over-alkylation

The Pierce 6 Protein Digest Standard contains an equimolar mixture of highly pure bovine cytochrome c, lysozyme, alcohol dehydrogenase, bovine serum albumin, apo-transferrin, and beta-galactosidase. The proteins are reduced, alkylated (iodoacetic acid), digested (MS-grade trypsin), and desalted (C18) after trypsin digestion to provide a quality LC/MS-grade standard free of intact protein. The peptide digest provides excellent sequence coverage with minimal over-alkylation and missed cleavages. This digest has been robustly tested for peptide quality, digestion efficiency, and lot-to-lot uniformity and is suitable for LC and LC-MS applications.

AbC™ Anti-Mouse Bead Kit (Invitrogen™)

The AbC™ Anti-Mouse Antibody Compensation Bead Kit provides a consistent, accurate, and simple-to-use technique for the setting of flow cytometry compensation when using fluorochrome-conjugated mouse antibodies with any laser in flow cytometry.

Accurate compensation is crucial for effective multicolor analysis in flow cytometry. Often, however, cellular-based compensation controls are not completely effective, as many antigens are not highly expressed, and dimly stained cells can lead to inaccurate compensation settings. How it works
Each kit provides polystyrene microspheres that have fluorescent antibody conjugate capture capacity (positive compensation beads) or are inert (negative compensation beads). After mixing with a fluorophore-conjugated mouse antibody, the two components provide a distinct high-signal positive control with an appropriate negative population that can then be used to set compensation properly regardless of the intensity of the cells in the actual experiment.

Species reactivity
The AbC™ Anti-Mouse Bead Kit is ideal for compensation with all isotypes of mouse (IgG1, IgG2a, IgG2b, IgG3, IgM) immunoglobulins.

Maximum laser compatibility
All AbC™ compensation bead kits are ideal for use with any excitation source, including the 405 nm laser which has historically provided higher background signals due to auto-fluorescence with other kits on the market.

CHCA MALDI Matrix, Single-Use (Thermo Scientific™)

The Thermo Scientific CHCA MALDI Matrices are highly purified, recrystallized reagents packaged in convenient single-use tubes sufficient for analyzing up to 96 samples by matrix assisted laser desorption ionization.

Matrix assisted laser desorption ionization (MALDI) matrices produce the cleanest mass spectrometric (MS) spectra when they are recrystallized and prepared daily, which is inconvenient and typically results in significant waste. The unique packaging of purified, recrystallized alpha-cyano-4-hydroxy-cinnamic acid (CHCA) MALDI matrices makes it easy to prepare high-quality MALDI reagents in minutes.

MALDI and electrospray ionization (ESI) are the most common methods to ionize peptides for MS analysis. MALDI is widely used because it is significantly faster than ESI and the ionized molecules are almost exclusively singly charged, making interpretation easier. Also, MALDI is readily automated and highly sensitive.
