DS37 Matrix Standard Kit (Dye Set J6-T) (Applied Biosystems™)

The DS-37 Matrix Standard Kit (Dye Set J6-T) is used to perform the spectral calibration required to analyze 6-FAM™, VIC™, TED™, SID™, TAZ™, and LIZ™ dye-labeled DNA fragments on the Applied Biosystems 3130 and 3500 series systems. The data collection software for these instruments uses the multi-component matrix to automatically analyze the 6 differently colored fluorescent dye-labeled samples in a single capillary. The DS-37 Matrix Standard Kit (Dye Set J6-T) contains 6 specific sizes of a unique fluorescent dye label.

QuantStudio™ 3/5 Spectral Calibration Plate 3 (TAMRA™, NED™, Cy™5 dyes), 96-well, 0.2 mL (Applied Biosystems™)

This plate establishes the pure dye spectra and multicomponent values for TAMRA™, NED™, and Cy®5 dyes on Applied Biosystems™ QuantStudio™ 3 and 5 96-well 0.2-mL real-time PCR systems. It enables simultaneous multiple dye calibrations in one plate, significantly reducing effort and time required for dye calibration. TetraSpeck™ Microspheres, 0.2 µm, fluorescent blue/green/orange/dark red (Invitrogen™)

The 0.2 µm TetraSpeck™ microspheres are stained throughout with four different fluorescent dyes, yielding beads that each display four well-separated excitation/emission peaks - 360/430 nm (blue), 505/515 nm (green), 560/580 nm (orange) and 660/680 nm (dark red). These microspheres can greatly facilitate the calibration of conventional fluorescence microscopes, confocal laser scanning microscopes, and associated image-processing equipment for both scientific and commercial imaging, especially for multicolor applications.

DS-33 GeneScan™ Installation Standards with GeneScan™ 600 LIZ™ Size Standard v2.0 (Applied Biosystems™)

The DS-33 GeneScan™ Installation Standard consists of pooled PCR products labeled with 6-FAM™, VIC®, NED™ and PET® dyes. To generate the pooled products, CEPH individual 1347-02 genomic DNA has been amplified with 8 fluorescent PCR primer pairs which amplify selected microsatellite loci (as listed in the package product insert). The kit also contains a tube of GeneScan™ 600 LIZ™ Size Standard 2.0.

Matrix assisted laser desorption ionization (MALDI) matrices produce the cleanest mass spectrometric (MS) spectra when they are recrystallized and prepared daily, which is inconvenient and typically results in significant waste. The unique packaging of purified, recrystallized 2,5-dihydroxybenzoic acid (DHB) MALDI matrices makes it easy to prepare high-quality MALDI reagents in minutes.

MALDI and electrospray ionization (ESI) are the most common methods to ionize peptides for MS analysis. MALDI is widely used because it is significantly faster than ESI and the ionized molecules are almost exclusively singly charged, making interpretation easier. Also, MALDI is readily automated and highly sensitive. ViiA™ 7 Array Card RNaseP Verification Kit (Applied Biosystems™)

The ViiA™ 7 System TaqMan® Low Density Array Instrument Verification RNase P kit is used to validate the performance of the Applied Biosystems® ViiA™ 7 Real-Time PCR Systems with the TaqMan® Low Density Array Block Upgrade.

The ViiA™ 7 System TaqMan® Low Density Array Instrument Verification RNase P kit contains an empty TaqMan® Low Density Array and 8 vials of RNase P detection reagents to detect and quantitate genomic copies of the human RNase P gene, a single-copy gene encoding the RNA moiety of the RNase P enzyme. Each vial contains premixed reagents (1X TaqMan® Universal PCR Master Mix, RNase P primers, and FAM™ dye-labeled probe) and template.

The ViiA™ 7 System TaqMan® Low Density Array Instrument Verification RNase P kit must demonstrate the ability to distinguish between 5,000 and 10,000 genomic equivalents with a 99.7% confidence level for a subsequent sample run in a single well. Pierce™ Peptide Retention Time Calibration Mixture (Thermo Scientific™)

The Thermo Scientific Pierce Peptide Retention Time Calibration Mixture enables reversed phase liquid chromatography (LC) and mass spectrometry (LC-MS) users to optimize and confirm correct operation of their equipment.

Features of Peptide Retention Time Calibration Mixture:

• Assess chromatography and MS instrument performance
• Predict peptide retention across multiple instrument platforms
• Predict peptide retention time from sequence using calculated hydrophobicity factor

The Pierce Peptide Retention Time Calibration Mixture is used to optimize and assess LC parameters, identify the total peptide elution window, and optimize MS parameters. Testing the mixture frequently throughout the lifetime of a column or including it in quantitative samples makes it possible to monitor and normalize LC-MS performance between samples and over time. When used with Thermo Scientific Pinpoint 1.1 software, the calibration mixture can predict peptide retention times based upon peptide sequence or relative retention time between instruments. The mixture can also be used with Pinpoint software to establish normalized collision energies between platforms. This allows normalization of methods and eases the transfer of quantitative methods between LC-MS platforms. The Pierce Peptide Retention Time Calibration Mixture contains 15 synthetic heavy peptides mixed at an equimolar ratio that elute across the chromatographic gradient. The observed retention times and hydrophobicity factors (HF) for these calibrant peptides are fit to a linear equation to determine the slope of the retention time/HF relationship. This equation and the HF of an uncharacterized peptide are then used to predict retention time.

The prediction of peptide retention time assists in the development of multiplexed, high throughput mass spectrometric assays. The Pierce Peptide Retention Time Calibration Mixture and Thermo Scientific Pinpoint software streamline the transition from qualitative protein discovery results to the development of targeted MS assays on Thermo Scientific triple quadrupole, Orbitrap, Exactive, and ion trap mass spectrometers. The Pierce Peptide Retention Time Calibration Mixture permits optimization and regular assessment of chromatographic performance and for rapid development of multiplexed, scheduled targeted MS assays for the quantification of dozens to hundreds of peptide targets per run.

Cell Sorting Set-up Beads (for UV lasers) (Invitrogen™)

Molecular Probes® Cell Sorting Set-up Beads are reliable standards for flow cytometry sorter instrument setup and calibration. The Cell Sorting Set-up Beads approximate the size, emission wavelength, and intensity of many biological samples, and so can be used to calibrate a flow cytometer’s cell sorting system, including laser source, optics, and stream flow. The beads are optimized for checking cell sorter settings such as drop delay and efficiency (cell loss during sorting).

Cell Sorting Set-up Beads are available for use with UV, blue, green/yellow, and red lasers.These 6.0 µm beads for UV lasers are fluorescent-dye infused microspheres that are excitable between 350–370 nm with emission in the range of 400–470 nm.

Bead Labeling
As with all of our flow cytometry standard microspheres, the Cell Sorting Set-up Beads are stained internally rather than on the surface. The dyes are therefore insulated from environmental interactions that could cause variable fluorescence output, resulting in excellent signal stability. These 6.0 µm fluorescent polystyrene microspheres are supplied as suspensions packaged in dropper vials for convenient dispensation and are available in a choice of four colors (UV, blue, green/yellow, red) to match your laser.

The dyes used in the manufacture of the Cell Sorting Set-up Beads have been carefully selected to provide emission peaks coincident with cells labeled with commonly used fluorescent dyes. The emission profiles for these standards are intentionally narrow in comparison to cells labeled with a corresponding fluorescent dye. Due to their narrow emission profiles, beads of two different fluorescent colors exhibit minimal spectral overlap, and little or no color compensation is needed when setting up for multicolor experiments.
