Applied Biosystems GeneAmp 10X PCR Gold Buffer is formulated to provide flexible, efficient activation of AmpliTaq Gold DNA Polymerase, resulting in highly specific, robust PCR amplification.

The ionic strength and pH of GeneAmp 10X PCR Gold Buffer have been optimized to provide a wider activation temperature and time range when used in conjunction with AmpliTaq Gold DNA Polymerase.

GeneAmp 10X PCR Gold Buffer is both magnesium ion-free and gelatin-free. A separate MgCl2 solution is provided. When used together, GeneAmp 10X PCR Gold Buffer and the MgCl2 solution offer more versatility in optimizing the magnesium ion concentration to achieve optimal PCR amplification with any specific set of primers and DNA template.

Yellow Sample Buffer (40X) (Thermo Scientific™)

Thermo Scientific 40X Yellow Sample Buffer is used with Luminaris Color qPCR Master Mixes to color and track the samples. When using the blue Luminaris Color qPCR Master Mix the qPCR reaction mix is blue before sample addition. After adding the sample the reaction mix turns green, making the overall dispensing of qPCR reaction components quick, easy and error-free. This is particularly important for users of opaque white reaction plates and tubes.

The Yellow Sample Buffer is an aqueous solution of the inert yellow dye (pH 8.0). The buffer is provided as a 40X concentrate and used in 1X concentration in the final reaction (for detailed recommendations refer to the manual of Luminaris Color qPCR Master Mix). The yellow dye does not affect the specificity or sensitivity of qPCR assays. The absorption maximum of the yellow dye is at 413 nm.

Phusion Green HF Buffer Pack (Thermo Scientific™)

Two optimized buffers are available for Thermo Scientific Phusion High-Fidelity DNA Polymerases:

The error rate of Phusion DNA Polymerase in HF Buffer (4.4 × 10-7) is lower than that in GC Buffer (9.5 × 10-7). Therefore, the HF Buffer should be used as the default buffer for high-fidelity amplification. However, GC Buffer can improve the performance of Phusion DNA Polymerase on some difficult or long templates, such as GC-rich templates or those with complex secondary structures. For applications such as microarray or DHPLC, where the DNA templates need to be free of detergents, we recommend using detergent-free reaction buffers.

5X Phusion Green Buffers include a density reagent and two tracking dyes for direct loading of PCR products on a gel. The colored buffers do not interfere with the enzyme performance and are compatible with downstream applications such as DNA sequencing, ligation and restriction digestion.

Taq Buffer (10X), with KCl (Thermo Scientific™)

Thermo Scientific 10X Taq Buffers are ready-to-use buffers for PCR using Taq DNA Polymerases (both recombinant and native). Available in different compositions and also without detergents.

Optimized for Easy and Challenging Targets
Challenging targets include AT-rich, GC-rich, primer-dimer forming amplicons, homopolymer repeats, and amplicons that pose sequencing challenges. Amplicons that previously required specialized enzymes and reaction conditions can now be reproducibly amplified with a single reagent under standardized conditions (Figure 1). Competitive benchmarking across more than 40 amplicons distinguishes AmpliTaq Gold® 360 as the best-performing enzyme, ensuring the highest probability of success for the amplification of both everyday and challenging targets (Table 1).

As shown in Figure 1, GC-rich regions are especially poorly amplified with competitor DNA polymerases and the original AmpliTaq Gold, while AmpliTaq Gold 360 provides successful, robust amplification.

Taq DNA Polymerase PCR Buffer (10X) (Invitrogen™)

Taq DNA Polymerase PCR Buffer is a 10X buffer [200 mM Tris HCl (pH 8.4), 500 mM KCl] supplied with 1 ml of 50 mM MgCl2. It is included with Platinum® Taq, Taq, and the SuperScript® First-Strand Synthesis System for RT-PCR.

SuperFi™ Buffer (Invitrogen™)

Invitrogen™ SuperFi™ Buffer is a 5X buffer solution optimized for use with Invitrogen™ Platinum™ SuperFi DNA Polymerase. The buffer contains 7.5 mM MgCl2 which provides 1.5 mM MgCl2 in the final reaction.

For direct loading of PCR products on gels, use SuperFi™ Green Buffer which includes a density reagent and two electrophoresis tracking dyes. The colored buffer does not interfere with the enzyme performance and is compatible with downstream applications such as DNA sequencing, ligation, and restriction digestion.

Phire Reaction Buffer (Thermo Scientific™)

The reaction buffer for Thermo Scientific Phire Hot Start II DNA Polymerase is available in three formats: non-colored version with and without detergents (F-524L and F-525L, respectively) or Green version (F-527L) containing a density reagent and two tracking dyes for direct loading of PCR products on gel. The colored buffer does not interfere with enzyme performance and is compatible with downstream applications such as DNA sequencing, ligation and restriction digestion.

The concentration of Mg2+ in the reaction buffers is optimized to work well for most amplicons. The buffer contains 1.5 mM MgCl2 at final reaction concentration. Separate tubes of MgCl2 and DMSO are provided for further optimization.

DreamTaq™ Green Buffer (10X) (Thermo Scientific™)

Thermo Scientific™ 10X DreamTaq™ Green Buffer is a proprietary PCR buffer that contains KCl and (NH4)2SO4 at a ratio optimized for robust performance of DreamTaq™ DNA Polymerase and DreamTaq™ Hot Start DNA Polymerase in PCR and includes MgCl2 at a concentration of 20 mM. In addition, the buffer includes a density reagent and two tracking dyes. The density reagent allows direct loading of PCR products on a gel. The blue dye (migrates with 3 to 5 kb DNA fragments in 1% agarose gel) and the yellow dye (migrates faster than 10 bp DNA fragments in 1% agarose gel) are included for monitoring electrophoresis progress.

The dyes have excitation peaks at 424 nm and 615 nm. For applications that require PCR product analysis by absorbance or fluorescence excitation, we recommend using the colorless DreamTaq™ Buffer (10X) or purifying the PCR product using the GeneJET PCR Purification Kit prior to analysis.
