Yellow Sample Buffer (40X) (Thermo Scientific™)

Thermo Scientific 40X Yellow Sample Buffer is used with Luminaris Color qPCR Master Mixes to color and track the samples. When using the blue Luminaris Color qPCR Master Mix the qPCR reaction mix is blue before sample addition. After adding the sample the reaction mix turns green, making the overall dispensing of qPCR reaction components quick, easy and error-free. This is particularly important for users of opaque white reaction plates and tubes.

The Yellow Sample Buffer is an aqueous solution of the inert yellow dye (pH 8.0). The buffer is provided as a 40X concentrate and used in 1X concentration in the final reaction (for detailed recommendations refer to the manual of Luminaris Color qPCR Master Mix). The yellow dye does not affect the specificity or sensitivity of qPCR assays. The absorption maximum of the yellow dye is at 413 nm.

Applied Biosystems GeneAmp 10X PCR Gold Buffer is formulated to provide flexible, efficient activation of AmpliTaq Gold DNA Polymerase, resulting in highly specific, robust PCR amplification.

The ionic strength and pH of GeneAmp 10X PCR Gold Buffer have been optimized to provide a wider activation temperature and time range when used in conjunction with AmpliTaq Gold DNA Polymerase.

GeneAmp 10X PCR Gold Buffer is both magnesium ion-free and gelatin-free. A separate MgCl2 solution is provided. When used together, GeneAmp 10X PCR Gold Buffer and the MgCl2 solution offer more versatility in optimizing the magnesium ion concentration to achieve optimal PCR amplification with any specific set of primers and DNA template.

Thermo Scientific 10X Buffer R ensures the optimum reaction conditions for restriction enzymes and is premixed with BSA for enhanced stability. Our Five Buffer System ensures the optimum reaction conditions for each restriction enzyme. This system consists of 10X B (blue), G (green), O (orange), R (red), and Tango (yellow) buffers. All restriction enzymes are supplied in color-coded tubes to indicate the recommended reaction buffer. The recommended buffer and/or the universal Tango buffer are supplied with each enzyme.

Thermo Scientific restriction enzymes exhibit 100% of their certified activity in the recommended buffer. However, some enzymes require additives to achieve 100% activity. For example, AjuI, AlfI, BdaI, BplI, BseMII, FaqI, Eco57I, Eco57MI, Hin4I, and TsoI require S-adenosylmethionine, which is supplied with the enzyme, while AarI and BveI require an oligonucleotide (also supplied with the enzyme). Esp3I requires DTT.

Optimized for Easy and Challenging Targets
Challenging targets include AT-rich, GC-rich, primer-dimer forming amplicons, homopolymer repeats, and amplicons that pose sequencing challenges. Amplicons that previously required specialized enzymes and reaction conditions can now be reproducibly amplified with a single reagent under standardized conditions (Figure 1). Competitive benchmarking across more than 40 amplicons distinguishes AmpliTaq Gold® 360 as the best-performing enzyme, ensuring the highest probability of success for the amplification of both everyday and challenging targets (Table 1).

As shown in Figure 1, GC-rich regions are especially poorly amplified with competitor DNA polymerases and the original AmpliTaq Gold, while AmpliTaq Gold 360 provides successful, robust amplification.

Terminal Deoxynucleotidyl Transferase (TdT) Buffer (Invitrogen™)

Terminal Deoxynucleotidyl Transferase, Recombinant (rTdT) is a DNA polymerase that catalyzes the addition of deoxynucleotides to the 3fi hydroxyl terminus of DNA. A TdT Technical Bulletin is available.

Applications:
Homopolymer tailing of vector and insert for cloning. Labeling oligonucleotides with biotin (1,2), 32P- or 35S-label (3), or in apoptosis (TUNEL) (4,5).

Purified from a baculovirus clone of calf thymus TdT.

Endonuclease, 3´ and 5´ exodeoxyribonuclease, and levels of incorporation tested.

Unit Definition:

T4 DNA Ligase Buffer (Invitrogen™)

T4 DNA Ligase catalyzes the formation of phosphodiester bonds in the presence of ATP between double-stranded DNAs with 3´ hydroxyl and 5´ phosphate termini. The unique T4 DNA Ligase buffer optimizes ligation, which can be performed in 5 minutes (1). Single-stranded nucleic acids are not substrates for this enzyme. A T4 DNA Ligase Technical Bulletin is available.

Applications: Cloning (blunt-end or cohesive-end ligation) (2). Adding linkers or adapters to blunt-ended DNA (2).

Unit Definition: One unit catalyzes the exchange of 1 nmol 32P-labeled pyrophosphate into ATP in 20 min. at 37°C. (One unit is equal to approximately 300 cohesive-end ligation units.)

Unit Reaction Conditions: 66 mM Tris-HCl (pH 7.6), 6.6 mM MgCl2 , 10 mM DTT, 66 µM ATP, 3.3 µM 32 P-labeled pyrophosphate, and enzyme in 0.1 ml for 20 min. at 37°C.
