eBioscience™ Flow Cytometry Staining Buffer (Invitrogen™)

Our Flow Cytometry Staining Buffer is designed for use in immunofluorescent staining protocols of cells in suspension. This Flow Cytometry Staining Buffer is a buffered saline solution containing fetal bovine serum and sodium azide (0.09%) as a preservative. This buffer can be used for antibody and cell dilution steps, as well as all the wash steps required for the surface staining and flow cytometric analysis. This buffer contains animal serum proteins to help minimizing non-specific binding of antibodies.

eBioscience™ Mouse Cytokine Positive Control Cells (Invitrogen™)

The Mouse Cytokine Positive Control Cells are recommended for use as positive control cells when establishing an intracellular cytokine staining protocol. Mouse splenocytes were stimulated with ConA (3 µg/mL) for two days, followed by recombinant IL-2 (20 ng/mL) and IL-4 (20 ng/mL) for three days. The cells were then restimulated with immobilized anti-mouse CD3 (10 µg/mL) and soluble anti-mouse CD28 (2 µg/mL) in the presence of Brefeldin A for 5 hours. These cells contain subpopulations that express IL-2, IL-3, IL-4, IL-10, GM-CSF, and IFNγ. The cells have been fixed with formaldehyde and are provided in a stabilizing buffer so that they may be stored at -80°C for up to six months or at 2-8°C for up to one month.

Attune™ Focusing Fluid (1X)

Attune® Focusing Fluid is an optimized buffer solution that is used in the Attune® Acoustic Focusing Cytometer for focusing and transportation of particles throughout the fluidics system.

Maintains System Cleanliness
Attune® Focusing Fluid is a sterile, buffered, azide-free support reagent that contains a surfactant that helps clean the fluidic lines of the Attune® Acoustic Focusing Cytometer. The Attune® Focusing Fluid was designed to minimize bubble formation and to prevent build-up of cellular material in the flow cell, resulting in less instrument clogging.

Convenient
The ready-to-use 1X focusing fluid was designed to eliminate the need for dilution prior to use, as well as access to a purified water system. The convenient 10 L packaging provides enough focusing fluid to operate for at least 10 days (the average focusing fluid consumption per day is less than 1 L) and has a spigot to to make filling of the onboard fluidics tanks easier.

Each batch of Attune® Focusing Fluid is traceable by its lot number designation and has been validated for use on the Attune® Acoustic Focusing Cytometer.

Attune™ Debubble Solution (1X)

Attune® Debubble Solution is used to remove excess air bubbles that may develop in the fluidic lines of the Attune® NxT Acoustic Focusing Cytometer.

Minimizes bubble formation
Attune® Debubble Solution has been formulated to minimize bubble formation that may occur in the system lines overnight or after system maintenance. This solution is used during the automated Debubble function of the Attune® acoustic focusing cytometer. It is also used after the regular quarterly maintenance/system decontamination function to help ensure that all air introduced during this cleaning process is removed prior to analyzing samples on the system.

Convenient
This ready-to-use 1X debubble solution was designed to eliminate the need for dilution prior to use and the need for access to a purified water system.

UltraComp eBeads™ Compensation Beads (Invitrogen™)

UltraComp eBeads™ react with antibodies of mouse, rat, and hamster origin, and are immunoglobulin light chain-independent. They are designed for use in compensation with all fluorochromes excited by ultraviolet (355 nm) violet (405 nm), blue (488 nm), green (532 nm), yellow-green (561 nm), and red (633-640 nm) lasers. The beads are spherical particles that can be stained with individual fluorochrome-conjugated antibodies for use as single-color compensation controls.

Each drop of beads contains two populations: a positive population that will capture any mouse, rat, or hamster antibody and a negative population that will not react with antibody. When a fluorochrome-conjugated antibody is added to the beads, both positive and negative populations result. This bimodal distribution can be used for single-color compensation controls in multicolor flow cytometry experiments.

UltraComp eBeads are compatible with all fluorochromes excited by an ultraviolet (355 nm) or violet (405 nm) laser. They should be used with standard staining buffers which contain PBS or HBSS, protein such as BSA or FBS, and sodium azide. No other additives should be used. OneComp eBeads™ Compensation Beads (Invitrogen™)

OneComp eBeads react with antibodies of mouse, rat and hamster origin, and are immunoglobulin light chain-independent. The beads are spherical particles that can be stained with individual fluorochrome-conjugated antibodies for use as single-color compensation controls.

Each drop of beads contains two populations: a positive population that will capture any mouse, rat or hamster antibody and a negative population that will not react with antibody. When a fluorochrome-conjugated antibody is added to the beads, both positive and negative populations result. This bimodal distribution can be used for single-color compensation controls in multicolor flow cytometry experiments.

OneComp eBeads cross-react to some antibodies of rabbit origin, but have not been extensively tested for this reactivity. OneComp eBeads are designed for use in compensation with all fluorochromes excited by blue (488 nm), green (532 nm), yellow-green (561 nm), and red (633-635 nm) lasers. This product is compatible with eFluor™ 450 but is not optimized for compensation of other fluorochromes excited by a violet (405 nm) laser.

Super Bright Staining Buffer, eBioscience™ (Invitrogen™)

The Super Bright Staining Buffer, eBioscience™, is designed for use as a supplement to Flow Cytometry Staining Buffer in immunofluorescent staining protocols of cells in suspension. Super Bright Staining Buffer is only necessary when using more than one Super Bright-conjugated antibody in the same experiment to prevent non-specific polymer interactions, which can result in data appearing under-compensated. Super Bright Staining Buffer is provided in a convenient 5 µL/test format and is compatible with traditional fluorochromes, Brilliant Violet dyes and standard flow cytometry protocols.

Super Bright Staining Buffer is not compatible with UltraComp eBeads microspheres. If using UltraComp eBeads microspheres as a compensation tool, Flow Cytometry Staining Buffer should be exclusively used for all dilutions and washes.

Super Bright Complete Staining Buffer, eBioscience (Invitrogen™)

Super Bright Complete Staining Buffer, eBioscience, is designed for use as a supplement to Flow Cytometry Staining Buffer in immunofluorescent staining protocols of cells in suspension. Super Bright Complete Staining Buffer is only necessary when using more than one polymer dye-conjugated antibody in the same sample to prevent non-specific polymer interactions, which can result in data appearing under-compensated. Super Bright Complete Staining Buffer is provided in a convenient 5 µL/test format and is compatible with traditional fluorochromes, Brilliant Violet dyes, and standard flow cytometry protocols.

Super Bright Complete Staining Buffer is not compatible with UltraComp eBeads microspheres. If using UltraComp eBeads microspheres as a compensation tool, Flow Cytometry Staining Buffer should be used exclusively for all dilutions and washes during staining of the microspheres.

Super Bright Complete Staining Buffer is a fully compatible replacement for Super Bright Staining Buffer (Cat. Nos. SB-4400-42, SB-4400-75), and it shows decreased background in the 450/40 channel on the violet laser.

123count eBeads™ Counting Beads (Invitrogen™)

123count eBeads are intended for use in absolute counting of cells or other particles by flow cytometry. These are 7 μm microparticles containing encapsulated dyes compatible with blue (488 nm) and violet (405 nm) excitation sources and emitting fluorescence between approximately 500 nm and 750 nm. They are supplied at a known concentration in 0.05% Tween-20 with 2 mM sodium azide. Collecting carefully suspended and measured quantities of cell:bead mixtures on a flow cytometer makes the ratiometric enumeration of cells possible.

eBioscience™ BrdU Staining Buffer Set for Flow Cytometry (Invitrogen™)

The BrdU Staining Buffer Set contains the optimized buffers needed for optimal BrdU staining with the BU20A monoclonal antibody for flow cytometric analysis. This set in combination with BrdU and DNase I steps allows for detection of BrdU pulse-labeled cells thereby enabling the examination of proliferating cells. Cycling cells are incubated with 5-bromo-2’-deoxyuridine (BrdU), a synthetic analog of thymidine which incorporates into newly synthesized genomic DNA during the S-phase of mitosis. Following DNA denaturation, the cells are stained for BrdU incorporation along with any other cell surface and/or intracellular targets of interest.

Attune™ Shutdown Solution (1X)

Attune® Shutdown Solution is a cleaning buffer that is used in the automated shutdown procedure for the Attune® Acoustic Focusing Cytometer.

Minimizes Bubble Formation
This azide-free shutdown solution has been formulated to minimize bubble formulation that can occur in the system lines overnight, to help keep the fluid lines clean, and to minimize microbial growth in the fluid lines.

Use for Instrument Maintenance
The Shutdown cycle is used at the end of each day to ensure that all sample fluids and dyes have been removed from the system, and that the fluidic lines and pumps are filled with Attune® Shutdown Solution to prevent salt deposits from forming in the lines. Shutdown takes approximately 20 minutes, but most of the steps are automatic and under the control of the Attune® Cytometric Software. Shutdown procedures should be run at least once a day, even if the instrument is intended for continuous use. Proper cleaning of the instrument ensures its consistent and accurate operation.

Attune® Wash Solution is an optimized cleaning buffer that is used in the Attune® Acoustic Focusing Cytometer to maintain cleanliness of the fluidics lines, minimizing contamination or carryover.

Maintains System Cleanliness
Attune® Wash Solution has been formulated to remove cellular debris, prevent protein building up, and minimize dyes sticking to the fluidics lines of the system. It is recommended for use during the routine maintenance and automated shutdown procedures of the Attune® Acoustic Focusing Cytometer.

Convenient
The ready-to-use 1X wash solution was designed to eliminate the need for dilution prior to use, as well as access to a purified water system. The wash solution is stable for 30 days after the bottle is opened.
